Verfahren zur Herstellung von cu-Dimethylaminoalkylarylketonen ci-Dimetliylaminoallcylaryllcetone
der Zusammensetzung:
worin i und a Alkyl oder Wasserstoff und K Aryl bedeuten, sind Zwischenprodukte
für die Darstellung wertvoller Heilmittel. Zu co-Diinetlivlaminoalkylarylketonen
gelangt man nach dem Schrifttum auf folgendem Wege: Man läßt auf co-Halogenalkylarylketone
Dimethylamin einwirken, wobei man neben höheren, basischen Ketonen und quaternären
Ammoniumsalzen in zahlenmäßig nicht angegebenen Ausbeuten geringe Mengen der gesuchten
Aminoketone erhält. Im Archiv der Pharmazie Bd..237 (i899) S. 234/35 wird die Darstellung
von co-Dimethylarninoacetophenon auf diesem Wege ohne Nennung der Ausbeute beschrieben.
Wie durch Nacharbeiten der dort gegebenen Vorschrift festgestellt wurde, beträgt
die Ausbeute hierbei nur 20 °/". Es wurde nun gefunden, daß man in durchweg guten
Ausbeuten von etwa 6o bis 75 % zu co-Dimethylaminoalkylarylketonen gelangt, wenn
man auf a-Dimethylaminocarbonsäuredialkylamide Aryimagnesiumhalogenide einwirken
läßt.Process for the preparation of cu-Dimethylaminoalkylarylketonen ci-Dimetliylaminoallcylaryllcetone of the composition: where i and a are alkyl or hydrogen and K are aryl are intermediates for the preparation of valuable medicinal products. According to the literature, co-diinetlivlaminoalkylaryl ketones are obtained in the following way: Dimethylamine is allowed to act on co-haloalkylaryl ketones, and in addition to higher, basic ketones and quaternary ammonium salts, small amounts of the desired amino ketones are obtained in yields not given in numbers. In the Archives of Pharmacy Vol. 237 (1899) p. 234/35 the representation of co-dimethylarninoacetophenone is described in this way without mentioning the yield. As was established by reworking the instructions given there, the yield here is only 20%. It has now been found that co-dimethylaminoalkylaryl ketones are obtained in consistently good yields of about 60 to 75% if a-dimethylaminocarboxylic acid dialkylamides are aryimagnesium halides can act.
Es. ist bekannt (vgl. z. B. Chemisches Zentralblatt, 1926, Bd. 1I,
S. 75q., und i9,30 Bd. 1, S.2721), daß durch Einwirkung von Arylmagnesiumhalogeniden
auf N-dialkylierte Säureamide Ketone dargestellt werden können. Es war jedoch nicht
zu erwarten, daß durch Einwirkung von Arylmagnesiumhalogeniden auf a-Dimethylraminocarbonsäuredialkylamide
in guten Ausbeuten die entsprechenden ci-Dimethylaminoalltylarylketone erhalten
werden. Ein solches Verfahren zur Herstellung von orDimethylaminoalkylarylketonen
durch Grignardierung der entsprechenden a-Dimethylaminocarbonsäureamide mit Arylmagnesiumlialogeniden
ist bis jetzt aus dem .Schrifttum noch nicht bekanntgeworden. Ausführungsbeispiele
i. Zu einer Grign@rdlösung aus 72,5 g Brombenzol, 11,2 g Magnesium und Zoo
ccm absolutem Äther fügt man tropfenweise eine Lösung von 2o g Dimethylaminoessigsäuredimethylamid
in 5o ccm absolutem Äther. Durch zweistündiges Erhitzen auf dem Wasserbad
wird
die Umsetzung zu Ende gebracht und das Umsetzungsgut mit Eis und Salzsäure zersetzt.
Die mit Ammoniak gefällte Base wird in Äther aufgenommen, . mit , Kaliumcarbonat
getrocknet und im luftver.=.-dünnten Raum destilliert. Man erhält in:tva' 8o°/oiger
Ausbeute reines co-Dimethylaminö= acetophenon.It. It is known (see, for example, Chemisches Zentralblatt, 1926, Vol. 11, p. 75q., and 19, 30, Vol. 1, p. 2721) that ketones can be prepared by the action of aryl magnesium halides on N-dialkylated acid amides . However, it was not to be expected that the action of aryl magnesium halides on α-dimethylraminocarboxylic acid dialkylamides would give the corresponding ci-dimethylaminoalltylaryl ketones in good yields. Such a process for the preparation of dimethylaminoalkylaryl ketones by Grignardation of the corresponding α-dimethylaminocarboxamides with arylmagnesium halides has not yet become known from the literature. Embodiments i. A Grign @ rdlösung of 72.5 g of bromobenzene, 1 1.2 g of magnesium and Zoo cc absolute ether added dropwise a solution of 2o g Dimethylaminoessigsäuredimethylamid in 5o cc absolute ether. The reaction is brought to an end by heating on a water bath for two hours and the reaction material is decomposed with ice and hydrochloric acid. The base precipitated with ammonia is taken up in ether,. dried with potassium carbonate and distilled in an air-diluted = .- thin space. Pure co-dimethylamino = acetophenone is obtained in a yield of about 80%.
