ノ ィ ォセ ン サおよびそ の製造法
技術分野
本発明は、 種々の生体試料液中の特定成分を高精度で、 迅速か つ容易に定量する こ と のでき るバイ ォセ ン サおよびその製造法 に関する も のである。
背景技術
近年、 酵素の有する特異的触媒作用を利用し た種々のバイ オ セ ン サが開発され、 特に臨床分野への応用が試みられてい る、 検査項目及び検体数が増加してい る現在、 迅速に精度よ く 測定 で き るバイ ォセ ン サが望まれている。
グル コ ース セ ン サを例に と る と、 糖尿病の増加が著し い今日、 血液中の血糖値を測定し管理する には、 従来のよ う に血液を遠 心分離し て血漿を測定する のでは非常に時間を要する ため、 全 血で測定でき るセ ン サが要求されている。 簡易型と し ては、 尿 検査の時に使用されてい る検査紙と 同様に、 ス テ ィ ッ ク状の支 持体に糖 (グルコ ー ス) にのみ反応する酵素と、 酵素反応時ま たは酵素反応の生成物によ り 変化する色素と を含有する担体を 設置し たも のがある。 こ の担体上に血液を滴下し、 一定時間後 の色素の変化を目視または光学的に測定する方式であ るが、 血 液中の着色物によ る妨害が大き く 精度は低い。
そ こ で、 第 1 図のよ う な多層式の分析担体が提案されてい る (実開昭 5 4 — 1 7 8 4 9 5号公報) 。 こ れは透明な支持体 5 1 の上に試薬層 5 2、 展開層 5 3、 防水層 5 4、 濾過層 5 5 が 順に積層された構造となってい る。 血液サ ン プ ルを上部か ら滴 下する と、 まず濾過層 5 5によ り血液中の赤血球、 血小板など の固形成分が除去され、 防水層 5 4にある小孔 5 6か ら展開層 5 3へ均一に浸透し、 試薬層 5 2 において反応が進行する。 反 応終了後、 透明な支持体 5 1 を通して矢印の方向から光を照射 し、 比色分析によ り 基質濃度を測定する方式である。 従来の簡 易なステ ィ ッ ク状の担体と比較する と複雑な構造であ るが、 血 球除去な どによ り精度は向上した。 しかし、 血液の浸透および 反応に時間がかかる ため、 サンプル の乾燥を防 ぐ防氷層 5 4を 必要と し、 また、 反応を速める ために高温でのイ ンキュベー ト を要するなど、 装置および担体が複雑化する とい う問題がある。
—方、 血液などの生体試料中の特定成分について、 試料液の 希釈や撹拌などの操作を行なう こ とな く 高精度に定量する方式 と しては、 第 2図に すょ ぅ なパィ ォセ ン サが提案されている (例えば、 特開昭 5 9— 1 6 6 8 5 2号公報)。 こ のパイ ォ セ ン サは、 絶縁基板 6 3 に リ ー ド 6 1、 6 2 を各々有する 白金な どから なる測定極 6 4および対極 6 5を埋設し、 これら の電極 系の露出部分を酸化還元酵素および電子受容体を担持した多孔 体 6 6 で覆ったも のであ る。 試料液を多孔体上へ滴下する と、 試料液中に酸化 ¾元酵素と電子受容体が溶解し、 試料液中の基 質との間で酵素反応が進行し、 電子受容体が還元される。 反応 終了後、 こ の還元された電子受容体を電気化学的に酸化し、 こ の とき得られる酸化電流値から試料中の基質濃度を求める。
しかし、 こ のよ う な構成では、 多孔体については測定毎に取 り 替える こ とによ り簡単に測定に供する こ とができ るが、 電極 については洗浄等の操作が必要であ る。 一方、 電極系をも含め て測定毎の使い捨てが可能と なれば、 測定操作上、 極めて簡易 になる も のの、 白金等の電極材料や構成等の面か ら、 非常に高 価なも の にな ら ざる を得ない。 ま た白金電極の構成方法と して、 スパ ッ タ法ゃ蒸着法を用いる こ と もでき る 力 製造上高価なも の となる。
また、 電極系をも含めて使い捨てに し得る方式と し ては、 特 開昭 6 1 - 2 9 4 3 5 1 号公報に記載のバイ オセ ン サを提案し た。 こ のバイ オセ ン サは、 第 3 図に示すよ う に絶縁性の基板 7 1 の上に ス ク リ ー ン 印刷等の方法で 力一 ボ ン な どから な る電極 系 7 2 ( 7 2 ' )、 7 3 ( 7 3 ' )、 7 4 ( 7 4 ' ) を形成し、 絶縁層 7 5 を設けた後、 電極系の上を酸化還元酵素と電子受容 体を担持した多孔体 7 7 で覆い保持枠 7 6 と カバー 7 8 で全体 を一体化し たも のであ る。 試料液を多孔体上へ滴下する と、 多 孔体に担持されてい る,.酸化還元酵素と電子受容体が試料液に溶 解し、 試料液中の基質と の間で酵素反応が進行し電子受容体が 還元される。 反応終了後、 こ の還元された電子受容体を電気化 学的に酸化し、 こ の と き得られる酸化電流値か ら試料液中の基 質濃度を求め る。
