本発明は、 高分子材料製人工関節部材に関し、 特に、 摺動部 位の潤滑状態を長期間に渡って良好に維持する こ とのできる高 分子材料製人工関節部材に関するものである。 背景技術
人体における実際の生体内関節部の構造を考える と、 人工関 節摺動部に良好な潤滑状態を与えるには、 その構造が、 関節部 材摺動面に繊毛の如 く 微細なひげ状の物質が配列されて、 該繊 毛状物質に潤滑液である体液が保持され得る構造のものが好ま しいと考えられる。 しかし、 こ の様な状態を人工的に再現する こ とは難し く 、 更に前記状態を長期間に渡って維持する こ とは 至難のこ とであって未だ実現されていない。
現状では、 骨頭や臼蓋等の人工関節部材と して樹脂等の軟質 材料と金属等の硬質材料が組み合わせて用い られ、 例えばステ ンレス、 コバル ト ク ロム合金、 チタ ン合金等の金属材料を用い た骨頭部材と、 超高分子量ポ リ エチレン等の樹脂を用いた臼蓋 を組み合わせた人工関節が、 わが国だけでも年間数万件の手術 に用い られている。 こ の様に人工関節に置換する こ とで、 患者 は苦痛や寝たき り の状態から開放され、 健康な人と同様の生活 を営むまでに生活の質を向上させる こ とができる。
しかしながら前記人工関節は、 日常生活の動作によ り 、 その 摺動部位で頻繁に前記金属部材と樹脂部材の摩擦が生じるため 、 特に人工関節の樹脂側で摩耗が進行する。 こ の様な摩耗によ り 生じた超高分子量ポ リ エチレン等の摩耗粉は、 骨の溶解を誘 発し易 く 、 骨の一部が溶解する と人工関節部材と骨の固着力が 弱まる、 いわゆるルーズニングが生じ、 人工関節と しての機能 が十分に発揮され難く なる。 前記超高分子量ポ リ エチレンの通 常の摩耗量は年間 0 . 1 〜 0 . 2 mm程度で、 置換手術後しば ら く は問題も生じないが、 5 年程度経過する と前記ルーズニン グが著し く なるので新しい人工関節部材に取 り 替える必要が生 じて く る。
最近では、 この様な樹脂の摩耗を抑える手段と して、 超高分 子量ポ リ エチレンにガンマ線や電子線 (エレク ト ロ ンビーム) を照射する こ とによ り分子を架橋状態にした、 いわゆる (ク ロ ス リ ンク) ポ リ エチレンが提案され、 摩耗量が照射前の超高分 子量ポリ エチレンと比較して 1 ノ 5 〜 1 1 0 にまで低減でき た旨報告されている ( Biomaterials, Vol.2 0 ( 1 9 9 9 ) , p 1 6 5 9 〜 1 6 8 8 )。 しかしながら臨床的応用の経過年数 が少なく 、 その有用性は未だ立証されていない。
また、 摩耗し易い前記超高分子量ポ リ エチレン等の樹脂を用 いず、 骨頭および臼蓋に硬質部材同士を組み合わせて使用する こ と も提案されてお り 、 例えば、 コバル ト ク ロム骨頭とコバル ト ク ロ ム 合 金 カ ッ プ ( 臼 蓋 ) の 組 み 合 わ せ ( Clinicalorthopaedics and Related Research, No333 1 9 9 6 ) p.96- 107) や、 アルミ ナ骨頭とアルミ ナカ ッ プ (臼蓋)の 組み合わせ ( The Journal of Arthroplasty, Vol.14 No.7 ( 1 9 9 9 ) p.781- 787) 等が臨床的に人工股関節に使用 されてい る。 しかしながら、 前記コノ ル ト ク ロム骨頭と コバル ト ク ロム 合金カ ッ プ (臼蓋) の組み合わせは、 コバル ト ク ロム合金同士 の摩擦で生じる摩耗粉が、 元来、 細胞毒性を有するので、 長期 の使用については安全性が危惧されている。
また、 前記アルミ ナ骨頭とアルミ ナカ ッ プ (臼蓋) の組み合 わせは、 アルミ ナ材料が脆性材料であるため、 手術中や術後の 使用中に破損が生じる こ とがあ り 、 実用に際しては更なる改善 が必要である。
