Description
Die vorliegende Erfindung betrifft neue Tetrahydronaphthalin-
und Indanderivate der allgemeinen Formel
worin X und Y -CH2- oder ≦λτC(CH2)2; Z einen Rest
-CHR8-, ≦λτCO, ≦λτCR8OR7, -CHR8-CHR8-,
-CHOR7-CH2-, -CO-CHOR7 oder -CHOR7-CHOR7-
darstellt; R1 Fluor, Chlor, Jod, o- oder m-Brom,
nieder-Alkoxy, Acyloxy, Nitro, Hydroxy, Amino, nieder-
Alkylamino, di-nieder-Alkyl-amino oder Phenyl; oder
nieder-Alkyl in o- oder m-Stellung; R2 und R3
Wasserstoff, nieder-Alkyl, Trifluormethyl oder Halogen
und einer der Reste R2 und R3 Wasserstoff,
Trifluormethyl oder nieder-Alkyl darstellt, R4 und
R5 Wasserstoff, Alkyl, Alkoxy oder Halogen; R6
Wasserstoff, Halogen, nieder-Alkyl oder einen Rest
-OR7; R7 Wasserstoff, nieder-Alkyl oder Acyl; R8
Wasserstoff oder nieder-Alkyl; m eine ganze Zahl von 0
bis 5 darstellt; mehrere anwesende Reste R1, R7 oder
R8 untereinander verschiedenen sein können; R6
Wasserstoff ist, wenn gleichzeitig Z -CH2-CH2- und
m = 0 sind; mit Ausnahme des 1,2,3,4-Tetrahydro-1,1,4,4-
tetramethyl-6-(α-methylstyryl)naphthalins und dessen
im terminalen Phenylring hydroxylierten Derivaten.
Die Bezeichnung "nieder" bezieht sich auf Gruppen mit
1-6 C-Atomen. Alkyl- und Alkoxygruppen können geradkettig
oder verzweigt sein, wie Methyl, Aethyl, Propyl, Isopropyl,
Butyl, sek.-Butyl bzw. Methoxy, Aethoxy, Propoxy, Isopropoxy,
Butoxy und sek.-Butoxy. Alkyl- und Alkoxygruppen R4
und R5 enthalten vorzugsweise bis zu 10 C-Atome, wie
Octyl, Nonyl, Decyl und 2,2-Dimethyloctyl bzw. Octyloxy,
Nonyloxy, Decyloxy und 2,2-Dimethyloctyloxy. Beispiele von
Acyloxygruppen sind Alkanoyloxygruppen, vorzugsweise nieder-
Alkanoyloxygruppen, wie Acetoxy, Propionyloxy, Butyryloxy,
Pivaloyloxy und Caproyloxy; oder Aroyloxygruppen wie
Benzoyloxy, p-Nitrobenzoyloxy und Toluoyloxy; oder Aralkanoyloxygruppen
wie Phenylacetoxy; oder Alkoxycarbonylgruppen
wie Methoxycarbonyl und Aethoxycarbonyl. Halogen
umfasst Fluor, Chlor, Brom und Jod.
Die Verbindungen der Formel I können als trans- oder
cis-Isomere oder cis/trans-Isomerengemische vorliegen. Im
allgemeinen sind die trans-Verbindungen der Formel I bevorzugt.
Von den Verbindungen der Formel I sind weiterhin diejenigen
bevorzugt, in denen X und Y eine Gruppe ≦λτC(CH3)2
darstellen, und solche, in denen m = 1 oder 2, insbesondere
1, ist. In Bezug auf die Substituenten R2 und R3 sind
Wasserstoff für R2 und nieder-Alkyl, insbesondere Methyl
für R3 bevorzugt. R4 ist vorzugsweise Wasserstoff oder
Alkyl oder Alkoxy mit bis zu 10 C-Atomen. R5 und R6 sind
vorzugsweise Wasserstoff.