以上の様な従来の構成では、 電極系を含む基板面の濡れが必 ずしも一様と なら ないため、 多孔体と基板と の間に気泡が残り、 応答電流に影響を与えた り反応速度が低下する場合があ っ た。 ま た、 周囲の湿度が低い と き には測定中に試料液の水分が蒸発 し、 応答の変動が見受け られた。 さ ら に、 試料液中に電極に吸 着しやすい物質ゃ ァ ス コ ル ビ ン酸な どの酸化されやすい物質が 存在する と、 セ ン サ の応答に影響が見受け られた。
そ こ で、 血液などの生体試料中の特定成分を簡易かつ迅速、 高精度に測定するためのセ ン サ と しては、 微量の試料波を予め 希釈あるいは定量する こ とな く 単にセ ン サ に添加する だけで測 定値が得られるも のが望まれる。 さ らには、 洗铮、 拭き取り な どの操作を伴わない、 デ イ スポーザブルタ イ プのものが望まれ る
発明の開示
本凳明のバイ オセ ン サは、 絶緣性の基板上に少な く と も剠定 極と対極から なる電極系と該電極系の上に酵素と電子受容体を 含む反応層を設け、 さ ら に力パーと一体化する こ とによ り、 該 反応層を含む空間部を形成し、 前記空間部に被検液を導入する 導入口と前記空間部の気体を前記被検液の流入によっ て排出す る排出口を設け、 前記酵素と前記被検液の反応に際しての物質 濃度変化を前記電極系で検知し、 前記被検液中の基質濃度を測 定するも のである。
また、 電極系を複数組の電極系から構成し、 これら複数組の電 極系に各々対応した反応層を設ける こ とによ り 独立したセ ン サ 系を構成する こ とができ る。 これによ り、 2種以上の基質濃度 を同時に測定する こ とができ る。 また、 2組の電極系を用い、 一方の電極系を測定用に、 他方の電極系をプ ラ ン ク用に用い、 雨電極系の応答差を求め る こ とによ り、 ァ ス コ ル ビ ン酸などに よる妨害を除去する こ とができ る。
また、 基板と電極系と反応層および力パーを一体化して空間 部を設け、 該空間部を構成する部材の表面を親水性とする こ と によ り、 微量の試料液にセ ン サ の導入口を接する だけの簡易な 操作で上記空間部内に一定量の試料液を速やかに導入する こ と ができ る。
導入された試料液は、 反応層の電子受容体や酵素、 親水性高 分子を溶解し粘調な液と な り ながら酵素反応が進行する。 酵素 反応によ り試料液中の基質濃度に対応した還元型の電子受容体 が生成する。 次に、 こ の還元型の電子受容体を測定極上で電気 化学的に酸化する こ と に よ り、 その酸化電流値から基質濃度を 知る こ と がで きる。
本発明の ィ ォ セ ン サ においては、 親水性高分子を用い る こ と によ り、 血液サ ン プル中の赤血球など、 固形成分の電極への 影響を防止する こ と ができ る と と も に、 さ ら に、 空間部を設け る こ と によ り 電極上の液の流動を効果的に防止する こ と ができ る ため、 測定中の振動による影響を防止で き る。
一方、 本発明のバイ オセ ン サ の反応層は、 酵素層き ら には こ の上に設けた電子受容体層から構成されるが、 さ らに これら両 層の間に親水性高分子層を設け る こ とによ り酵素と電子受容体 を分離し、 保存性に優れたバイ オセ ン サを提供する も のであ る。
本発明によれば、 電極系をも含めたデ ィ ス ポーザブルタ イ プ のバイ オセ ン サを構成する こ と がで き、 試料中の基質濃度、 た と えば血液中のグル コ ー ス濃度につ いて、 予め試料の希釈ゃ定 量をする こ と な く、 極めて微量の試料液を添加するだけの簡易 な操作で迅速かつ高精度に測定する こ とができ る。

発明を実施するため の最良の形態
実施例 1
以下の実施例の説明用図面において、 共通す 要素には同一 番号を付し、 一部説明を省略する。
パイ ォセ ン サの一例と して、 グル コ ー ス セ ン サ について説明 する。 第 4図は本癸明のバイ オセ ン サの一実施例として作製し たグル コ ー ス セ ン サ の分解斜視図であ り、 第 5 図はその外観図、 また第 6図は、 第 5図に示すバイ オセ ン サを長手方向に中央部 で切断した場合の断面図である。 なお、 第 4図には、 第 6図に 示す反応層 1 4は示されていない。
以下に、 セ ン サの作製方法に つい て説明する。 ポ リ エ チ レ ン テ レ フ タ レ ー トから なる絶縁性の基板 1 に、 ス ク リ ー ン印刷に よ り銀ペース ト を印刷し リ ー ド 2, 3 ( 3 ' ) を形成する。 次 に、 樹脂バィ ンダーを含む導電性カーボ ンペー ス トを印刷し、 加熱乾燥する こ と に よ り、 測定極 4、 対極 5 C 5 ' ) から なる 電極系を形成する。 さ ら に、 電極系を部分的に覆い、 電極の露 出部分の面積を一定どし、 かつ リ ー ド の不要部を覆う よ う に絶 縁性ペー ス ト を印刷し、 加熱処理を して絶縁層 6 を形成する。
