Description
- 1. Gebiet der Erfindung
- Diese Erfindung bezieht sich auf Verfahren und Zusammensetzungen, die Hautpigmentierung zur Folge haben und/oder die Depigmentierung der Haut verursachen. Diese Erfindung bezieht sich weiter insbesondere auf Verbindungen, welche Melanogenese beeinflussen und als Depigmentierungs-Mittel oder als Mittel zum Verdunkeln der Haut verwendet werden können.
- 2. Hintergrund der Erfindung
- Die Färbung der Haut ist seit vielen Jahren ein Anliegen der Menschen. Insbesondere ist die Fähigkeit, Hyperpigmentierung zu entfernen, wie sie in Altersflecken, Sommersprossen oder in alternder Haut im Allgemeinen gefunden wird, von Interesse für Individuen, die eine uniforme Hautfarbe/Teint wünschen. In bestimmten Gebieten der Welt ist allgemeines Körperbleichen wünschenswert. Es gibt auch Hypopigmentierungs- und Hyperpigmentierungs-Störungen, deren Behandlung wünschenswert ist. Die Fähigkeit, eine gebräunte Erscheinung hervorzurufen, ohne daß Fotoschaden („photodamage") aufgrund von Sonnenstrahlung erlitten wird, ist für viele Individuen gleichermaßen bedeutsam. Es wurden viele Methoden vorgeschlagen, um Depigmentierung zu erreichen sowie um Verdunkelung der Haut zu erreichen. Zum Beispiel wurden Koji-Säure, Hydrochinon, Retinoide und andere chemische Verbindungen für Depigmentierung verwendet. Dihydroxyaceton und ähnliche chemische Verbindungen wurden aufgrund ihrer Fähigkeit verwendet, die Haut zu „bräunen", ohne daß sie der Sonne ausgesetzt wird.
- Viele dieser vorherigen Lösungen wurden als nicht akzeptabel befunden. Es gibt oftmals eine ausgeprägte Grenzlinie zwischen den Gebieten der Haut, auf welche die vorherigen Zusammensetzungen aufgetragen wurden. Daher ist die präzise Applikation aller dieser Verbindungen notwendig, um das gewünschte Ergebnis zu erreichen. Von vielen dieser Verbindungen wurde gefunden, daß sie die Haut ziemlich reizen und daher unerwünscht für die Verwendung sind.
- Das Verständnis der chemischen und enzymatischen Grundlage der Melanogenese wurde in großem Umfang dokumentiert. Melanozyten migrieren aus der embryonalen Neuralleiste in die Haut, um sekretorische Granula, Melanosomen, welche Melanin produzieren, zu produzieren. Melanogenese tritt innerhalb des Melanosoms auf und das Melanin wird später zu den Keratinozyten über die Melanozyten-Dendriten verteilt. Das Schlüsselenzym in der Melanogenese ist Tyrosinase, welche eine Kaskade von Reaktionen initiiert, welche Tyrosin in das Biopolymer Melanin umwandelt. Zwei Tyrosinase-verwandte Proteine (TRP's) sind bekannt, TRP-1 und TRP-2. Diese Proteine teilen mit Tyrosinase ungefähr 40% Homologie und haben katalytische Aktivitäten sowie regulatorische Rollen in der Melanogenese. TRP-1 ist das am reichlichsten vorkommende Glykoprotein in Melanozyten.
- Trotz der Tatsache, daß die chemische und enzymatische Grundlage der Melanogenese gut dokumentiert ist, wird ihre Regulation auf dem zellulären Niveau nur teilweise verstanden. Tyrosinase und die TRP's teilen strukturelle und biologische Eigenschaften mit der Genfamilie der lysosomal-assoziierten Membranproteine (LAMP), daher könnte ihr Targeting zur melanosomalen Membran ihre Aktivierung induzieren. Eine Phosphorylierungs-/Dephosphorylierungs-Reaktion an den zytoplasmatischen Schwänzen dieser Proteine könnte in die Regulation der Melanogenese involviert sein. Von der β-Isoform der Proteinkinase C (PKC)-Famile wurde gezeigt, daß sie die humane Melanogenese durch Tyrosinase-Aktivierung reguliert. Die Genexpression von Tyrosinase, TRP-1 und TRP-2 ist koordiniert. Alle drei Enzyme werden in der humanen Epidermis exprimiert. In Melanozyten, die mit Keratinozyten ko-kultiviert wurden, werden diese Transkripte in einem Verhältnis von 45:45:10 exprimiert. In Melanozyten, die allein kultiviert werden, sind nur TRP-1-Transkripte anwesend, was anzeigt, daß ein Keratinozyten-abgeleitetes Signal in die koordinierte Expression dieser Gene involviert ist. Die Regulation der Interaktionen zwischen Keratinozyten und Melanozyten und der Mechanismus des Melanosom-Transfers in die Keratinozyten werden bisher nicht verstanden.