更にこれらの硬質部材は、 弾性の乏しい剛体であって前記超 高分子量ポリ エチレンの様なク ッ ショ ン機能を有さないので、 外力に対する緩衝作用がなく 骨に直接負荷がかかるので好ま し く ない。
本発明は、 この様な事情に鑑みてなされたものであ り 、 その 目的は、 繰り返し行われる 日常の動作に対しても、 摺動部位の 摩擦を抑えて摩耗粉の発生を抑制する こ とができ、 かつ衝撃吸 収機能を備えて長期間に渡り使用できる高分子材料製人工関節 部材を提供する こ とにある。 発明の開示
本発明に係る高分子材料製人工関節部材とは、 高分子物質で 形成される人工関節部材の摺動面がホスホリ ルコ リ ン基を有す る高分子からなる と ころに特徴を有してお り 、 その好ま しい実 施形態と して以下のものが例示される。
①前記ホスホ リ ルコ リ ン基が (メ タ) ァク リ ロイルォキシァ ルキル基を介して高分子物質に結合されたもので、 その中でも 、 前記高分子物質にホスホ リ ルコ リ ン基を有する重合性単量体 がグラフ ト結合されている ものは、 化学的に安定しているので 望ま しい。
②前記高分子物質にホスホ リ ルコ リ ン含有化合物の単独重合 体または共重合体がコーティ ングされている もので、 該ホスホ リ ルコ リ ン含有化合物と しては、 (メ タ) ァ ク リ ロイルォキシ アルキルホスホ リルコ リ ンを用いたものが好ま しい形態と して 推奨される。
前記高分子物質と しては、 分子量 1 0 0 万以上の超高分子量 ポ リ エチレンが好ま しく 使用される。 また、 前記高分子物質と して架橋処理されたものを用いれば、 優れた耐摩耗性を確保で きるので望ま しい。
更に本発明は、 この様な高分子材料製人工関節部材を用いて 得られる人工関節も含むものである。
尚、 前記 「高分子物質」 とは人工関節部材のベース部分を示 し、 前記 「高分子材料」 と は、 「高分子物質」 および 「ホスホ リ ルコ リ ン基を有する高分子からなる人工関節部材の摺動面」 を併せた人工関節部材全体を指すものとする。 図面の簡単な説明

本発明者らは、 前述した様な状況の下で、 金属等の硬質材料 よ り も優れた衝撃吸収機能を有する高分子材料を人工関節部材 と して用いた場合に、 摺動部位の摩擦が極力抑制され、 かつ摩 擦によ り 生じる摩耗粉が免疫機能を発揮する細胞に対して不活 性となるよ う、 人工関節部材に用いる高分子物質の表面修飾に ついて様々 な角度から検討を行った。 その結果、 生体組織を構 成する細胞と同様の化学構造を有する化合物を高分子物質の表 面に形成すればよいこ とを見出し、 本発明に想到した。 本発明 にかかる高分子材料製人工関節部材について以下に詳述する。
図 1 に示す様に、 生体関節部等の骨材表面を構成する生体膜 は、 リ ン脂質分子の集合体で、 その表面は微視的にホスホリ ル コ リ ン基で覆われている (石原 : 外科 6 1 巻 1 3 2 頁 ( 1 9 9 9 ) )。 そこで人工関節部材についても、 関節の骨頭および Zま たは臼蓋を構成する高分子材料の摺動面が、 ホスホリルコ リ ン 基を有する高分子で構成されるよ う にすれば、 前記潤滑液との 親和性を確保する こ とができるので、 潤滑液を保持して良好な 潤滑状態を維持でき、 結果と して摩擦を著し く 低減できる こ と がわかった。 更に、 前記ホスホリルコ リ ン基を有する化合物は 、 生体組織を構成する細胞と同様の化学構造を有しているので 、 たとえ摩擦によ り摩耗粉が発生したと しても、 人体に悪影響 を与える こ とがなく 良好な生体適合性を示す。 本発明は、 こ の様に高分子物質で形成される人工関節部材の 摺動面をホスホ リ ルコ リ ン基を有する高分子で構成する こ とに よって、 上記効果を得たもので、 ホスホ リ ルコ リ ン基を有する 高分子の具体的化学構造まで規定する ものではないが、 ホスホ リルコ リ ン基が (メ タ) ァク リ ロイルォキシアルキル基を介し て高分子物質に化学結合したものは、 化学的に安定であ り 、 か つ後述する重合法によって形成し易いので望ま しい。