次に、 電極系 4、 5 ( 5 ' ) の露出部分を研磨後、 空気中で 1 0 0 °Cにて 4時間熱処理を施し た。 こ の よ う に し て電極部分 を構成し た後、 親水性高分子と し て、 カル ボキ シ メ チルセル 口 ー ス (以下 C M C と 略す) の 0. 5 wt%水溶液を電極上へ展開、 乾燥し C M C層を形成する。 そ して こ の C M C層を覆 う よ う に、 酵素と し て グルコ ー ス ォ キ シ ダ一ゼ ( G O D ) を リ ン 酸緩衝液 に溶解し たも のを展開し、 乾燥させ、 C M C — G O D層か ら な る反応層 1 4を形成した。 こ の場合、 C M C と G O Dは部分的 に混合された状態で厚さ数 ミ ク 口 ン の薄膜状と な っている。
次に、 こ の基板 1 と樹脂扳か ら な る スぺ ーサ 7 と カバ一 9の 3 つの部材について、 第 4図に示す様に、 各部材間が破線で示 す位置関係になる よ う に接着し、 第 5図の外観図に示すよ う に 一体化し た。 こ こ で、 ス ぺーサは約 3 0 0 〃 mの厚みを有し、 そ の中央部を幅 2 (^、 長さ 7™の U ^型に切 り と つた形状をし てお り、 切 り と られた部分の端部は一体化し た と きに試料液の 導入口 1 0 と な り、 中央部は空間部 8を形成する。 ま た、 カバ 一 9は直径 2 の穴を有してお り、 一体化し た と き に排出口 1 1 となる。
上記のよ う に構成し たグル コ ー ス セ ン サの先端の導入口を試 料液であ る グルコ ー ス標準液 ( 2 0 0 m g Z d 1 ) に接触させ る と、 試料液は導入口 1 0よ り 内部へ導かれる。 こ の と き空間 部 8内の空気は排出口 1 1から速やかに排出 さ れ、 同時に空間 部は排出口近傍まで試料液で満たされた。 こ の よ う に試料液は 電極面上を速やかに拡がり空間部を満たすため、 気泡の残留は 認め られなか όた。
これは、 導入口と排出口を設ける こ と によ り 試料液の流れが —方向と な り、 また、 電極上に予め形成された親水性高分子層 により電極面の濡れが向上し、 気液の交換が円滑に行なわれる こ とによる も のと考えられる。
さ ら に、 力パーゃスぺーサなど、 空間部を構成する部材の表 面を界面活性—剤で予め処理して親水性とする こ とによ り、 さ ら に円滑な試料液の導入が可能であつ た。
一方、 添加された試料液は C M C を溶解し粘調な液体と な り、 試料液中のグル コ ー スは電極上に担持されたグル コ ー ス ォ キ シ ダーゼの作用で酸素と反応して過酸化水素を生成する。 そ こで、 試料液を導入して 1 分後に電極間に 1 Vの電圧を印加する こ と により、 測定極をア ノ ー ド方向へ分極した。 上記のア ノ ー ド方 向への電圧印加によ り、 生成し過酸化水素の酸化電流が得られ た。 こ の電流値は基質である グル コ ー ス の濃度に対応する。
上記構成になるセ ン サ の応答特性の一例と して、 電圧印加 5 秒後の電流値とグル コ ー ス濃度との関係を第 7図 Αに示すが、 良好な応答特性が得られた。
また、 上記と同一仕様のダルコ —センサス 3 0個につ い て同 じ試料液で試験した と こ ろ、 変動係数は約 5 % と良好であ っ た。
さ ら に、 第 3図に示す従来例の構成で試作し たグル コ ー ス セ ン サと とも に、 湿度が 3 0 % と 8 0 %の条件下で、 蒸発による 影響についてそれぞれ 1 0個のセ ン サを用いて比較検討し た こ ろ、 本発明になる グル コ ー ス セ ン サ においては 5分放置後に おいても その影響は無視し得る も のであ つ たが、 従来構成のも の については低湿度の方が 2分後において約 3 %高値を示し た c 以上のよ う に、 本発明にな る上記構成の グル コ ー ス セ ン サは、 微量の試料液をすみやかに反応層領域へ吸入し、 周囲の湿度等 による影響を受ける こ と な く、 簡易、 迅速かつ高精度に ダル コ ー ス濃度を測定する こ と ができ る。
実施例 2
C M C — G 0 D層を形成する までは実施例 1 と全 く 同様に作 製した後、 さ らに以下に示す方法で C M C — G O D —電子受容 体層から なる反応層を形成し た。
ト ル エ ン に界面活性剤と し て レ シ チ ン ( ホ ス フ ァ チ ジ ル コ リ ン ) を溶解して l w t %溶液を調製し、 これに電子受容体と し て フ ユ リ シ ア ン化力 リ ゥ ム の微結晶 (平均粒径 1 0 ^ ™以下) をたも のを分散し、 得られた分散液を C M C — G O D層上へ展 開した。 こ の後、 ト ル エ ンを蒸発さ せて、 C M C — G O D —電 子受容体層か らなる反応層を電極系上に形成し、 さ ら に実施例
