Abstract

The invention relates to a method for identifying specific η-secretase inhibitors which can be used for treating neuro-degenerative diseases, in particular to a method which can be carried out in vitro. The invention also relates to a test kit which can be used for said method, in addition to the use of this test kit or method to identify substances which specifically inhibit η-secretase. A further embodiment relates to the use of these substances to produce a medicament for treating neuro-degenerative diseases and to pharmaceutical formulations which contain said substances.

Ferner betrifit diese Erfindung einen Testkit, mit dem das erfindungsgemäße Verfahren durchgeführt werden kann, sowie die Verwendung dieses Testkits oder des Verfahrens zum Auffinden von Substanzen, die spezifisch γ-Sekretase inhibieren. Hintergrund der Erfindung Aggregation und Präzipitation von Proteinen sind an der Entstehung von verschiedenen neurodegenerativen Erkrankungen wie Alzheimer, Parkinson und Veitstanz (Engl.: „Chorea Huntington") beteiligt. Bei der Alzheimer Erkrankung aggregiert das Amyloid-ß-peptid (Aß) und führt zu unlöslichen senilen Plaques, welche einen der pathologischen Marker der Erkrankung darstellen (Selkoe et al, 1996). Aß entsteht durch proteolytische Spaltung eines Vorläuferproteins, dem Amyloid- Vorläuferprotein (Engl.: „amyloid precursor protein"; Engl.: APP). Im nicht amyloidogenen Metabolismus des APPs spaltet die α-Sekretase innerhalb der Aß Region des APPs und führt damit zur Sekretion des löslichen N-terminalen Bereiches des Proteins (α- APPs) sowie nach erfolgtem γ-Sekretase-Schnitt zur Freisetzung von p3. Dagegen führt der amyloidogene Weg zur Enstehung von Aß, indem zwei Proteasen den N-Terminus (ß-Sekretase) bzw. den C-Terminus (γ-Sekretase) von Aß generieren (Haass, 1993; Selkoe, 1994). In vivo ist Aß im humanen Plasma und der Zerebrospinalflüssigkeit nachzuweisen. Auch in Zellkultur kann sekretiertes Aß im Zellkulturüberstand in verschiedenen Zelltypen nachgewiesen werden, die APP oder Fragmente davon endogen exprimieren oder überexprimieren. Sowohl Aß- Produktion und damit auch die Amyloid-Plaqueentstehung werden durch verschiedene genetische Risikofaktoren beeinflußt. Dazu gehören Mutationen in den homologen Proteinen Presenilin 1 und Presenilin 2 als auch im APP selbst. Die Analyse dieser Mutationen an Fibroblasten von Alzheimer-Patienten mit familiär vererbter Alzheimerkrankheit (Engl.: „Familiär Alzheimer Disease" (FAD)), zeigten den Einfluß, den sie auf die Aß-Entstehung haben. Dies konnte durch Untersuchungen an transfizierten Zellen und transgenen Tieren bestätigt werden. Alle Mutationen erhöhen die Produktion von Aß und führen im Fall der Presenilin Mutationen zur selektiven Erhöhung der längeren Aß Variante, dem Aß42 (Selkoe, 1996; Price, 1998). Dieses Peptid aggregiert im höheren Maße als die kürzere Form, dem Aß40, und wird in den diffusen Plaques und zusammen mit Aß40 in den senilen Plaques gefunden (Lemere et al., 1996; Mann et al., 1996). Neben diesem Einfluß der Mutationen gibt es Hinweise, daß auch die Wildtypform der Preseniline eine fundamentale Funktion in der physiologischen Entstehung von Aß haben. In Neuronen von Mausembryonen, bei denen das PS 1 -Gen (PS: Presenilin) auf gentechnischen Wege ausgeschaltet wurde, konnte eine drastische Reduktion von Aß 40 und Aß 42 nachgewiesen werden. Außerdem akkumulieren in diesen Zellen die C-terminalen Fragmente des APPs, welches zu der Ansicht führte, die Preseniline aktivieren die γ-Sekretase oder besitzen selbst γ-Sekretase