この様に、 ホスホ リ ルコ リ ン基が (メ タ) ァク リ ロイルォキ シアルキル基を介して高分子物質に化学的に結合された構造を 得る方法と して、 モノ マーを高分子物質に直接重合してグラフ ト化する方法、 ホスホリ ルコ リ ン基を有する反応性ポリ マーを 予め合成し、 これを高分子物質と反応させる方法、 架橋能を持 つホスホリ ルコ リ ン基を有するポリ マーを予め合成し、 高分子 物質に被覆した後、 架橋反応によ り 高分子物質表面に固定化す る方法等が挙げられるが、 その中でも特に、 グラ フ ト重合で高 分子物質にホスホリ ルコ リ ン基を有する重合性単量体を結合さ せる方法が、 高分子物質の有する強度等の性能を劣化させる こ となく 高分子物質表面のみを修飾する こ とができ、 かつ結合部 分が化学的に安定し、 更に、 多量のホスホ リ ルコ リ ン基を人工 関節部材の摺動面に形成して潤滑液を十分に保持する こ とがで きるので好ま しい。
前記ホスホ リ ルコ リ ン基を有する重合性単量体と しては、 例 えば、 2 — メ 夕ク リ ロイルォキシェチルホスホ リ ルコ リ ン、 2 —ァク リ ロイルォキシェチルホスホ リ ルコ リ ン、 4 一メ タク リ ロイルォキシブチルホスホリ ルコ リ ン、 6 _ メ夕ク リ ロイルォ キシへキシルホスホ リ ルコ リ ン、 ω —メタク リ ロイルォキシェ チレンホスホ リ ルコ リ ン、 4 —スチリ ルォキシブチルホスホリ ルコ リ ン等を用いる こ とができる。 その中でも特に、 下記化学 式 1 に示す 2 — メタ ク リ ロイルォキシェチルホスホ リルコ リ ン
(以下、 単に 「 M P C」 という こ とがある) は、 ホスホ リ ルコ リ ン基と重合性のメ 夕ク リル酸ュニッ 卜からな り 、 ラジカル重 合で容易に高分子量の重合体を得る こ とができるので好ま しい
( Ishihara ら : Polymer Journal 誌 2 2 巻 3 5 5 頁 ( 1 9 9 0 ))。
一例と して前記 M P C を用いてグラフ ト重合を行う と、 2 — メタク リ ロイルォキシェチルホスホリルコ リ ンを含む重合体 ( 以下、 単に 「 M P Cポリ マー」 という こ とがある) を高分子物 質に結合させて、 多量のホスホ リ ルコ リ ン基を人工関節部材の 摺動面に形成させる こ とができる。 そして M P Cポリ マーの形 成された表面は、 前記図 1 に示した様な生体膜と類似の形態に なる こ とから、 潤滑液を十分に保持して摩擦を抑制する こ とが でき、 かつ生体成分や細胞との反応性も小さ いので望ま しい。
尚、 本発明では、 前記グラ フ 卜重合の際に他のビニル化合物 等を共存させ、 ホスホリ ルコ リ ン含有化合物との共重合体とな つていても差し支えない。 化 1
本発明では、 高分子物質で形成さ れる人工関節部材の摺動面 がホスホ リ ルコ リ ン基を有する高分子か らなる場合のその他の 態様と して、 前記高分子物質にホス ホ リ ルコ リ ン含有化合物の 単独重合体または共重合体がコーティ ングされたものが挙げ ら れる。
この場合も、 ホスホ リ ルコ リ ン含有化合物には、 高分子を形 成させるのに容易な重合性化合物である (メ タ) ァク リ ロイ ル ォキシアルキルホスホ リ ルコ リ ンを用い るのがよ く 、 例えば 2 — メ タ ク リ 口イ リレオキシェチルホスホ リ Jレコ リ ン、 2 — ァク リ ロイルォキシェチルホスホ リ ルコ リ ン、 4 _ メ タ ク リ ロイ リレオ キシブチルホスホ リ ルコ リ ン、 6 — メ タ ク リ ロイルォキシへキ シルホスホ リ ルコ リ ン、 ω — メ 夕 ク リ ロイ ルォキシエチ レンホ スホ リ ルコ リ ン、 4 —スチ リ ルォキシブチルホスホ リ ルコ リ ン 等が挙げられるが、 その中でも重合性の良好な 2 — メ タ ク リ 口 ィ ルォキシェチルホスホ リ ルコ リ ンを用 いる こ とが最も好ま し い
