Abstract

A method for treating a disorder of the central nervous system includes administering to the respiratory tract of a patient a drug which is delivered to the pulmonary system, for instance to the alveoli or the deep lung.
Particles that include the drug can be employed. Preferred particles have a tap density of less than about 0.4 g/cm3. In addition to the medicament, the particles can include other materials such as, for example, phospholipids, amino acids, combinations thereof and others. The drug is administered at a dose which is at least about two-fold less than the dose required by oral administration.

Claims (58)

1. A method for providing rescue therapy for a disorder of the central nervous system comprising administering to the respiratory tract of a patient in need of rescue therapy particles comprising an effective amount of a medicament, wherein the particles have a tap density less than about 0.4 g/cm3 and are delivered to the alveoli region of the pulmonary system.
2. A method for providing rescue therapy for Parkinson's disease comprising administering to the respiratory tract of a patient in need of rescue therapy particles comprising an effective amount of all anti-Parkinson disease medicament, wherein the particles are delivered to the alveoli region of the pulmonary system.
3. The method of Claim 2, wherein the particles have a tap density less than about 0.4 g/cm3.
4. The method of Claim 3, wherein the particles have a tap density less than about 0.1 g/cm3.
5. The method of Claim 3, wherein the particles have an mass median aerodynamic diameter of less than about 5 microns.
6. The method of Claim 5 wherein the particles have an aerodynamic diameter of less than about 3 microns.
7. The method of Claim 3, wherein the particles have a volume median geometric diameter in the range of between about 5 microns and about 30 microns.
8. The method of Claim 2, wherein the medicament is levodopa.
9. The method of Claim 8 wherein levodopa is present in the particles in an amount ranging from about 1 to about 90 weight percent.
10. The method of Claim 9 wherein levodopa is present in the particles in an amount of at least about 20 weight percent of.
11. The method of Claim 2, wherein the medicament is carbidopa.
12. The method of Claim 2 wherein the medicament is apornorphine.
13. The method of Claim 2 wherein the particles additionally comprise a phospholipid.
14. The method of Claim 13 wherein the phospholipid is present in the particles in an amount of at least 10 weight percent.
15. The method of Claim 13 wherein the particles have a matrix transition temperature that is no higher than the patient's physiological temperature.
16. The method of Claim 2 wherein the particles additionally comprise an amino acid.
17. The method of Claim 16 wherein the amino acid is present in the particles in an amount of at least about 10 weight percent.
18. The method of Claim 2 wherein the particles additionally comprise citrate.
19. The method of Claim 18 wherein the citrate is present in the particles in an amount of at least about 10 percent.
20. The method of Claim 18 wherein the particles further comprise a divalent metal salt.
21. The method of Claim 20 wherein the divalent metal salt is calcium chloride.
22. The method of Claim 2 wherein the medicament is administered in a dose that is at least about two times less than that required by oral delivery.
23. The method of claim 2 wherein the medicament reaches its site of action, thereby providing rescue therapy in less than about thirty minutes.
24. The method of claim 23 wherein the medicament reaches its site of action thereby providing rescue therapy in less than about fifteen minutes.
25. The method of Claim 2 wherein the particles are administered as a dry powder.
26. The method of Claim 25 wherein the medicament is present in the dry powder in an amount of at least 20 weight percent.
27. The method of Claim 26 wherein the medicament is present in the dry powder in an amount of at least 40 weight percent.
28. The method of Claim 27 wherein the drug is present in the dry powder particles in an amount of at least 50 weight percent.
29. The method of Claim 2 wherein administering particles is in combination with ongoing therapy.
30. The method of Claim 2 wherein the particles further comprise at least one bioactive agent.
31. A method for treating a disorder of the central nervous system comprising administering to the respiratory tract of a patient in need of treatment or rescue therapy ketoprofen, wherein ketoprofen is administered in a dose that is at least about two times less than that required by oral administration and wherein delivery is to the alveoli region of the pulmonary system.
32. A method for treating a disorder of the central nervous system comprising administering to the respiratory tract of a patient in need of ongoing treatment or rescue therapy a benzodiazepine drug, wherein the benzodiazepine drug is administered in a dose that is at least about two times less than that required by oral administration and wherein delivery is to the alveoli region of the pulmonary system.
33. A method for treating a disorder of the central nervous system comprising administering to the respiratory tract of a patient in need of rescue therapy particles comprising an effective amount of a benzodiazepine, wherein the particles have a tap density less than about 0.4 g/cm3 and are delivered to the alveoli region of the pulmonary system.
34. The method of Claim 33, wherein the particles have a tap density less than about 0.1 g/cm3.
35. The method of Claim 33, wherein the particles have an mass median aerodynamic diameter of less than about 5 microns.
36. The method of Claim 33, wherein the particles have an mass median aerodynamic diameter of less than about 3 microns.
37. The method of Claim 33, wherein the particles have a volume median geometric diameter in the range of between about microns and about 30 microns.
38. The method of Claim 33, wherein the medicament is alprazolam.
39. The method of Claim 38 wherein benzodiazepine is present in the particles in an amount ranging from about 1 to about 90 weight percent.
40. The method of Claim 39 wherein the benzodiazepine is present in the particles in an amount of at least about 20 weight percent.
41. The method of Claim 2 wherein the particles additionally comprise a phospholipid.
42. The method of Claim 41 wherein the phospholipid is present in the particles in an amount of at least 10 percent by weight.
43. The method of Claim 41 wherein the particles have a matrix transition temperature which is no higher than the patient's physiological temperature.
44. The method of Claim 33 wherein the particles additionally comprise an amino acid.
45. The method of Claim 44 wherein the amino acid is present in the particles in an amount of at least about 10 percent.
46. The method of Claim 33 wherein the particles additionally comprise citrate.
47. The method of Claim 46 wherein the citrate is present in the particles in an amount of at least about 10 percent.
48. The method of Claim 46 wherein the particles further compose a divalent metal salt.
49. The method of Claim 48 wherein the divalent metal salt is calcium.

50. The method of Claim 33 wherein the benzodiazepine is administered in a dose, as measured by plasma levels, that is at least about two times less than that required by oral delivery.
51. The method of claim 33 wherein the medicament reaches its site of action, thereby providing rescue therapy in less than about thirty minutes.
52. The method of claim 51 wherein the drug reaches its site of action thereby providing rescue therapy in less than about fifteen minutes.
53. The method of Claim 33 wherein the particles are administered as a dry powder.
54. The method of Claim 53 wherein the medicament is present in the dry powder in an amount of at least 20 weight percent.
55. The method of Claim 54 wherein the medicament is present in the dry powder in an amount of at least 40 weight percent.
56. The method of claim 55 wherein the drug is present in the dry powder particles in an amount of at least 50 weight percent.
57. The method of Claim 33 wherein administering particles is in combination with ongoing therapy.
58. The method of Claim 33 wherein the particles further comprise at least one bioactive agent.