Die Erfindung betrifft weiterhin ein Verfahren zur Herstellung
der Verbindungen der Formel I, pharmazeutische
Präparate auf Basis der Verbindungen der Formel I, die Verbindungen
der Formel I bei der Behandlung und Prophylaxe von
Neoplasien und Dermatosen sowie der Verwendung der Verbindungen
der Formel I bei der Herstellung von pharmazeutischen
Präparaten zur Behandlung und Prophylaxe solcher Erkrankungen.
Die Verbindungen der Formel I können erfindungsgemäss
dadurch erhalten werden, dass man eine Verbindung der allgemeinen
Formel
mit einer Verbindung der allgemeinen Formel
umsetzt,
wobei entweder
A einen der Reste -CH(R3)P⁺(Q)3Y- oder -CH(R3)-P(O)(OAlk)2 darstellt, und B einen Rest R21-CO- darstellt; oder
A einen Rest R31-CO- und B einen der Reste -CH(R2)P⁺(Q)3Y-, oder -CH(R2)-P(O)(OAlk)2 oder -CH(R21)MgHal darstellt; oder
A einen Rest -CH(R31)MgHal und B einen Rest R2-CO- darstellt; wobei in den obigen Formeln Q Aryl: Y- das Anion einer organischen oder anorganischen Säure; Alk eine niedere Alkylgruppe; Hal Halogen; R21 und R31 Wasserstoff, Trifluormethyl oder nieder-Alkyl ist; und R1, R2, R3, R4, R5, X, Y, Z und m die oben angegebene Bedeutung haben und das 1,2,3,4-Tetrahydro-1,1,4,4-tetramethyl-6- (α-methylstyryl)naphthalin und dessen im terminalen Phenylring hydroxylierte Derivate von der Herstellung ausgenommen sind,
worauf gewünschtenfalls eine Nitrogruppe R1 zur Aminogruppe reduziert wird, gewünschtenfalls eine Aminogruppe R1 mono- oder di-alkyliert, gewünschtenfalls eine Acyloxygruppe R1 oder OR7 verseift, eine in Z enthaltene Carbonylgruppe zur Hydroxygruppe reduziert und gewünschtenfalls eine Hydroxygruppe R1 oder OR7 oder eine in Z enthaltene Hydroxygruppe alkyliert oder acyliert.

A einen der Reste -CH(R3)P⁺(Q)3Y- oder -CH(R3)-P(O)(OAlk)2 darstellt, und B einen Rest R21-CO- darstellt; oder
A einen Rest R31-CO- und B einen der Reste -CH(R2)P⁺(Q)3Y-, oder -CH(R2)-P(O)(OAlk)2 oder -CH(R21)MgHal darstellt; oder
A einen Rest -CH(R31)MgHal und B einen Rest R2-CO- darstellt; wobei in den obigen Formeln Q Aryl: Y- das Anion einer organischen oder anorganischen Säure; Alk eine niedere Alkylgruppe; Hal Halogen; R21 und R31 Wasserstoff, Trifluormethyl oder nieder-Alkyl ist; und R1, R2, R3, R4, R5, X, Y, Z und m die oben angegebene Bedeutung haben und das 1,2,3,4-Tetrahydro-1,1,4,4-tetramethyl-6- (α-methylstyryl)naphthalin und dessen im terminalen Phenylring hydroxylierte Derivate von der Herstellung ausgenommen sind,
worauf gewünschtenfalls eine Nitrogruppe R1 zur Aminogruppe reduziert wird, gewünschtenfalls eine Aminogruppe R1 mono- oder di-alkyliert, gewünschtenfalls eine Acyloxygruppe R1 oder OR7 verseift, eine in Z enthaltene Carbonylgruppe zur Hydroxygruppe reduziert und gewünschtenfalls eine Hydroxygruppe R1 oder OR7 oder eine in Z enthaltene Hydroxygruppe alkyliert oder acyliert.
Die Umsetzung der Verbindungen der Formeln II und III
kann nach den bekannten Methoden der Wittig-, Horner- oder
Grignard-Reaktion durchgeführt werden.