この様なホスホ リ ルコ リ ン含有化合物の単独重合体または、 ビニル化合物、 メ タ ク リ ル酸エステル、 アク リ ル酸エステルま たはスチレン誘導体等との共重合体、 具体的にはメ タク リ ル酸 プチル、 メ 夕 ク リ ル酸 2 —ェチルへキシル、 アク リ ル酸ブチル 、 アク リ ル酸 2 —ェチルへキシル、 スチ レン等との共重合体を 、 例えばエタ ノ ール、 プロパノ ール、 テ ト ラ ヒ ド ロ フ ラ ン等の 溶媒に溶解させた ものを高分子物質表面に塗布、 浸漬等の方法 でコ一ティ ングして得る こ とができる。
高分子物質で形成される人工関節部材の摺動面がホスホ リ ル コ リ ン基を有する高分子か らなる も のの更に他の態様と して、 超高分子量ポ リ エチレン等の高分子物質に前記ホスホ リ ルコ リ ン含有化合物を予め練り込んでから成形するプレン ド法等を採 用 してもよい。
本発明に係る人工関節部材のベース となる高分子物質と して は、 超高分子量ポ リ エチレンを用いるのが望ま しい。 前記超高 分子量ポリ エチレンは、 その分子量が高いほど摺動摩耗が生じ 難く なるので分子量 1 0 0 万以上のものを用いるのが好ま し く 、 よ り好ま し く は分子量 3 0 0 万以上である。
上記高分子物質と しては、 架橋処理を施したものを用いれば 、 優れた耐摩耗性を確保できるので望ま しい。 該架橋処理の具 体的な方法と しては、 公知の方法を採用すればよ く 、 例えば X 線照射やガンマ線照射、 電子線照射等の他、 マイ ク ロ波による 架橋や超音波による架橋等が挙げられる。
また本発明は、 前記グラフ ト重合させるにあたって詳細な条 件まで規定するものではなく 、 例えば、 後述する実施例に示す 如く 波長 3 0 0 〜 4 0 0 n mの紫外光を照射して行う他、 過酸 化べンゾィル、 過酸化水素等の水素を引き抜く 作用を有する化 合物を用い、 炭素原子にラジカルを生じさせ、 これを利用 して モノ マーを重合する方法や、 芳香族ニ ト ロ化合物やメルカプ ト 化合物を高分子物質に吸着させた後、 該化合物が溶出 しない溶 媒中にてラジカル発生剤を用いてモノ マーを重合し、 生成する ポ リ マーラジカルを吸着させた芳香族ニ ト ロ化合物やメルカ プ ト化合物によ り捕捉する こ とで、 高分子物質表面にポ リ マー鎖 をグラフ ト化する方法等によ り行う こ とができる。
本発明にかかる高分子材料製人工関節部材を人工関節に適用 するにあたっては、 骨頭および臼蓋の双方を本発明の人工関節 部材とする こ との他、 骨頭および臼蓋の一方を本発明の人工関 節部材と し、 他方を、 例えばステン レス、 コバル ト ク ロム合金 等の金属、 アルミナゃジルコニァ等のセラ ミ ッ クスなどからな る部材と してもよい。 また、 骨頭および/または臼蓋の人工関 節部材の摺動部位のみを本発明の人工関節部材と し、 その他の 部分が他の高分子材料や上記金属、 セラ ミ ッ クス等で構成され ている複合形態のものであってもよい。 実施例
以下、 実施例を挙げて本発明をよ り具体的に説明するが、 本 発明はもとよ り 下記実施例によって制限を受ける ものではな く 、 前 · 後記の趣旨に適合し得る範囲で適当に変更を加えて実施 する こ とも可能であ り 、 それらはいずれも本発明の技術的範囲 に含まれる。
実施例 I : 濡れ性について
人工関節部材の潤滑性の尺度と して、 まず、 液体に対する濡 れ性を評価した。
分子量 3 0 0 〜 5 0 0 万の超高分子量ポリ エチレン角材 (断 面 : 2 0 mm X 2 0 mm, 長さ : 5 0 mm) の表面 (一面のみ ) に、 次の方法でホスホリ ルコ リ ン基を有する高分子層を形成 した。
まず、 前記超高分子量ポリ エチレン角材を、 ベンゾフ エノ ン 含有 ( 1 . O g Z d L ) のアセ ト ン溶液に 3 0 秒間浸漬した後 、 直ちに引き上げて室温で溶媒を除去した。