1 と同様に し てス ぺ ーサ、 カバー と と も に一体化し、 第 5 図に 示す構造のグル コ ー ス セ ン サ と し た。
次に、 前記同様に してセ ン サ先端部分よ り グル コ ー ス標準液 を導入し、 約 1 分後に両極間に 6 0 0 m V の電圧を印加する こ と によ り、 酵素反応で生成し た フ Λ ロ シ ア ンィヒカ リ ゥ ム を測定 極で酸化し、 この と き の電圧印加 5秒後の電流値を測定し た。 こ の結果、 第 7図 Β に示す様に グル コ ー ス濃度と応答電流値の 間に極めて良好な直線関係が得られた。 ま た、 3 0個のセ ン サ を用いて変動係数を調べたと こ ろ、 グルコ ー ス標準液について
2. 6 %、 全血試料について 3 %と極めて良好な値を示し た。 また、 蒸発の影響につい'て実 S 例 i と同様に試験した と こ ろ、 前記同様の効果が得られた。
上記実施例において、 レ シ チ ン の濃度が 0 . 0 1 w t %以上にな る と フ ェ リ シ ア ン化力 リ ゥ ムが う ま く ト ル エ ン中で分散するた め滴下が容易とな り、 薄膜状の フ ェ リ シ ア ンィ匕カ リ ウ ム一レ シ チ ン層が形成できた。 レ シチ ンがない場合は、 フ ヱ リ シ ア ンィ匕 力 リ ゥ ム層が不均一に形成された り基 を曲げる とはか'れる と い う欠点が見られたが、 レ シ チ ンを添加する こ と によ り均一で は がれに く いフ · シ ア ン化力 リ ゥ ム層が容易に形成でき た。 レ シチ ン の濃度が高 く なる と と もに、 フ ェ リ シ ア ン化カ リ ウ ム 層がはがれに く く なるが、 フ ェ リ シ ア ン化カ リ ゥ ム の溶解速度 も落ちるため、 0 .0 1 - 3 w t %が適当と考え られる。 レ シ チ ン の かわ り にポ リ エチ レ ン グ リ コ ー ル ア ルキル フ エ ニルエ ー テ ル ( 商品名: ト リ ト ン X ) を用いた と こ ろ、 フ リ シ ァ ン ィ匕 力 リ ゥ ム の微粒子を ト ルェ ン中に分散させるためには 0 . 1 %以上必要で あ つたが、 レ シ チ ン と同様に良好な フ ユ リ シ ア ンィ匕カ リ ゥ ム層 が形成できた。 界面活性剤と しては、 前記の例の他に、 ォ レ イ ン 酸ゃ ポ リ オ キ シ エチ レ ン グ リ セ リ ン脂肪酸エ ス テルゃ シ ク ロ デキス ト リ ン など、 電子受容体を有機溶媒に分散させ、 かつ酵 素活性に影響をおよぼさないも の であれば、 特に制限される こ と-はない。 また、 電子受容体を混合する有機溶 と し ては、 ト ル ェ ンや石油ェ一テルな ど、 G O D活性および印刷電極への影 響の少ないも のであればよい。 上記に用い た フ ヱ リ シ ア ン 化 カ リ ウ ム微結晶の粒径に ついて は、 市販の フ リ シ ア ンィヒカ リ ウ ム の結晶を粉砕 し、 ふる いに よ り所定の粒径の も の を集めて フ リ シ ア ン ィ匕 カ リ ゥ ム層を形 成し、 各種の粒径の も の で作成 し た前記同様の グル コ ー ス セ ン ザについて応答を比較し た。 第 1 5 図は、 横軸にふる いの メ ッ シ ュ の大き さ、 縦軸に グル コ ー ス 4 0 0 m g Z d 1 に対す る反 応終了時間を示し た。 ( ) の中は穴の径 ( m ) を表わ し て い る。 第 1 5 図に示すよ う に細かい粒径の方が速やかに溶け反 応終了に必要な時間が短か つ た。 1 4 5 メ ッ シ ュ ( 日 本二業規 格) を通過し た フ リ シ ア ンィヒ カ リ ウ ム (粒径 1 0 0 m以下) で作製し たセ ン サは、 2 分以内に反応が終了 し た。 さ ら に、 フ X リ シ ア ンィヒ カ リ ゥ ム層を作製する と き粒径が小さ い方が均一 に膜がで き応答のば ら つ き が少なか つ た。 フ X リ シ ア ンィ匕 カ リ ゥ ムの微結晶は粉砕でも作製で き る が、 フ ユ リ シ ア ンィヒカ リ ウ ム の水溶液を エ タ ノ ール中で再結晶 させる と簡易に 1 0 / m 以下 の粒径が作成でき、 フ ユ リ シ ア ン化 カ リ ウ ム層を形成 さ せ る と 密な膜と な り、 反応終了時間も短縮で き た。
1 0 0 m以下に微粒化し た フ ヱ リ シ ア ンィヒ カ リ ゥ ム を ト ル エ ン に混ぜて滴下する と、 ト ル エ ン がすみやかに気化し、 微粒子の ま ま の フ ヱ リ シ ア ン ィヒカ リ ウ ム層が形成で き、 溶解速度も 速 く 迅速に測定で き た。 さ ら に有機溶媒を用い る こ と に よ り、 C M
C一 G O D層の上に フ ユ リ シ ア ンィヒ カ リ ゥ ム の層を分離し て形 成でき、 保存性を向上す る こ と がで き た。