- Der Protease-aktivierte Rezeptor-2 (PAR-2) ist ein Sieben-Transmembran-, G-Proteingekoppelter Rezeptor, der mit den Thrombin-Rezeptoren (TR, auch bezeichnet als PAR-1 und PAR-3) verwandt ist, aber in seiner Sequenz verschieden ist. Beide Rezeptoren werden proteolytisch durch eine Arginin-Serin-Spaltung in der extrazellulären Domäne aktiviert. Die neugeschaffenen N-Termini aktivieren dann diese Rezeptoren als angebundene Liganden.
- Beide Rezeptoren können durch Trypsin aktiviert werden, aber nur die TRs werden durch Thrombin aktiviert. Nur PAR-2 wird durch Mastzell-Tryptase aktiviert. Unabhängig von der Spaltung des Rezeptors können beide Rezeptoren auch durch die Peptide aktiviert werden, die mit ihren neuen N-Termini korrespondieren. SLIGRL, das Maus-PAR-2-aktivierende Peptid, ist gleichermaßen potent („equipotent") in der Aktivierung des humanen Rezeptors. Während die Funktion der TR gut dokumentiert ist, wurde die Biologie des PAR-2 bisher nicht vollständig identifiziert. Eine Rolle der PAR-2-Aktivierung in der Inhibierung des Keratinozyten-Wachstums und der Keratinozyten-Differenzierung wurde kürzlich beschrieben (Derian et al., „Differential Regulation of Human Keratinocyte Growth and Differentiation by a Novel Family of Protease-activate Receptors", Cell Growth & Differentiation, Vol. 8, S. 743–749, Juli 1997).
- Zusammenfassung der Erfindung
- Gemäß dieser Erfindung haben wir ein Verfahren zur Reduzierung der Hautpigmentierung eines Säugetiers gefunden, welche die topische Applikation eines Bowman-Birk-Inhibitors auf die Haut eines Säugetiers in Abwesenheit des Sojabohnen-Trypsin-Inhibitors umfaßt.

Die graphische Darstellung quantifiziert die Prozentzahl der Inhibierung der Pigmentablagerung in der Schweinehaut nachfolgend BBI- oder STI-Behandlung. - Detaillierte Beschreibung der bevorzugten Ausführungsformen
- Während wir nicht durch die folgende Theorie gebunden werden wollen, stellen wir die Theorie auf, daß die Verbindungen deshalb zur Beeinflussung der Pigmentierung der Haut fähig sind, weil sie direkt oder indirekt mit dem Keratinozyten-PAR-2 oder mit seiner aktivierenden Protease interagieren und dabei Melanogenese direkt oder indirekt beeinflussen. Möglicherweise induzieren die Verbindungen dieser Erfindung im Fall der verstärkten Pigmentierung – oder reduzieren im Fall der verminderten Pigmentierung – das Signal, Melanosomen mittels Melanozyten zu transportieren oder Melanosomen durch Keratinozyten in der Haut zu empfangen.
- Kürzlich haben wir identifiziert, daß der Bowman-Birk-Inhibitor (BBI), eine Gruppe von Hülsenfrucht-abgeleiteten Proteinen, depigmentierende Mittel sind.