M P C ( 2 — メタ ク リ ロイルォキシェチルホスホ リ ルコ リ ン ) を 0 . 5 m o 1 Z L含む水溶液を十分に脱気した後、 ベンゾ フエ ノ ンを十分に吸着させた角材を該水溶液に浸漬し、 次いで 液温 6 0 でで波長が 3 0 0 〜 4 0 0 n mの紫外光を角材に 3 0 分間照射し、 その後、 試料を純水で十分に洗浄して M P Cポ リ マ一を表面に形成させた試験片を得た。
尚、 得られた試験片の M P Cポ リ マー形成表面を X線光電子 スぺク トル装置で調べ、 3 9 9 e Vの窒素原子のシグナルと 1 3 3 e Vの リ ン原子のシグナルが検出されたこ とによ り 、 M P C 中のホスホリ ルコ リ ン基が高分子材料の表面に存在する こ と を確認した。
こ の様に して M P Cポ リ マーが形成された超高分子量ポ リ エ チレン角材および M P Cポ リ マーを形成しなかった未処理の超 高分子量ポ リ エチレン角材を試験片と して用い、 濡れ性を評価 した。 濡れ性は、 前記角材表面に表 1 に示す液体を 1 滴 (約 3 0 mm 3 ) 滴下して表面上の液滴の接触角度を測定した。 その 結果を表 1 に併記する。 表 1
表 1 に示す結果よ り 、 M P Cポ リ マーを形成した超高分子量 ポリ エチレン角材では、 いずれの液体を滴下した場合にも、 M P Cポ リ マーを形成していない未処理のものと比較して接触角 度が著し く 小さ く 、 濡れ性が良好である こ とが分かる。 実施例 π: 摩擦係数について
次に、 人工関節部材の潤滑性を摩擦係数で評価した。
実施例 I と 同様に して得た試験片に、 潤滑液と して純水を介 して表 2 に示す金属製の直径 5 mmの丸棒の端面を押え付け、 面圧 1 c m 2 になる大きさ の荷重を角材と金属丸棒の接触 面に垂直に加え、 次に速度 1 0 O mmZm i nで丸棒のみ角材 表面上を滑らせた場合の摩擦係数を求めた。 その結果を表 2 に 併記する。 表 2
表 2 よ り 、 M P Cポ リ マーを形成した場合には、 摩擦係数が 小さ く 潤滑性が著し く 改善されている こ とがわかる。 これは、 数分子層程度の M P Cポ リ マーが表面に形成される こ とで親水 性が増加 し、 最高 9 0 %程度の 自 由水含有率の高い層が表面に 形成されているため と考え られる。
実施例 III : 回転揺動摩耗試験 ( 1 )
人工関節のモデルと して、 臼蓋に超高分子量ポ リ エチ レ ン力 ッ プを用い、 骨頭にステン レス製球体を用 いて回転揺動摩耗試 験を実施 した。
図 2 は回転揺動摩擦試験に用 いた装置を示すものであ り 、 主 軸 X— Xに沿っ て 自 由 に回転する主軸 B に、 直径 1 2 . 1 0 m mの半球状の凹部 2 を有する超高分子量ポ リエチレン製カ ッ プ (分子量 3 0 0 〜 5 0 0 万) 1 を取り付け、 一方、 ステンレス ( S U S 3 1 6 ) 製の直径 1 2 . 0 0 m mの鋼球体 3 を治具に 取り付け、 空気圧によ り 力 Fで鋼球体 3 をカ ッ プ 1 の内壁凹部 面 2 に押し付けた状態で、 鋼球体 3 の支持軸が主軸 X — Xに対 して任意の角度 6» に傾いた状態で回転させるよ う に した。 尚、 鋼球体 3 は、 図示していないが回転数を調整する こ とのできる モーターで駆動させる。 主軸 B には、 摩擦 トルク を測定するた めのアームが取り付けられ、 鋼球体 3 の回転に伴う摩擦力で主 軸 Bが回転しょ う とする ときの摩擦 トルク を測定できるよう に した。 鋼球体 3 の表面粗さは R a O . 0 2 mで一定と し、 超 高分子量ポリ エチレン製カ ッ プ 1 には、 凹面に前述の様な方法 で M P Cポリ マーの形成されたものと、 M P Cポリ マーの形成 されていない未処理のものを用いた。 この様な装置を用いて以 下の実験①〜③を行なった。