上記実施例において、 導入口及び排出口 を設け る に あた っ て は、 第 4 図〜第 6 図に示し た配置以外に、 第 8 図の分解斜視図 および第 9図の外観図に示すよ う に、 カバー 9 と基板 1 にそれ ぞれ穴を設けて導入口 1 0 と排出口 1 1 と しても よい。
また、 第 1 0図の分解斜視図に示すよう に、 スぺ—サを 7 と 7 ' の 2 つに分ける こ とによ り、 空間部 8 と導入口 1 0及び排 出口 1 1 とする こ ともできる。
さ らに、 第 5図に示す構造のバイ オセ ン サ の先端部分の形状 を変えて、 丸みを持つよ う にし たも のについて、 その外観図を 第 1 1 図に示す。 こ うする こ と によ り、 試料液を導入する にあ たって接液する部分がほぼ導入口 1 0 に限定されるため、 導入 口周囲に試料液の回り''込みがな く、 さ ら に少量の試料液に対し ても円滑に導入する こ とができ た。
実施例 3
実施例 1 と 同様の方法によ り、 第 1 2図に示す電極部を構成 し た。 すなわち、 基板上に銀リ ー ド 3、 2 1、 2 2、 2 3 と こ れらに対応する対極 5及び 3 ケ の測定極 4 1、 4 2、 4 3 を形 成する こ と によ り、 対極を共有する 3組の電極系を構成した。 次に、 各々の測定極近傍に実施例 2 と同様にして C M C— G 0 D — フ ヱ リ シ ア ン化力 リ ゥ ム層から なる反応層を形成した。
次に、 スぺーサおよび力パ ーを実施例 1 と同様に接着して一 体化した。 こ こで、 ス ぺーサはその一部が 「 く し状」 に切 り と られてお り、 こ の切 り と られた部分が空間部 8 を形成している。 こ の空間部はそれぞれの電極系上に分割されているが、 全体と しては連続しており、 共通の空間部となっ てい る。 また、 端部 は導入口 1 0 となる。
これら の各電極系上に分割されて構成された空間部に対応し て、 カバー 9 には 3 個の穴をあ け排出口 1 1、 1 2、 1 3 を設 けている。 導入口 1 0 を試料液に接触させる と、 3個の排出口 よ り空間部内の空気が排出 されなが ら試料液は空間部内へ速や かに導入 される。
実施例 2 と 同様に して、 グル コ ー ス標準液に対する応答を 3 個の電極系について測定し、 その平均値を求めて 1 個のセ ン サ の応答値と し た。 これを、 3 0 個の同一仕様のグルコ ー ス セ ン サにっき試験した と こ ろ、 変動係数は 2 % と良好な値を示し た。
デ ィ ス ポ 一 ザ ブル タ イ プの セ ン サ に おい て は、 各セ ン サ 毎の 電極面積などの差異をは じめ と する誤差要因をいかに小さ く す るかが信頼性向上に いて重要であ る。 これら セ ン サ間の差異 は同一試料液に対する測定値の差を生ずる原因と なっ てい る。 しかし ながら、 上記のよ う に、 同一セ ン サ に複数の電極系を設 け、 その応答値の平均値を得る こ と によ り、 よ り 高精度な測定 を迅速に行な う こ と がで き る。
第 1 2 図には 3組の電極を有する例について示した力 これ に限定される こ と はな く、 さ ら に多数の電極系を設置する こ と が可能である。 また空間部の形状については、 電極系の形状や 配置も同様であるが、 図示し た も の に限定される も のではない。
実施例 4
第 1 3 図に複数の電極系を有する構成の変形例を示す。 すで に述べた実施例 1 と 同様の方法で、 基板上に銀 リ ー ド 2 1、 2 2、 3 と これ らに対応する測定極 4 1、 4 2 と共通の対極 5 力 ら なる 2組の電極系を構成し た。 次に、 実施例 2 と同様に し て、 C M C— G O D — フ ヱ リ シ ア ンィヒカ リ ゥ ム層か ら なる反応層を 各々の電極系上へ形成した後、 スぺーサ 7 と 力パ ー 9 を接着し た。 こ こ で ス ぺ 一 サは独立し た空間部 8 1、 8 2 となる 2 ケ の 穴を有してお り、 これに対応する排出口 1 1、 1 2 と共通の導 入口 1 0 が力パーに設けてある。
上記構成のグル コ ー ス セ ン サ について、 実施例 3 に述べたと 伺様に、 グル コ ー ス標準液に対する 2組の電極系の応答値を平 均して 1 個のセ ン サ の測定値と し、 3 0個の同一仕様のセ ン サ について試験した と こ ろ、 変動係数は 2. 2 % と良好であ っ た。
こ の様に、 第 1 3—図に示すセ ン サにおいては、 よ り ¾精度な 測定ができ る ほか、 独立した空間部を有してい るので、 1 個の セ ン サで異なる試料液、 あるいは時間や温度等の異なる条件下 での測定に供する こ ともでき る。