Bei der Wittig-Reaktion, d. h., bei Verwendung einer Verbindung
der Formel II mit A = -CH(R3)P⁺(Q)3Y- oder
der Formel III mit B = -CH(R2)P⁺(Q)3Y-, werden die
Komponenten in Gegenwart eines säurebindenden Mittels, z. B.
in Gegenwart einer starken Base, wie z. B. Butyllithium,
Natriumhydrid oder dem Natriumsalz von Dimethylsulfoxyd,
vornehmlich aber in Gegenwart eines gegebenenfalls durch
niederes Alkyl substituierten Aethylenoxyds wie 1,2-Butylenoxyd,
gegebenenfalls in einem Lösungsmittel, z. B. in einem
Aether, wie Diäthyläther oder Tetrahydrofuran, oder in einem
aromatischen Kohlenwasserstoff, wie Benzol in einem zwischen
der Raumtemperatur und dem Siedepunkt des Reaktionsgemisches
liegenden Temperaturbereich miteinander umgesetzt.
Von den anorganischen Säureanionen Y- ist das Chlor-
und Brom-ion oder das Hydrosulfat-ion, von den organischen
Säureanionen ist das Tosyloxy-ion bevorzugt. Der Arylrest Q
ist vorzugsweise ein Phenylrest oder ein substituierter
Phenylrest wie p-Tolyl.
Bei der Horner-Reaktion, d. h. bei Verwendung einer Verbindung
der Formel II mit A = -CH(R3)-P(O)(OAlk)2 oder
der Formel III mit B = -CH(R2)-P(O)(OAlk)2 werden die
Komponenten mit Hilfe einer Base und vorzugsweise in Gegenwart
eines inerten organischen Lösungsmittels, z. B. mit
Hilfe von Natriumhydrid in Benzol, Toluol, Dimethylformamid,
DMSO, Tetrahydrofran, Dioxan oder 1,2-Dimethoxyäthan, oder
auch mit Hilfe eines Natriumalkoholates in einem Alkanol,
z. B. Natriummethylat in Methanol, in einem zwischen 0° und
dem Siedepunkt des Reaktionsgemisches liegenden Temperaturbereich
kondensiert.
Die Alkoxyreste OAlk sind vornehmlich niedere Alkoxyreste
mit 1-6 Kohlenstoffatomen, wie Methoxy oder Aethoxy.
Die Umsetzung einer Verbindung der Formel II mit A =
-CH(R31)MgHal bzw. der Formel III mit B = -CH(R21)MgHal
kann in an sich bekannter Weise unter den Bedingungen einer
Grignard-Reaktion vorgenommen werden, z. B. in einem Aether
wie Diäthyläther oder Tetrahydrofuran bei Raumtemperatur und
anschliessender Wasserabspaltung mit sauren Agentien, z. B.
mit organischen Säuren wie p-Toluolsulfosäure.
Verbindungen der Formel I, die im Phenylring eine Aminogruppe
enthalten (d. h., in denen ein Rest R1 Amino ist)
werden zweckmässig über die entsprechenden Nitroverbindungen
hergestellt. Eine in einer Verbindung der Formel I enthaltene
Nitrogruppe kann in an sich bekannter Weise durch
Reduktion, z. B. mit nascierendem Wasserstoff in eine Aminogruppe
übergeführt werden. Eine in einer Verbindung I vorhandene
Aminogruppe kann in an sich bekannter Weise mono-
oder di-alkyliert werden, z. B. durch Behandlung mit Alkylierungsmitteln
wie Alkylhalogeniden oder -sulfaten, oder durch
reduktive Alkylierung mit Aldehyden wie Formaldehyd oder
Acetaldehyd und Natriumcyanoborhydrid. Die Reduktion einer
in Z enthaltenen Carbonylgruppe sowie die Alkylierung und
Acylierung von Hydroxygruppen kann ebenfalls in an sich bekannter
Weise vorgenommen werden. Beispielsweise kann eine
Carbonylgruppe durch Behandlung mit Reduktionsmitteln wie
Natriumborhydrid zur Hydroxygruppe reduziert werden.