実験①
まず表 3 に示す様に押え付け荷重を変えて、 摩擦 トルクおよ び摩擦 トルク の変動 (一回転中での変動) を、 M P Cポ リ マー の形成有無別に調べた。 その結果を表 3 に併記する。

3 よ り 、 荷重の増加とと もに摩擦 トルクが大きく なる傾向 は、 M P Cポ リ マー形成の有無に関係な く みられるが、 M P C ポ リ を形成した場合の方が、 摩擦 トルクの絶対値は極めて 小さ く 、 かつ摩擦 トルク の変動もほとんどないこ とか ら、 M P
Cポリ マー形成によ り摩擦軽減効果が発揮されている こ とがわ かる
実験②
潤滑液と しての純水を供給せずに稼動させ、 試験途中で骨頭 3 とカ ッ プ (臼蓋) 1 の間に潤滑液と して純水を供給した後の 摩擦 トルクの変動を M P Cポ リ マーの形成有無別に調べた。 尚 、 押え付け荷重は 9 8 Nで一定と した。 その結果を図 3 に示す 図 3 よ り 、 M P Cポ リ マーの形成されていない超高分子量ポ リ エチレンま まの場合、 回転数 (摺動距離) の増加に伴い摩擦 トルクが増大し、 途中で純水を供給する と摩擦 トルク は一時的 に減少するが、 再び摩擦 トルクが急激に増大している。 これに 対し、 M P Cポ リ マーの形成された超高分子量ポ リ エチレン製 カ ッ プでは、 摺動開始から摩擦 トルクは増加してい く が、 途中 で純水を供給する と急激に摩擦 トルクは減少し、 その後は低 ト ルク を維持している こ とがわかる。 これは、 形成された M P C ポリ マーに純水が保持されて良好な潤滑状態を長時間確保する こ とができ、 摩擦が小さ く なるためと考え られる。
実験③
次に、 回転数を増加させて摺動距離を長く した場合の摩擦 ト ルク の変動を、 M P Cポリ マーの形成有無別に調べた。
実験では、 骨頭 3 と'カ ッ プ (臼蓋) 1 の間に潤滑液と して純 水を 5 . e m L Zm i nの割合で供給しつつ摺動させた。 尚、 押え付け荷重は 9 8 Nで一定と した。 実験結果を、 M P Cポリ マーの形成有無別に示した摺動回転数と摩擦 トルク の関係と し て図 4 に示す。
この図 4よ り 、 M P Cポ リ マーが形成された超高分子量ポ リ エチレン製カ ッ プでは、 摺動回数に関係なく低 トルク を維持し ている こ とがわかる。 これに対し、 M P Cポリ マーを形成して いない未処理の超高分子量ポ リ エチレン製カ ッ プでは、 摺動開 始か ら回転数と と もに摩擦 トルクが増加し、 しばら く 高摩擦 ト ルク を維持した後、 一旦減少する傾向が見られた。 尚、 こ の様 に M P Cポ リ マー形成なしの場合に摩擦 トルクが減少したのは 、 摺動面になじみができて、 摩耗現象におけるいわゆる定常摩 耗域に達したためと考え られる。 しかしながら実際の使用では 、 こ の様な領域に達するまでに摩耗粉が多量に発生し、 前記ル ーズニングが生じるため好ま し く ない。 この図 4 にて、 M P C ポ リ マーを形成した場合の 1 0 0万回摺動回転後の摩擦 トルク は、 M P Cポ リ マ一形成なしの場合の 4分の 1程度と長時間使 用後においても摩擦が著し く 抑え られている こ とが分かる。 実施例 IV : 回転揺動摩耗試験 ( 2 )
人工関節部材を形成する高分子物質の架橋処理が、 該部 " の 耐摩耗性向上に有効である こ とを確認する実験を行った。
人工関節のモデルと して、 臼蓋には、 架橋処理した超高分子 量ポ リ エチレンカ ッ プまたは該処理を施していない超高分子量 ポ リ エチレンカ ッ プに、 前述の方法で M P Cポ リ マーを形成し たものを用い、 骨頭には市販のコバル ト ク ロム合金 (直径 2 2 mm) を用いて、 回転揺動摩耗試験を実施した。 尚、 前記架橋 処理は、 強度が 5 M r a d の ァ線を 3 2 0 分間超高分子量ポリ エチレン製カ ッ プに照射して行った。