上記においては、 ガ'パーに 1 個の穴を設けて共通の導入口と したが、 こ の構成に限定される こ と はない。 複数組の電極系と 複数の独立した空間部からなる測定系を搆成する こ とが重要な 点であ り、 2個の穴を設けて、 それぞれの空間部に対する導入 口を構成する こ と も でき る。
実施例 5
第 1 3 図に示す構成の電極部、 ス ぺーサ、 力パーを用いて以 下に述べるグル: 7—ス セ ンサを構成した。 まず、 測定極 4 1 の 上には実施例 1 と同様にして C M C— G 0 Dか らなる反応層を 形成した。 一方、 測定極 4 2の上には C M C層のみを形成した。
上記の様に して得られた 2組の電極系を有する グル コ ー ス セ ン サに つ いて、 種々 の濃度の ァ スコ ル ビ ン酸を含む グル コ ー ス 標準液 ( 2 0 0 m g Z d l ) を導入口 1 0 に滴下し、 各々の電 極系の上へ導いた。 次に、 実施例 1 と同様に、 滴下後、 約 1 分 後に 1 Vの電圧を印加し、 5秒後の電流値を測定し た。 結果を 第 1 4図に示す。 C M C — G 0 D層の電極系の出力を Aで、 ま た、 C M C層だけの電極系の出力 ( ブ ラ ン ク 出力) を B でそれ ぞれ示す。 図よ り 明 らかなよ う に、 A の出力は ァ ス コ ル ビ ン酸 の濃度増加と と も に増大し、 一方 B の出力も 同様な増加がみ ら れる。 これは ァ ス コ ル ビ ン酸に対する各々の電極系の感度がほ ぼ等し い こ と を示し てい る。 こ れよ り、 両電極系の出力の差 ( A— B ) を検出する と ダル ニ ー ス に基 く 電流値が得ら れる。 す なわち、 2組の電極系を用いる こ と によ り 電極活性な物質によ る誤差を大幅に低減する こ とができ る。 こ の様な効果はァ ス コ ル ビ ン酸以外にも、 尿酸などについても認め られた。
この様に、 2組の電極系を設け、 一方の電極系に親水性高分 子一酵素層、 他方の電極系に親水性高分子層だけを形成し て、 セ ン サを構成する こ と によ り、 妨害物質を含む試料液中の基質 濃度を精度よ く 測定する こ とができ た。
上記において、 両方の電極系に C M C — G ◦ D層を形成した 後、 一方の電極系につ い て のみレーザ照射によ る局部加熱、 紫 外線照射などを施すこ と によ り G 0 Dを失活させて、 ブラ ン ク 出力用の電極系とする こ と もで き る。 こ う する と酵素活性以外 は両電極系の構成が同一とな り、 両電極系の妨害物質によ る出 力電流をさ ら によ く 一致させる こ と ができ、 セ ン サ検出精度を 向上する こ と ができ る。
また、 以上の実施例においては電極部分が測定極と対極の 2 電極から な る電極系について述べた力 電極系を銀ノ塩化銀を 加えた 3 電極から構成する こ と によ り、 さ ら に精度を向上する こ とができ る。 電極系を構成する方法の一例と しては、 3本の 銀 リ ー ド を基扳上に印刷した後、 2 本の リ ー ド先端部の上にだ けカーボ ンペース ト を印刷し、 絶縁層をコ ー ト した後、 銀が露 出してい る残り 1本の リ ー ド先端部について、 その表面を処理 して塩化銀を形成し、 銀 Z塩化銀電極とするな どによ り構成す る こ とができ た。
実施例 6
実施例 &において、 測定極— 4 1 の上に実施洌 2 と同様:こ し て C M C— G 0 D—フ ヱ リ シ ア ンィヒカ リ ゥ ムから なる反応層を、 また測定極 4 2の上に C M C—フ ユ リ シ ア ン化力 リ ゥ ムか らな る層をそれぞれ形成した。 測定は、 印加 圧を 0. 6 Vと した 以外は実施例 5 と同様に行なつ たと ころ、 前記同様にァ ス コ ル ビ ン酸の影響を除去する こ とができた。
実施例 7
実施例 5 において、 ,測定極 4 1 の上に実施例 2 と同様にして C M C— G O D—フ : c リ シ ア ンィヒカ リ ゥ ムからなる反応層を形 成した。 また測定極 4 2 の上には、 C M C層を形成し た後、 グ ル コ ースォキ シ ダーゼと ム タ ロ タ ーゼおよ び ^ ー グル コ シ ダ一 ゼからなる酵素層を形成し、 さ ら に フ ェ リ シ ア ン化カ リ ウ ムか ら なる電子受容体層を形成して反応層と した。 測定は、 印加電 圧を 0. 6 Vと し、 試料液と してグルコ ース と シ ョ糖 (ス ク ロ ー ス) を含む水溶液を用いたと ころ、 測定極 4 1 においてはグ ル コ ー ス濃度に対応した電流が得られ、 また、 測定極 4 2 にお いてはグルコ ース と ス ク ロ ースの合計濃度に対応した電流が得 ら れた。 こ れ ら の電流値の差か ら ス ク ロ ー ス濃度を測定で き た c こ の よ う に し て、 2 種類の基質濃度を測定する こ と がで き る。 実施例 8