- Während STI ein 21 kD Protein mit primärer Trypsin-inhibitorischer Aktivität ist, ist der Sojabohnen-abgeleitete BBI ein kleineres 8 kD Protein, welches Chymotrypsin und Trypsin in hibiert. Ungleich STI hat BBI keine Kunitz-Typ-Domäne, was verschiedene Interaktionen mit Serinproteasen vorschlägt. BBI ist für seine Fähigkeit bekannt, Karzinogenese in zahlreichen in vivo und in vitro Modellen zu verhindern. In einigen tierischen Karzinogenese-Modellen wurde von BBI gefunden, daß es starke anti-inflammatorische Effekte aufweist. BBI ist gegenüber Hitze-Denaturierung resistenter als STI. - Die Verbindungen, die in den Zusammensetzungen und Methoden dieser Erfindung aktiv sind, können topisch durch jedes Mittel, das dem Fachmann auf diesem Gebiet bekannt ist, zugeführt werden. Wenn es die Zufuhrparameter der topisch aktiven pharmazeutischen oder kosmetischen Mittel erfordern, kann die topisch aktive Zusammensetzung dieser Erfindung bevorzugterweise weiterhin zusammengesetzt sein aus einem pharmazeutisch oder kosmetisch akzeptablen Vehikel, das in der Lage ist, als ein Zufuhrsystem zu fungieren, um die Penetration des topisch aktiven Mittels in die Haut zu ermöglichen.
- Ein akzeptables Vehikel für die topische Zufuhr für einige der Zusammensetzungen dieser Erfindung kann Liposomen enthalten. Die Liposomen sind weiter bevorzugt nicht-ionisch und enthalten a) Glycerol-Dilaureat (bevorzugt in einer Menge zwischen ungefähr 5 Gewichts-% und ungefähr 70 Gewichts-%); b) Verbindungen, die ein Steroid-Rückgrat aufweisen, das in Cholesterol gefunden wird (bevorzugt in einer Menge von zwischen ungefähr 5 Gewichts-% und ungefähr 45 Gewichts-%) und c) ein oder mehrere Fettsäure-Ether, die von ungefähr 12 bis ungefähr 18 Kohlenstoffatome aufweisen (bevorzugterweise in einer Menge von gemeinsam ungefähr 5 Gewichts-% und ungefähr 70 Gewichts-%), wobei die konstituierenden Verbindungen der Liposomen bevurzugterweise in einem Verhältnis von ungefähr 37,5:12,5:33,3:16,7 vorliegen. Liposomen umfassend Glycerol-Dilaureat/Cholesterol/Polyoxyethylen-l0-Stearylether/Polyoxyethylen-9-Laurylether (GDL-Liposomen) werden am meisten bevorzugt. Bevorzugterweise sind die Liposomen in einer Menge vorhanden, basierend auf dem Gesamtvolumen der Zusammensetzung, von ungefähr 10 mg/ml bis ungefähr 100 mg/ml und weiter bevorzugt von ungefähr 20 mg/ml bis ungefähr 50 mg/ml. Ein Verhältnis von ungefähr 37,5:12,5:33,3:16,7 ist am meisten bevorzugt. Geeignete Liposomen können bevorzugterweise präpariert werden in Übereinstimmung mit dem Protokoll, das in Beispiel 1 dargelegt wird, obwohl andere Methoden, die im Stand der Technik gewöhnlicherweise verwendet werden, auch akzeptabel sind.
- Die oben beschriebene Zusammensetzung kann präpariert werden durch Kombinieren der gewünschten Komponenten in einem geeigneten Behälter und Mischen der Komponenten unter Umgebungsbedingungen in irgendeinem konventionellen Mischungsmittel mit hoher Scherung, die im Stand der Technik zur Präparation nicht-ionischer Liposomen bekannt sind, wie z.B. diejenigen, die in Niemiec et al., „Influence of Nonionic Liposomal Composition On Topical Delivery of Peptide Drugs Into Pilosebacious Units: An In Vivo Study Using the Hamster Ear Model," 12 Pharm. Res. 1184–88 (1995) („Niemiec") offenbart werden, welche hiermit in ihrer Gesamtheit durch Referenz eingefügt wird. Wir haben gefunden, daß die Anwesenheit dieser Liposomen in den Zusammensetzungen dieser Erfindung die Depigmentierungs-Fähigkeiten einiger dieser Zusammensetzungen dieser Erfindung verstärken kann.
- Andere bevorzugte Formulierungen können z.B. flüssige Formulierungen enthalten, die direkt von Hülsenfrüchten oder anderen geeigneten Pflanzen abgeleitet werden.
