The invention concerns an electrochemical process for the quantitative determination of binding proteins in which the binding protein is a component of an antibody-conjugate system applied to an electrode. Die Bestimmung von Bindungsproteinen besitzt in der klinischen Diagnostik eine besondere Relevanz. Bei¬ spielhaft hierbei sei die Bestimmung von kardialen humanen Fettsäurebindungsproteinen (H-FABP) genannt. Insbesondere die Bestimmung von FABP besitzt in der Diagnostik besondere Bedeutung, da im Falle von Herz¬ muskelschädigungen kardiales FABP (H-FABP) aus dem Herzmuskel austritt und nach wenigen Stunden im Blut nachweisbar ist. Aus diesem Grund ist auch FABP als Herzinfarktmarker anzusehen. Einem schnellen und ge- nauen Verfahren zum Nachweis von FABP kommt deshalb besondere Bedeutung zu. Klassische Methoden der FABP- Bestimmung beruhen auf ELISA- oder RIA-Verfahren. Neben diesen klassischen Methoden ist weiterhin in Spener, F. et al. , 1992, Dechema Biotechnol. Conf. ein amperometrisches Verfahren zur Bestimmung von FABP bekannt. Dieses Verfahren beruht auf einer Immo¬ bilisierung von Fänger-Antikörpern auf einer Membran und einer Konkurrenzreaktion zwischen FABP der Probe und zugesetztem Katalase gelabelten FABP. Nachteilig bei diesem Konkurrenzverfahren ist jedoch die Anzahl der nötigen Verfahrensschritte, die aufwendige Präpa¬ ration der Antikörpermembranen, die sehr langen Ana¬ lysezeiten, die nicht ausreichende Empfindlichkeit, der bedingte Einsatz in reinem Plasma oder Blut und die Höhe der unspezifischen Meßwerte.The determination of binding proteins is of particular relevance in clinical diagnostics. Wesentlich beim anmeldungsgemäßen Verfahren ist die Verwendung einer Einmalgraphitelektrode und die Immo¬ bilisierung des Antikörpers direkt auf der Elektro¬ den-Oberfläche. Dadurch steht eine Elektrode zur Ver¬ fügung, die in einfacher Weise für das erfindungsge¬ mäße Meßverfahren eingesetzt werden kann. Besonders hervorzuheben ist, daß, wenn die mit Antikörpern im¬ mobilisierte Elektrode z.B. mit einer BSA-Lösung be¬ handelt und getrocknet wird, über eine längere Zeit problemlos gelagert werden kann. Es hat sich dabei gezeigt, daß die so präparierten Graphitelektroden selbst über mehrere Wochen bei 4 °C stabil sind. Be¬ sonders bevorzugt ist es, wenn als Einmalelektrode eine auf einem Trägermaterial mit Siebdruck aufge¬ brachte Graphitschicht eingesetzt wird. Als Trägerma¬ terial ist hierbei besonders eine Laminierfolie be- vorzugt. Verfahrensmäßig wird dabei so vorgegangen, daß z.B. ein Leit-C-Spezialkleber für die Elektronen- mikroskopie mit einem Siebdrucköl versetzt und durch eine Schablone oder durch ein Sieb mit entsprechender Elektrodenform auf eine Laminierfolie aufgebracht wird. In entsprechender Weise kann auch die Referen¬ zelektrode hergestellt werden, indem z.B. eine Sil¬ ber/Silberchloridpaste auf eine weitere Laminierfolie aufgebracht oder zusammen mit der Graphitelektrode auf eine Folie aufgebracht wird. Die¬ se letzte Variante hat den Vorteil, daß das Mischen von Konjugat und Bindungsprotein vor der Messung ent¬ fällt und somit ein Pipettierschritt eingespart wird. Die mit Konjugat und Antikörper beschichteten Elek¬ troden sind im übrigen mehrere Wochen stabil. Die Durchführung der Messung ist in sehr einfacher Weise möglich. Die in Abhängigkeit zur Konzentration an Bindungsprotein bindet nämlich in der alkalischen Phosphatase Bindungsprotein-Antikörper und der immo¬ bilisierte Bindungsprotein-Antikörper an das Bin¬ dungsprotein. Das amperometrische Stromsignal entwickelt sich in ca. 10 bis 20 sec bei 300 mV gegen Silber/Silber¬ chlorid und wird entsprechend der Kalibrationskurve in die Konzentration von FABP umgerechnet.