実施例 1 と 同様に し て電極部分を作成し た後、 親水性高分子 と し て、 カ ル ボキ シ メ チ ルセ ル ロ ー ス (以下 C M C と 略す) の 0 . 5 水溶液を電極系上へ展開、 乾燥し C M C層を形成す る。 次に、 こ の C M C層を覆 う よ う に、 グル コ ース ォ キ シ ダー ゼ水に溶解し たも のを展開し、 乾燥 させ、 C M C — G 0 D層を 形成し た。 こ の場合、 C M C と G 0 D は—部分的に混会 さ れ た ·ί犬 態で厚さ 数 ミ ク ロ ン の薄膜状と な っ てい る。 さ ら に、 こ の C Μ C一 G O D層か ら な る第 1 の層上を完全に覆う よ う に し て、 ポ リ ビュ ル ピ ロ リ ド ン (以下 P V P と 略す) の 0 . 5 % ェ タ ノ 一 ル溶液を展開 し、 乾燥さ せ、 P V P層か ら な る第 2 の層を形成 し た。 こ の P V P層の上へ、 界面活性剤であ る レ シ チ ン の 1 % ト ルエ ン溶液中に電子受容体であ る フ リ シ ア ンィヒカ リ ゥ ム の 微結晶を混ぜたも の を滴下、 展開 し、 乾燥 さ せ る こ と に よ っ て フ エ リ シ ア ンィ匕カ リ ゥ ム 一 レ シ チ ン層か ら な る第 3 の層を形成 し た。 そ し て、 実施例' 1 と 同様に ス ぺーサ、 カバー と と も に一 体化し た。
こ の フ リ シ ア ンィ匕 カ リ ウ ム一レ シ チ ン層を形成す る 際の溶 媒と し て P V Pが難溶性を示す ト ル エ ン を用い る こ と に よ っ て、 フ エ リ シ ア ンィヒカ リ ゥ ム 一 レ シ チ ン 溶液を P V P層上に一様に 広げる こ と が可能と な り、 そ の結果、 均一な フ ユ リ シ ア ンィ匕カ リ ゥ ム ー レ シ チ ン層を得る こ と がで き た。
こ の よ う に電子受容体と界面活性剤を展開す る際の溶媒 と し て、 第 2層を搆成する親水性高分子が難溶性を示す溶媒を用い る と、 極めて均一性の高い電子受容体層 (第 3 の層) を形成す る こ とができ た。
上記のよ う にして構成したグルコ ー ス セ ン サ に試料液と して グル コ ー ス標準液を導入口から加え、 1 分後に測定極にァ ノ ー ド方向へ + 0. 6 Vのパル ス電圧を印加し、 5秒後の応答電流 を測定し たと ころ、 9 0 0 ni g/d l以上と い う高濃度まで良好な直 線関係が得られた。 上記のグル コ ー スセ ン サに血液サ ンプルを 5 H 1 _滴下し て 1分後の 答電流を澍定し た と こ ろ |Ξ常 S現 性のよい応答が得られた。
上記の方法で作成したグル コ ー ス セ ン サ と、 該グル コ ー ス セ ン サの う ち親水性高分子から なる第 2 の層を設けなか っ たも の の 2種類について、 乾燥状態で 3 5 °C、 3 0 日間の保存試験を 行なった。 試料液と してグル コ ー ス標準液 ( 9 0 mg/d l ) を用い て 3 0 日後のセ ン サ応答—を比較した と ころ、 親水性高分子から なる第 2 の層を設けなかったも のについて の C V値は 5 . 3 と な つたが、 P V Pから なる第 2の層を設けたも のの C V値は 2. 5 と極めて良好な値が得られた。
実施例 9
実施例 8 と同様に して C M C — G 0 D層、 および P V P層を 形成した。 この P V P層の上へ、 界面活性剤である レ シ チ ン の 0 . 5 % エ タ ノ ール溶液中に電子受容体と して フ ェ リ シ ア ンィ匕 カ リ ウ ム の微結晶を混ぜたも のを滴下、 展開し、 乾燥させる こ とによってフ エ リ シ ア ン化カ リ ゥ ム 一 レ シ チ ン層を形成した。 こ の フ ヱ リ シ ア ンィ匕カ リ ゥ ム ー レ シ チ ン層を形成する際の溶媒 と し て P V P が容易 溶解する エ タ ノ ー ルを用 い る こ と に よ つ て、 フ ユ リ シ ア ン ィヒ カ リ ゥ ム ー レ シ チ ン層を P V P層上 の 一点 に集中的に展開する こ と がで き た。 すなわ ち、 セ ン サ の測定電 極上に集中 し て均—な フ リ シ ア ン ィヒカ リ ゥ ム ー レ シ チ ン 層を 構成する こ と が可能 と な り、 必要と される 最小量を展開す る だ けで安定な応答が得 ら れ る セ ン サを作成す る こ と がで き た。