- Öl-in-Wasser-Emulsionen; Wasser-in-Öl-Emulsionen, Lösungsmittel-basierende Formulierungen und wäßrige Gele, die dem Fachmann auf dem Gebiet bekannt sind, können auch als Vehikel für die Zufuhr der Zusammensetzungen dieser Erfindung verwendet werden.
- Produkte von natürlichen Extrakten können gemäß der Techniken, die im Stand der Technik bekannt sind, gereinigt werden. Rekombinante Quellen der Verbindungen sind dem Durchschnittsfachmann auf dem Gebiet auch verfügbar.
- In alternativen Ausführungsformen kann die topisch aktive kosmetische Zusammensetzung optional kombiniert werden mit anderen Inhaltsstoffen, wie z.B. Feuchtigkeitscremes, kosmetischen Hilfsmitteln, Anti-Oxidantien, bleichenden Mitteln, Tyrosinase-Inhibitoren und anderen bekannten depigmentierenden Mitteln, oberflächenaktiven Mitteln, Treibmitteln, Conditioners, Befeuchtungsmitteln, Düften, viskosierenden Mitteln, puffernden Mitteln, Konservierungsmitteln, Sonnenschutzmitteln und ähnlichem. Die Zusammensetzungen dieser Erfindung können auch aktive Mengen an Retinoiden (d.h. Verbindungen, die an irgendein Mitglied der Familie der Retinoid-Rezeptoren binden), einschließlich z.B. Tretinoin, Retinol, Ester von Tretinoin und/oder Retinol und ähnliches, enthalten.
- Die topisch aktive kosmetische Zusammensetzung sollte in einer Menge appliziert werden, die effektiv ist, um Änderungen in der Pigmentierung der Säugetierhaut zu beeinflussen. Wie hierin verwendet, bedeutet „effektive Menge" eine Menge, die ausreichend ist, um die Region der Hautoberfläche zu bedecken, wo eine Veränderung in der Pigmentierung gewünscht wird. Bevorzugterweise wird die Zusammensetzung großzügig auf die Hautoberfläche appliziert, so daß, basierend auf 1 cm2 Hautoberfläche, von ungefähr 2 μl/cm2 bis ungefähr 200 μl/cm2 des topisch aktiven Mittels vorhanden ist, wenn eine Veränderung in der Pigmentierung gewünscht wird.
- Flüssige Derivate und natürliche Extrakte, die direkt aus Pflanzen oder botanischen Quellen hergestellt werden, können in den Zusammensetzungen dieser Erfindung in einer Konzentration (w/v) von ungefähr 1 bis ungefähr 99% verwendet werden. Fraktionen von natürlichen Extrakten und natürlich-abgeleiteten Protease-Inhibitoren können einen unterschiedlichen bevorzugten Bereich von ungefähr 0,01% bis ungefähr 20% und weiter bevorzugt von ungefähr 1% bis ungefähr 10% der Zusammensetzung aufweisen.
- Wir haben überraschenderweise gefunden, daß, wenn ein BBI topisch auf die Haut eines Tieres appliziert wird, eine signifikante Veränderung in der Pigmentierung erreicht wird. Bevorzugterweise werden depigmentierende Mittel (sowie andere die Pigmentierung beeinflussende Mittel dieser Erfindung) auf die Haut eines Säugetieres in einer relativ hohen Konzentration und Dosis (von ungefähr 20% bis ungefähr 99% für flüssige Derivate und Extrakte aus botanischen Materialien und von ungefähr 1% bis ungefähr 20% für Fraktionen natürlicher Extrakte und natürlich-abgeleiteter Protease-Inhibitoren) zwischen ein- und zweimal täglich für eine Zeitspanne appliziert, bis die Haut eine Veränderung in der Pigmentierung zeigt. Dies kann für von ungefähr 4 bis ungefähr 10 Wochen oder mehr sein. Danach kann, wenn die Veränderung in der Pigmentierung erreicht wurde, eine niedrigere Konzentration und Dosis (von ungefähr 10% bis ungefähr 90% für flüssige Derivate und Extrakte aus botanischen Materialien und von ungefähr 0,01% bis ungefähr 5% für Fraktionen natürlicher Extrakte und natürlich-abgeleiteter Protease-Inhibitoren) des aktiven Inhaltsstoffes in einem weniger häufigen Zeitplan appliziert werden, z.B. einmal pro Tag bis ungefähr zweimal pro Woche. Die Effekte der aktiven Mittel dieser Erfindung sind reversibel, daher sollte, um diese Effekte aufrecht zu erhalten, kontinuierliche Applikation oder Verabreichung durchgeführt werden. Die Erfindung, die hierin illustrativ offenbart wird, kann geeigneterweise bei Abwesenheit irgendeiner Komponente, eines Inhaltsstoffes oder Schrittes, welcher nicht hierin spezifisch offenbart wird, praktiziert werden.