Die Verbindungen der Formel I können in trans- oder
cis-Form vorliegen. Bei der Herstellung fallen sie mehrheitlich
in der trans-Form an. Gegebenenfalls anfallende cis-Anteile
können in an sich bekannter Weise, falls erwünscht,
abgetrennt werden.
Die Ausgangsverbindungen der Formeln II und III können,
soweit ihre Herstellung nicht bekannt oder nachstehend
beschrieben ist, in Analogie zu bekannten oder den nachstehend
beschriebenen Methoden hergestellt werden.
Die Verbindungen der Formel I sind therapeutisch wirksam.
Sie besitzen insbesondere anti-seborrhoische, anti-
keratinisierende, anti-neoplastische und anti-allergische/
anti-inflammatorische Wirksamkeit, die mit den nachstehend
beschriebenen Versuchsanordnungen gezeigt werden
kann:
- A) Die anti-keratinisierende Wirkung kann nach dem folgenden Prozedere am Rhinomausmodel bestimmt werden. Die Haut der Rhinomaus zeichnet sich durch das Vorhandensein von mit Keratin ausgefüllten Utriculi der Epidermis und subkutanen Zysten aus, welche beide von Haarfollikeln herrühren. Die Verabreichung von Retinoiden führt zu einer Hyperproliferation der Epidermis und der epithelialen Auskleidung der Utriculi. Die Verdickung der Epidermis und die Reduktion der Grösse der Utriculi führen zu einer Normalisierung der veränderten Struktur der Epithelschicht. Die tägliche topische Applikation von 0,1 ml/cm2 Haut der Rhinomaus von einer 3%igen Acetonlösung einer wirksamen Versuchsverbindung während 3 Wochen bzw. die dreimalige wöchentliche orale Verabreichung in Arachisöl während 3 Wochen führt zu einer signifikanten Proliferation der Epidermis und markanten Verkleinerung der mit Keratin angefüllten Utriculi.
- B) Die Wirkung bei der Verhütung chemisch induzierter
Mammatumoren kann nach dem folgenden Prozedure bestimmt
werden. Weibliche Sprague-Dawley-Ratten werden unter Temperatur-
und Licht-kontrollierten Bedingungen gehalten, bei
freiem Zugang zu Trinkwasser und Futter. Im Alter von 50
Tagen werden jeder Ratte 15 mg in Arachisöl gelöstes Dimethylbenz(a)anthracen
mittels einer Magensonde verabreicht.
Die Behandlung mit den Versuchs-Verbindungen beginnt 1 Tag
nach der Verabreichung des Karzinogens. Die Körpergewichte
der Versuchstiere werden aufgezeichnet und die Tumoren
wöchentlich palpiert und mit einer Schublehre ausgemessen.
Die Volumina werden nach der Formel D/2 · d 2 berechnet,
wobei D der grössere und d der kleinere Durchmesser des
Tumorellipsoids darstellt. Der Versuch wird nach 11 Wochen
beendet und ausgewertet. In diesem Versuch werden neben den
30 Kontrolltieren, welche ausschliesslich normales Futter
erhalten, die folgenden zwei Gruppen von Versuchstieren eingesetzt:
- 1. 33 Ratten, denen mit dem Futter vermischt täglich 30 mg/kg Versuchs-Verbindung verabreicht werden.
- 2. 36 Ratten, denen mit dem Futter vermischt täglich 90 mg/kg Versuchs-Verbindung verabreicht werden.
- C) Die Wirkung auf Tumoren kann weiterhin am transplantablen
Chondrosarkom der Ratte nach der folgenden Methode bestimmt
werden. Der feste Tumor eines Spendertiers wird fein
zerteilt und in Phosphatpuffer/Kochsalzlösung suspendiert.
0,5 ml des 30%igen Tumorbreis wird Albinoratten subkutan
implantiert.