Aktivität (De Strooper et al., 1998; Sisodia et al., 1998). Erste in vitro Testsysteme kombiniert mit Mutationsstudien an konservierten Aspartaten des Presenilin 1 legen die Vermutung nahe, daß die Preseniline spezielle autokatalytisch aktive Aspartatproteasen sein könnten, die für den γ-Sekretase Schnitt in der Membran verantwortlich sind (Wolfe et al., 1999). Die Diskussion über die Identifizierung der γ-Sekretase als entscheidender Schritt in der Generierung von Aß und damit in der Entstehung von Alzheimer ist bis heute nicht abgeschlossen. Unabhängig davon stellt diese Protease ein interessantes Ziel dar, um pharmakologisch in den Prozeß der Aß-Entstehung eingreifen zu können, indem man Inhibitoren findet, die selektiv ihre Aktivität reduzieren. Das erfindungsgemäße Verfahren verwendet gereinigte Membranen, die aus Zellen isoliert werden, die ein Substrat der γ-Sekretase nachweisbar exprimieren und γ-Sekretase-Aktivität aufweisen. Unter γ-Sekretase soll im Rahmen dieser Erfindung ein Protein verstanden werden, das die Eigenschaft hat, APP oder Fragmente davon proteolytisch zu spalten, insbesondere das C99- Peptid (Shoji et al., 1992), in p3 oder Aß. Somit sind alle Zellen für das erfindungsgemäße Verfahren geeignet, in denen der Fachmann mittels Western Blot und Verwendung spezifischer Antikörper ein Substrat der γ-Sekretase bzw. dessen Spaltprodukte nachweisen kann. Geeignete θ Membranen sind alle Membranen, in denen der Fachmann mittels Western Blot und Verwendung spezifischer Antikörper ein Substrat der γ-Sekretase nachweisen kann, bevorzugt lysosomale und endosomale Membranen, besonders bevorzugt mikrosomale Membranen. Verfahren zur spezifischen Aufreinigung von Membranen sind dem Fachmann bekannt aus Methods in Enzymology, Vol. 219, Titel: Reconstitution of intracellular Transport, und dem Buch „Biochemische Arbeitsmethoden", T.G Cooper, De Gruyter Verlag, 1981. Zur Ausführung des erfindungsgemäßen Verfahrens können, wie in einer nicht limitierenden Ausführungsform im Beispiel beschrieben, Zellen mit einer DNS-Sequenz transfiziert werden (DNS: Desoxyribonukleinsäure), die für ein Substrat der γ-Sekretase kodiert. Bei einer beispielhaften Ausführungsform kann durch den Entzug von Doxycyclin die Expression dieses Substrates induziert werden. Die Zellen können geöffnet werden und ein postnuklearer Überstand (Engl.: „post nuclear supematant"; Abk.: PNS) hergestellt werden, der weiter aufgearbeitet wird, um z.B. die mikrosomale Fraktion zu isolieren. Diese Fraktion kann unter geeigneten Bedingungen inkubiert werden und die Bildung des Produktes der Reaktion der γ-Sekretase mit einem geeigneten Substrat durch Immunpräzipitation und anschließendem Nachweis mittels geeigneter Antikörper im Western Blot Verfahren bestimmt werden. Das γ-Sekretase-Substrat konnte im PNS und in einer höheren Konzentration in der mikrosomalen Fraktion gefunden werden (Fig. 1A). Die C-terminalen Fragmente (CTF) von PSl und PS2 wurde auch in der SDI-Fraktion angereichert (Fig. 1B). Die mikrosomale Fraktion enthielt keine endosomalen Membranen, aber ER und Golgi Kompartimente waren angereichert (Fig. IC). De novo generiertes Aß konnte in den mikrosomalen Fraktionen gefunden werden, die bei 37 °C inkubiert worden waren (Fig. 2A). Inkubation bei 4 °C verhinderte die de novo Aß-Entstehung. Geringe Mengen an Aß waren durch die Existenz von intrazellulären Aß in den frisch präparierten mikrosomalen Fraktionen bedingt (Fig. 2A, Spur c). Die de novo Aß-Entstehung war zeitabhängig und erreichte ein Maximum nach 3 bis 4 Stunden Inkubation (Fig. 2B). Jedoch zeigte eine Evaluierung der Substrat/ Produkt- Beziehung, daß die in vitro Aß-Generierung keine effiziente proteolytische Reaktion war (Fig. 2A). Die Inkubation der mikrosomalen Membranen in Abwesenheit von EDTA fiihrte zu einer dramatischen Verminderung der in vitro Aß-Generierung (Fig. 3). Den gleichen Effekt hatte der Calciumchelator BAPTA. Dies legt die Vermutung nahe, daß eine in derselben Fraktion anwesende Ca2+-abhängige Protease Aß abbaut. Die Behandlung der mikrosomalen Membranen mit 1 M KCl im Puffer oder eine Extraktion dieser Membranen mit 0,1 M Na23 pH 11,5 vor der Durchführung des γ-Sekretase-Testsystems zeigt, daß die γ-Sekretase membranständig oder zumindest fest an die Membran gebunden sein muß (Fig. 4). In einer erfindungsgemäßen Ausführungsform werden die beschriebenen gereinigten Membranen insbesondere mikrosomale Membranen mit einem geeigneten Reaktionspuffer und einer Testsubstanz vermischt. Unter Testsubstanz soll im Rahmen dieser Erfindung jede Substanz verstanden werden, die daraufhin getestet werden soll, ob sie inhibitorisch auf die γ-Sekretase- Aktivität wirken könnte. Dann wird die Mischung unter Bedingungen inkubiert, unter denen das Substrat der γ-Sekretase bei Abwesenheit der Testsubstanz gespalten wird. Dann wird die Menge eines gebildeten Spaltprodukts bestimmt und der erhaltene Wert mit dem Wert verglichen, der in Abwesenheit der Testsubstanz erhalten wird. Mit dem erfindungsgemäßen γ-Sekretase- Testsystem können nun vorteilhafterweise spezifische γ-Sekretase-Inhibitoren identifiziert und validiert werden und von Inhibitoren unterschieden werden, die die Sekretion von Aß40 und Aß42 verhindern. Eine Verbindung wurde, wie im Beispiel beschrieben, mit dem erfindungsgemäßen Testsystem getestet. Die Verbindung A ([αS-(o_R*, γR*, δR*)] N-butyl-γ- hydroxy-α-(l-methylethyl)-δ-[(4-methylpentyl)an ino]cyclohexanhexanamid-hydrochlorid; siehe Beispiel 20 von EP 778 266 AI) wurde gemäß den Angaben im Beispiel 20 der Europäischen Patentanmeldung EP 778 266 AI hergestellt und zeigte eine konzentrationsabhängige Inhibition der Aß-Generierung mit einem IC50-Wert von etwa 6 μM (Fig. 6A). Ähnliche Resultate wurden in einem Testsystem ermittelt, das sekretiertes Aß40 und Aß42 in einem quantitativen ELISA mißt, der spezifisch für beide Aß-Peptide ist (Steiner et al., 1998). Hier wird eine Astrocytoma Zellinie (U373) benützt, die Wildtyp APP751 überexprimiert und detektierbare Mengen beider Aß-Spezies sekretiert. Eine konzentrationsabhängige Reduktion in der Menge an extrazellulären Aß40 und Aß42 konnte nach einer Übernachtbehandlung mit der Verbindung A beobachtet werden (Fig. 6B). In einer Ausführungsform der Erfindung werden Zellen kultiviert, die ein endogenes Polypeptid exprimieren, das ein Substrat der γ-Sekretase ist. Unter dem Begriff endogen ist im Rahmen dieser Erfindung zu verstehen, daß diese Zelle oder Zelllinie das bezeichnete Polypeptid in einer ausreichenden Menge exprimiert, ohne daß dazu weitere Manipulationen, z.B. mittels gentechnologischer Methoden nötig sind. Im Rahmen der Erfindung soll unter einer ausreichenden Menge an Substrat der γ-Sekretase eine Menge verstanden werden, die in einer etablierten biochemischen Nachweismethode (z.B. ELISA, Western Blot) unter Verwendung spezifischer Antikörper ein nachweisbares, über dem Hintergrund liegendes Signal ergibt. In einer besonders bevorzugten Ausführungsform der Erfindung werden Zellen kultiviert, die ein exogenes Polypeptid exprimieren, das ein Substrat der γ-Sekretase ist. Unter dem Begriff exogen ist