Mehrere Beispiele werden unten dargelegt, um die Natur der Erfindung und die Art und Weise ihrer Ausführung weiter zu illustrieren, dienen aber nicht dazu, den Umfang der Methoden und Zusammensetzungen dieser Erfindung zu limitieren.
- Beispiel 1: BBI beeinflußt die Pigmentierung
- Um die möglichen Rollen von BBI bei der Pigmentierung zu studieren, wurde ein in vitro epidermales äquivalentes System, enthaltend Melanozyten, verwendet. Das epidermale äquivalente System, das in dieser Studie verwendet wurde, ist das MelanoDerm me1-300-System, welches kommerziell von MatTek Co., Ashland, MA erhältlich ist. Dieses System enthält humane normale Melanozyten zusammen mit normalen human-abgeleiteten epidermalen Keratinozyten, die von afroamerikanischer Vorhaut abgeleitet werden. Diese Zellen wurden kultiviert, um ein Multi-Schicht-, hochdifferenziertes Modell der humanen Epidermis zu bilden. In den folgenden Beispielen wurden Äquivalente mit BBI (0,1%) für drei Tage behandelt und am vierten Tag nach dem Beginn der Behandlung wurden Proben geerntet. Die geernteten Äquivalente wurden zuerst auf ihre Farbe ohne Färbung verglichen, gefolgt von histologischer Untersuchung mit Fontana-Mason (F&M) Färbung, einer Färbung, die dem Fachmann auf diesem Gebiet bekannt ist. F&M-Färbung ist eine Silber-Färbetechnik, die Melanine deutlich und rein markiert, welche hohe Silbernitrat-reduzierende Aktivität aufweisen. Die Bilder der gefärbten Schnitte wurden auch für Bildanalyse erfaßt. Mindestens drei Schnitte pro Äquivalent, drei Äquivalente pro Experiment wurden bearbeitet. Für das Erfassen der Bilder wurde Empire Images Database 1.1 auf einem Gateway 2000 P5-100 Computer (Media Cybernetics, Silver Springs, MD) verwendet. Image Pro Plus Version 3.0 wurde für die Bildanalyse verwendet. Die gemessenen Parameter waren das Oberflächengebiet der Silberablagerungen innerhalb der Melanozyten und die Dichte-Helligkeit jedes Pixels. Ein „Pigmentierungs-Faktor" wurde definiert als das Oberflächengebiet von Silberablagerungen geteilt durch das gesamte epidermale Oberflächengebiet. Ein Wert von 1 (100%) wurde den unbehandelten Kontrollen zugewiesen und Werte von Behandlungsgruppen wurden gegenüber ihren relevanten Kontrollen normalisiert.
- Wie in
1 gezeigt wird, sind unbehandelte me1-300 Äquivalente sichtbar dunkel ohne irgendeine Färbung. BBI-behandelte Äquivalente waren heller als diese Kontrollen, was die Fähigkeit von BBI demonstriert, sichtbar Pigmentierung zu reduzieren.2 zeigt die histologischen Schnitte dieser Äquivalente nachfolgend F&M-Färbung. In dieser Figur repräsentieren schwarze Gebiete Melaninablagerungen innerhalb von sowohl Melanozyten als auch von Keratinozyten. Wie in2 gezeigt wird, führt BBI-Behandlung zu reduzierter Melaninablagerung sowohl in den Melanozyten als auch in den Keratinozyten von behandelten Äquivalenten. Bildanalyse legte offen, daß BBI-behandelte Äquivalente nur 50,6% Melaninablagerungen relativ zu den Kontrollen aufweisen. - Beispiel 2: Der depigmentierende Effekt von BBI ist Dosis-abhängig.