上記の よ う に し て構成し た グル コ ー ス セ ン サ の グル コ ー ス標 準液に対する応答特性を実施例 8 と 同様に し て測定し た と こ ろ、 9 0 0 m g / d i以上 と い う 高濃度 まで良好な直線関 ¾が得 ら た。 ま た、 血液を試料液 と し て用い た場合に も 非常に再現性の よ い 応答が得 られた。 さ ら に、 実施例 8 と 同様に し て親水性高分子 か ら な る第 2 の層を設け たも の と設けなか っ た も のに つ い て、 乾燥状態で 3 5 °C、 3 0 日間の保存試験を行な っ た と こ ろ、 3 0 日後の応答は P V Pか ら な る第 2 の層を設け た も の は極めて 良好な C V値が得 ら れた。
従来の構成では、 セ ン サが作成さ れた時点ですでに G 0 D— C M C層 と フ ユ リ シ ア ン ィ匕カ リ ウ ム 一 レ シ チ ン層 とが接触し て いたため、 保存性能を高め る こ と が難し か っ た。 前記実施例 8 および本実施例 9 で用い た P V P か ら な る親水性高分子層は、 乾燥状態において G 0 D — C M C層 と フ ェ リ シ ア ンィ匕 カ リ ウ ム 一 レ シ チ ン層 と を完全'に分離する役 目 を果た し てい る。
さ ら に、 こ の親水性高分子層は試料液中 に電極に吸着し やす い物質や電極活性な物質が存在する場合に おいて も、 安定なセ ン サ応答を確保する のに極めて有効であ る。 血液を試料液 と し て前記グル コ ー ス セ ン サ で グル コ ー ス濃度を測定 し た場合に も、 試料液の粘度等に拘 らず安定したセ ン サ応答が得られた。
以上に述べた実施例 1〜 9 においては、 空間部につながる導 入口と排出口について-は、 その好適な位置関係の一例を実施例 と して図示したも のであ り、 排出口を導入口 と して、 また導 入口を排出口 と して用いても よい。 さ ら に、 各々の図に示した スぺーサおよび力パーは独立した部材から な つているが、 これ に限定される こ とはない。 成形などによ り スぺーサの形状を力 バ一に持たせた力パ一だけの一体部材とすれば量産上有利であ る。
また上記、 空間部を構成する部材表面を親水性とする こ と に よ り、 導入口から の試料液の導入がよ り 円滑と なった。 こ の効 果は親水性の材料を用いるか、 あるいは、 空間部を構成する部 材の表面を予め界面活性剤で処理し親水性と し たとき に得られ た。
—方、 基板と力パー の間隔は、 スぺ ーサの厚みあるいは これ に相当する部分の寸法を変える こ と によっ て調節する こ とがで きるが、 大きすぎる と空間部を満たすに要する試料液が多 く な り、 また毛管現象による液導入にも支障をきたす。 また、 間隔 が小さすぎる と、 電極間の抵抗の増大や電流分布の乱れを生ず る。 以上の点から、 間隔と しては 0. 0 5〜 1 . 0 (f¾が適当で あ り、 さ ら に好ま し く は 0. 1〜 0. 5 (ί であ る。
実施例においては、 親水性高分子と して C M Cを用いたが、 その役割は、 すでに述ベたよ う に試料液の円滑な導入の他に、 試料液に含まれる吸着性物質が電極へ吸着し応答に影響するの を防止する こ と にあ る。 さ ら には、 酵素層と電子受容体層を親 水性高分子層で分離する こ と に よ り 保存性を さ ら に向上す る こ と にあ る。 親水性高分子 と し ては C M C の他に ゼ ラ チ ン ゃ メ チ ノレ セ ル ロ ー ス な ど も 使用で き、 デ ン プ ン 系、 カ ル ボ キ シ メ 午 ノレ セ ノレ ロ ー ス系、 ゼ ラ チ ン 系、 ァ ク リ ノレ酸塩系、 ビ ニ ル ァ ノレ コ ー ル系、 ビ ニ ル ピ ロ リ ド ン 系、 無水マ レ イ ン 酸系の も のが好ま し い。 こ れ ら の吸水性あ る いは水溶性の親水性高分子を適当 な濃 度の溶液に し たも の を塗布、 乾燥す る こ と に よ り、 必要な膜厚 の親水性高分子層を形成する こ と ができ る。
なお、 以上の実施例に.おいては電極部分が測定極と 対極 2 電極か ら な る電極系について述べた力 電極系を参照極を加え た 3電極か ら 構成する こ と に よ り、 さ ら に精度を向上す る こ と がでる。 ま た、 電極材料と し ては、 実施例に示し た カ ー ボ ン は 安定で安価な材料と し て適し てい る 力、 こ れに制限さ れる こ と は な く、 他の貴金属材料や酸化物材料を用いて も よい。
ま た、 電子受容体と し ては、 実施例に示 し た フ ユ リ シ ア ン 化 カ リ ウ ム は安定性や反応速度の点で優れた も のであ る が、 こ れ 以外に、 例えばキ ノ ン系化合物や フ ユ 口 セ ン系化合物な どの レ ド ッ ク ス化合物を用い る こ と も で き る。
さ ら に、 酸化還元酵素 と し ては上記実施例に示し た グ ル コ 一 ス ォキ シ ダー ゼに限定さ れる こ と は な く、 ァノレ コ ール ォ キ シ ダ — ゼゃ コ レ ス テ ロ ー ルォ キ シ ダ ー ゼな ど種々の酵素を用い る こ と がで き る。