Die transplantierten Ratten werden auf Versuchsgruppen von je 8 Tieren verteilt. Die Versuchsverbindungen werden in Arachisöl suspendiert und während 24 Tagen fünfmal wöchentlich mittels Schlundsonde oral verabreicht. Die Tumoren werden am Tag 24 exzidiert und gewogen. Die Resultate werden im Quotienten C/T ausgedrückt, der sich wie folgt berechnet: - D) Die antimetaplastische Wirkung kann auch nach der folgenden Methode bei Ratten bestimmt werden. Weibliche Holtzmann-Ratten mit einem Gewicht von ca. 100 g werden nach einer Eingewöhnungszeit von 8 Tagen unter Thiogenalnarkose ovarektomiert und nach weiteren 14 Tagen in den Versuch genommen. Je zwei Tiere werden in einem Käfig untergebracht mit freiem Zugang zu Futter, das ca. 2000 IE analytisch bestimmtes Vitamin A enthält. Vor der oralen Verabreichung der Testverbindung werden die Tiere an 6 aufeinanderfolgenden Tagen täglich mit 1 µg Oestradiolbenzoat und 250 µg Testosteronpropionat, gelöst in 0,1 ml Sesamöl, subkutan behandelt. Die parenterale Hormonapplikation führt zur Ausbildung eines reinen Schollenstadiums im Vaginalbereich, d. h. einer squamösen Metaplasie. 2 Tage nach der oralen Verabreichung der Testsubstanz wird das Reaktionsergebnis wiederum am Vaginalepithel abgelesen. Für die Berechnung der mittleren wirksamen Dosen wird die Flächenmethode nach Behrens und Kärber herangezogen.
- E) Die Wirkung der Verbindungen I auf die Sebum-Ausscheidung bei Ratten wurde nach dem folgenden Prozedere bestimmt. Männliche Ratten von ungefähr 50-60 g Körpergewicht wurden im Alter von 21-22 Tagen kastriert. Eine Woche nach dieser Operation wurden die Ratten in einer Reinigungslösung gewaschen, um Sebum, das vor der Testperiode ausgeschieden wurde, zu entfernen. Einer Gruppe von Ratten wurden lediglich die verwendeten Trägermaterialien verabreicht. Eine weitere Gruppe von Ratten erhielt gleichzeitig auch noch 100 µg Testosteronpropionat in 0,2 ml Sesamöl pro Ratte und Tag. Einer weiteren Gruppe von Ratten wurde täglich pro Ratte 100 µg Testosteronpropionat in 0,2 ml Sesamöl subkutan und die Versuchsverbindungen in verschiedenen Dosen in 0,2 ml Propylenglykol oral verabreicht. Die Ratten wurden während 14 Tagen so behandelt. Am 15. Tag wurde das Sebum von der Hautoberfläche und dem Pelz entfernt, indem der ganze Körper der Versuchstiere in ein bestimmtes Volumen Aceton getaucht und während 2 Minuten darin gebadet wurde. Ein Aliquot des Lösungsmittelbades wurde eingedampft, und der feste Rückstand gravimetrisch bestimmt. Die Inhibierung der Testosteron-stimulierten Zunahme der Sebum-Ausscheidung im Vergleich zu den entsprechenden Werten von nur mit Testosteronpropionat behandelten Ratten wurde als Mass für die Wirkung verwendet.
Die Resultate dieser Versuche A-E mit Verbindungen der
Formel I sind in den nachstehenden Tabellen I-V aufgeführt.

Die Verbindungen der Formel I können zur topischen und
systemischen Therapie von benignen und malignen Neoplasien,
von prämalignen Läsionen, sowie ferner auch zur systemischen
und topischen Propylaxe der genannten Affektionen verwendet
werden.
Sie sind des weiteren für die topische und systemische
Therapie von Akne, Psoriasis und anderen mit einer verstärkten
oder pathologisch veränderten Verhornung einhergehenden
Dermatosen, wie auch von entzündlichen und allergischen
dermatologischen Affektionen geeignet. Die Verfahrensprodukte
der Formel I können ferner auch zur Bekämpfung von
Schleimhauterkrankungen mit entzündlichen oder degenerativen
bzw. metaplastischen Veränderungen eingesetzt werden.