2. co-Dimethylaminoacetophenon aus a-Dimethylaminodiäthylacetamid.
Man bereitet aus i 5o g Brombenzol, 23 g Magnesium und 4.0o ctn' absolutem Äther
eine Grignandlösung und fügt 6o g Dimethylarninodiäthylacetamid in Zoo cm3 Äther
hinzu. Nach einstündigem Erhitzen auf dem Wasserb d wird das Umsetzungsgut mit Eis
und Salzsäure zersetzt, die Base mit Ammoniak abgeschieden, mit Äther aufgenommen
und nach Entfernen des Äthers im Vakuum destilliert. Ausbeute: 4.6 g co-Dimethylaminoacetophenon
(74. °/o der Theorie).2. co-dimethylaminoacetophenone from α-dimethylaminodiäthylacetamid.
One prepares from 15o g of bromobenzene, 23 g of magnesium and 4.0o ctn of absolute ether
a Grignand solution and add 60 g of dimethylaminodiethylacetamide to zoo cm3 of ether
added. After heating on the water bath for one hour, the reaction mixture is mixed with ice
and hydrochloric acid decomposed, the base deposited with ammonia, taken up with ether
and, after removing the ether, distilled in vacuo. Yield: 4.6 g of co-dimethylaminoacetophenone
(74 ° / o of theory).
3. co-Dimethylaminomethyl-i, 3-dimethylphenylketon. 4.o g Dimethylaminodimethylacetamid,
in 150 cm' absolutem Äther gelöst, werden zu einer aus i50 g i, 3-Dimethylq.-brombenzol,
2o g Magnesium und 400 cm3 absolutem Äther dargestellten Grignardlösung gegeben.
Die Umsetzung wird durch zweibis d.-reistündiges Erhitzen auf dem Wasserbad zu Ende
geführt. Das co-Dimetliylaminor, 3-dimethylphenylketon wird aus dem mit Eis und
Salzsäure zersetzten Umsetzungsgemisch mit Hilfe von Ammoniak abgeschieden, mit
Äther aufgenommen, mit Kaliumcarbonat getrocknet und im Vakuum destilliert. Kpil
_,3 = 1a-4 bis i:46°. Die Ausbeute beträgt 6o bis 65 °/o der Theorie.3. co-dimethylaminomethyl-1,3-dimethylphenyl ketone. 4.og dimethylaminodimethylacetamide, dissolved in 150 cm 'of absolute ether, are added to a Grignard solution prepared from 150 g of 3-dimethylq-bromobenzene, 20 g of magnesium and 400 cm3 of absolute ether. The reaction is brought to completion by heating on the water bath for two to three hours. The co-dimethylaminor, 3-dimethylphenyl ketone is separated out from the reaction mixture, which has been decomposed with ice and hydrochloric acid, with the aid of ammonia, taken up with ether, dried with potassium carbonate and distilled in vacuo. Kpil _, 3 = 1a-4 to i: 46 °. The yield is 60 to 65% of theory.
q.. a-Dimethylaminopropiophenon. Eine Grignardlösung aus 8o g Brombenzol,.
12,4 g Magnesium und Zoo cm3 absolutem Äther wird mit einer Lösung von
30 g a-Dimethylaminodimethylpropionamid in ioo cm' absolutem Äther umgesetzt.
Das nach zweistündigem Erhitzen auf dem Wasserbad erhaltene Umsetzungsgut liefert
nach dem Aufarbeiten mit Eis und Salzsäure und nach dem Fällen mit Ammoniak 26 g
a-Diriiethylaminopropiophenon (70 °/o der Theorie).q .. a-Dimethylaminopropiophenone. A Grignard solution of 80 g of bromobenzene. 12.4 g of magnesium and zoo cm3 of absolute ether are reacted with a solution of 30 g of a-dimethylaminodimethylpropionamide in 100 cm of absolute ether. The reaction mixture obtained after heating for two hours on a water bath gives, after working up with ice and hydrochloric acid and after precipitation with ammonia, 26 g of α-diriethylaminopropiophenone (70% of theory).