- Epidermale Äquivalente, enthaltend Melanozyten, wie in Beispiel 1 beschrieben, wurden mit steigenden Konzentrationen an BBI von 0,001% bis 0,1% behandelt. Es wurde derselben experimentellen Prozedur, die in Beispiel 1 beschrieben wird, gefolgt. Und es wurde gefunden, daß der depigmentierende Effekt von BBI Dosis-abhängig ist.
3 zeigt F&M-gefärbte Schnitte der behandelten Äquivalente, die die Dosisantwort und den depigmentierenden Effekt bei so wenig wie 0,001% BBI demonstrieren. Die computerisierte Bildanalyse, die in4 gezeigt wird, quantifiziert diesen Effekt und demonstriert weiterhin seine Dosisabhängige Natur. - Beispiel 3: In vivo Demonstration des depigmentierenden Effekts von BBI
- Ein dunkelhäutiges Yucatan-Mikroschwein wurde mit 1% BBI oder STI in PBS mit 20 mg/ml Liposomen behandelt. Nicht-ionische Liposomen-Präparationen, wie diejenigen, die in Niemiec et al., „Influence of Nonionic Liposomal Composition On Topical Delivery of Peptide Drugs Into Pilosebacious Units: An In Vivo Study Using the Hamster Ear Model," 12 Pharm. Res. 1184–88 (1995) ("Niemiec"), welche hierin in ihrer Gesamtheit durch Referenz eingefügt wird, offenbart werden, sind im Stand der Technik gut bekannt und werden in JPB-430 beschrieben. Wir haben gefunden, daß die Anwesenheit dieser Liposomen in den Zusammensetzungen dieser Erfindung die depigmentierenden Fähigkeiten einiger dieser Zusammensetzungen dieser Erfindung verstärken kann. GDL-Liposomen wurden präpariert, wie in Niemiec et al. oben, dargelegt wird, mit der Ausnahme der folgenden Änderungen: die nicht-ionische liposomale Formulierung enthielt Glycerol-Dilaureat (Emulsynt GDL, ISP Van Dyk)/Cholesterol (Croda)/Polyoxyethylen-10-Stearylether (Brij76, ICI)/Polyoxyethylene-9-Laurylether in einem Verhältnis von 37,5:12,5:33,3:16,7. Hepes-Puffer, 0,05M, pH 7,4 (Gibco-BRL, Gaithersburg, MD) wurde als die wäßrige Phase bei der Präparation der Liposomen verwendet.
- BBI, STI und Liposomen-Vehikel-Präparationen wurden jeweils auf zwei Seiten der Flanke des Schweins, zweimal täglich, fünf Tage pro Woche, für acht Wochen appliziert. Nach acht Wochen der Behandlung resultierte die Applikation von entweder BBI oder STI in einem sichtbaren aufhellenden Effekt. Die histologische Analyse von F&M-gefärbten Hautschnitten von unbehandelten und behandelten Stellen bestätigten diese Beobachtung.
5 zeit die F&M-gefärbten Hautschnitte des behandelten Schweins und demonstriert eine dramatische Reduktion in der Pigmentablagerung an den Stellen, die mit BBI oder STI behandelt wurden. Computerisierte Bildanalyse, die in6 gezeigt wird, quantifiziert diesen Effekt und demonstriert weiter den depigmentierenden Effekt von BBI.
Claims (5)
- Verwendung eines Bowman-Birk-Inhibitors in Abwesenheit von Sojabohnen-Trypsininhibitor für die Reduktion der Hautpigmentierung eines Säugetiers, unter der Voraussetzung, daß es keine therapeutische Verwendung ist.
- Verwendung gemäß Anspruch 1, wobei der Bowman-Birk-Inhibitor aus einer oder mehreren der botanischen Familien Leguminosae, Solanaceae, Gramineae und Cucurbitaceae stammt.
- Verwendung gemäß Anspruch 2, wobei der Bowman-Birk-Inhibitor aus Hülsenfrüchten (Legumen) stammt.
- Verwendung gemäß Anspruch 3, wobei der Bowman-Birk-Inhibitor aus nicht denaturiertem Sojabohnen-Extrakt stammt.
- Verwendung gemäß Anspruch 4, wobei der Bowman-Birk-Inhibitor aus Fraktionen von nicht denaturiertem Sojabohnen-Extrakt stammt.