im Rahmen dieser Erfindung zu verstehen, daß diese Zelle oder Zelllinie mittels gentechnologischer Methoden so manipuliert wird, daß sie das γ-Sekretase-Substrat exprimiert. Enthält die Zelle oder Zellinie das γ-Sekretase-Substrat auch ohne die besagten Manipulationen, so soll mit diesem Begriff ausgedrückt werden, daß die Menge des γ-Sekretase-Substrats gegenüber dem Wert ohne Manipulation meßbar erhöht ist. Zur Herstellung einer das γ- Sekretase-Substrat exogen exprimierenden Zelle kann eine Nukleotidsequenz, die für die Aminosäuresequenz des γ-Sekretase-Substrats kodiert, in eine geeignete Expressionskassette eines eukaryontischen Expressionsvektors eingebracht werden. Geeignete Expressionskassetten haben einen in eukaryontischen Wirten funktioneilen Promoter wie z.B. den Cytomegalovirus- Promoter (CMV-Promoter) und ein funktionelles Polyadenylierungssignal z.B. vom SV40 Virus (Engl.: "Simian Virus"; Abk.: SV). Geeignete Expressionsvektoren sind Vektoren, die in eukaryontischen Wirten replizieren können, d.h. einen funktionellen Replikationsursprung (Engl. : "Origin of replication") besitzen. Femer ist auf dem selben Plasmid, einem weiteren Plasmid oder chromosomal integriert eine weitere Nukleotidsequenz zu finden, die für ein Fusionsprotein zwischen dem Tet-Repressor und einer sauren, aktivierenden Domäne kodiert, das konstitutiv exprimierbar ist. Unter saurer, aktivierender Domäne ist eine Proteindomäne zu verstehen, die einen hohen Anteil an sauren Aminosäuren hat und die Eigenschaft besitzt, die Transkription eines Gens zu vermitteln, wenn die Domäne in geeigneter Position in den vor dem Gen lokalisierten Transkriptionskomplex eingebracht wird. Diese Eigenschaft kann durch den bekannten "One- hybrid-Assay" ermittelt werden. Bei dieser Methode wird eine DNS-bindende Domäne mit einer zu untersuchenden Domäne fusioniert, wobei die DNS-bindende Domäne an eine Sequenz bindet, die sich vor einem Reporterprotein befindet, und die Aktivität des besagten Reporterproteins gemessen wird (Clontech, Heidelberg). In dem oben beschriebenen Fall wird die Expression des γ- Sekretase-Substrats nicht stattfinden, wenn die Konzentration von Tetracyclin oder einem Derivat von Tetracyclin wie z.B. Doxycyclin in der Zelle einen bestimmten Wert überschreitet, da der Tetracyclin-Repressor Tetracyclin oder das Derivat davon bindet, infolgedessen nicht an seine Bindungsstelle in der DNS bindet und daher nicht die Transkription des hinter diese Bindungsstelle geschalteten Gens induziert, das für das γ-Sekretase-Substrat kodiert. In Abwesenheit von Tetracyclin oder einem Derivat davon bindet das Fusionsprotein zwischen Tet- Repressor und saurer Aktivierungsdomäne an seine DNS-Bindungsstelle und induziert die Transkription des dahinter geschalteten Gens, das für das γ-Sekretase-Substrat kodiert. Damit ist gewährleistet, daß eine gesteuerte und gezielte Expression des γ-Sekretase-Substrats stattfindet. In einer Ausführungsform der Erfindung ist das γ-Sekretase-Substrat das Amyloid- Vorläuferprotein (APP; engl.: "amyloid precursor protein") oder ein Fragment davon, sofern es die γ-Sekretase Schnittstelle beinhaltet.. In einer bevorzugten Ausführungsform der Erfindung ist das γ-Sekretase-Substrat das C99-Fragment des Amyloid- Vorläufer-Proteins (Shoji et al., 1992). Das γ-Sekretase-Substrat kann ganz allgemein membranassoziert, bevorzugt aber membranständig, sein. Der Membranstabilisator im Rahmen dieser Erfindung ist bevorzugt Saccharose oder Sorbitol, wobei der Fachmann diese jedoch durch gleichwirkende Substanzen ersetzen kann.