前記摩耗試験は M T C社製の摩耗試験装置を用いて行った。 図 5 は該装置の概略側面図を示すものである。 本実験では、 図 5 の下部主軸 XI— XI に対して 4 5 ° の傾斜で取 り付けた面板 上に、 超高分子量ポリ エチレン製カ ッ プ 1 1 を固定し、 上部主 軸 XII _ XII 上に取り付けた骨頭 1 2 を該カ ッ プ 1 1 の凹部に 押し当て、 下部主軸 XIII— XIII を回転させて該カ ッ プ 1 1 の 摩耗量を測定した。 押え付け荷重は 2 7 4 4 N ( 2 8 0 k g f ) と し、 回転周期は 1 H z と し、 また 1 周期の荷重変動は P o u 1 の歩行カーブにあわせた。
本実験では、 生体内を模擬した状態での上記カ ッ プ 1 1 の耐 摩耗性を評価するため、 上記カ ッ プ 1 1 および骨頭 1 2 を 3 7 X:の一定温度に制御した牛血清 ( 2 5 質量% ) 中に浸潰させて 前記摩耗試験を行った。 尚、 上記牛血清は、 回転サイ クル 5 0 万回ごとに入れ替えた。
こ の様に、 架橋処理が施されかつ M P Cポ リ マーの形成され た超高分子量ポ リ エチレンカ ップ 1 1 、 または架橋処理されて いないが M P Cポリ マーは形成されている超高分子量ポ リ ェチ レンカ ッ プ 1 1 のそれぞれについて摩耗試験を行い、 回転サイ クル 5 0 万回ごとに各カ ッ プ 1 1 の質量を測定し、 摩耗試験前 のカ ッ プの質量との差から、 カ ッ プ 1 1 の摩耗量を求めた。 そ の結果を回転サイ クル回数とカ ッ プの摩耗量との関係と して図 6 に示す。
この図 6 から、 架橋処理した超高分子量ポ リ エチレンカ ッ プ を用いれば、 回転サイ クル数が 3 0 0 万回と摺動距離が非常に 長く ても、 該カ ップの摩耗量は極微量である こ とがわかる。 即 ち、 架橋処理した高分子材料を人工関節部材のベースに使用す れば、 M P Cポ リ マーの形成による優れた耐摩耗性を更に高め る こ とができ有効なのである。 産業上の利用可能性
本発明は以上のよ う に構成されており 、 長期間に渡って良好 な潤滑状態が維持され、 その結果、 摩耗が極めて小さ く 、 かつ 衝撃吸収機能も備えた人工関節部材を提供できるよ う になった 。 そして、 この様な人工関節部材を用いた人工関節を使用する こ とによって、 患者は長期間に渡り健康人と相変わらぬ日常生 活を営むこ とができる。

Claims

請求の範囲
1 . 高分子物質で形成される人工関節部材の摺動面がホスホリ ルコ リ ン基を有する高分子か らなる こ と を特徴とする高分子材 料製人工関節部材。
2 . 前記ホスホ リルコ リ ン基が (メタ) ァク リ ロイルォキシァ ルキル基を介して前記高分子物質に結合されたものである請求 項 1 に記載の高分子材料製人工関節部材。
3 . 前記高分子物質に、 ホスホ リ ルコ リ ン基を有する重合性単 量体がグラフ ト結合されている請求項 2 に記載の高分子材料製 人工関節部材。
4 . 前記高分子物質に、 ホスホ リ ルコ リ ン含有化合物の単独重 合体または共重合体がコ 一ティ ングされている請求項 1 に記載 の高分子材料製人工関節部材。
5 . 前記ホスホ リルコ リ ン含有化合物が (メ タ) ァク リ ロイル ォキシアルキルホスホリ ルコ リ ンである請求項 4 に記載の高分 子材料製人工関節部材。
6 . 前記高分子物質が分子量 1 0 0 万以上の超高分子量ポ リ エ チレンである請求項 1 〜 5 のいずれかに記載の高分子材料製人 ェ関節部材。
7 . 前記高分子物質が架橋処理されたものである請求項 1 〜 6 のいずれかに記載の高分子材料製人工関節部材。
8 . 請求項 1 〜 7 のいずれかに記載の高分子材料製人工関節部 材を用いて得られる人工関節。