Die Mittel können enteral, parenteral oder topisch
verabreicht werden. Für die enterale Applikation eignen sich
z. B. Mittel in Form von Tabletten, Kapseln, Drag´es, Sirupen,
Suspensionen, Lösungen und Suppositorien. Für die
parenterale Applikation sind Mittel in Form von Infusions-
oder Injektionslösungen geeignet.
Die Dosierungen, in denen die Präparate verabreicht
werden, können je nach Anwendungsart und Anwendungsweg sowie
nach den Bedürfnissen der Patienten variieren. Im allgemeinen
kommen für den Erwachsenen tägliche Dosen von etwa
0,1-50 mg/kg, vorzugsweise 1-15 mg/kg in Betracht.
Die Präparate können in einer oder mehreren Dosierungen
verabreicht werden. Eine bevorzugte Darreichungsform sind
Kapseln mit einem Gehalt von ca. 5-200 mg Wirkstoff.
Die Präparate können inerte oder auch pharmakodynamisch
aktive Zusätze enthalten. Tabletten oder Granula z. B. können
eine Reihe von Bindemitteln, Füllstoffen, Trägersubstanzen
oder Verdünnungsmitteln enthalten. Flüssige Präparate können
beispielsweise in Form einer sterilen, mit Wasser mischbaren
Lösung vorliegen. Kapseln können neben dem Wirkstoff zusätzlich
ein Füllmaterial oder Verdickungsmittel enthalten. Des
weiteren können geschmacksverbessernde Zusätze, sowie die
üblicherweise als Konservierungs-, Stabilisierungs-, Feuchthalte-
und Emulgiermittel verwendeten Stoffe, ferner auch
Salze zur Veränderung des osmotischen Druckes, Puffer und
andere Zusätze vorhanden sein.
Die vorstehend erwähnten Trägersubstanzen und Verdünnungsmittel
können aus organischen oder anorganischen
Stoffen, z. B. aus Wasser, Gelatine, Milchzucker, Stärke,
Magnesiumstearat, Talkum, Gummi arabicum, Polyalkylenglykolen
und dgl. bestehen. Voraussetzung ist, dass alle bei der
Herstellung der Präparate verwendeten Hilfsstoffe untoxisch
sind.
Zur topischen Anwendung werden die Wirkstoffe zweckmässig
in Form von Salben, Tinkturen, Crèmen, Lösungen, Lotionen,
Sprays, Suspensionen und dgl. verwendet. Bevorzugt sind
Salben und Crèmen sowie Lösungen. Diese zur topischen Anwendung
bestimmten Präparate können dadurch hergestellt werden,
dass man die Verfahrensprodukte als wirksamen Bestandteil
nichttoxischen, inerten, für topische Behandlung geeigneten,
an sich in solchen Präparaten üblichen festen oder flüssigen
Trägern zumischt.
Für die topische Anwendung sind zweckmässig ca. 0,1-5%ige,
vorzugsweise 0,3-2%ige Lösungen, sowie ca. 0,1-5%ige,
vorzugsweise ca. 0,3-2%ige Salben oder Crèmen geeignet.
Den Präparaten kann gegebenenfalls ein Antioxydationsmittel,
z. B. Tocopherol, N-Methyl-γ-tocopheramin sowie
butyliertes Hydroxyanisol oder butyliertes Hydroxytoluol
beigemischt sein.
Die nachstehenden Beispiele erläutern die Erfindung
weiter. Die Temperaturen sind in Celsiusgraden angegeben.
45 g [1-(5,6,7,8-Tetrahydro-5,5,8,8-tetramethyl-
2-naphthyl)-äthyl]-triphenylphosphoniumbromid werden in
200 ml 1,2-Butylenoxid suspendiert. Nach Zugabe von 8 g
4-Fluorbenzaldehyd kocht man das Gemisch während 16 Stunden
am Rückfluss. Nach dem Abkühlen giesst man die klare, gelbliche
Lösung in 1 l Methanol/Wasser (6 : 4) und extrahiert
mehrfach mit Hexan. Die organische Phase wird dreimal mit
Wasser gewaschen und nach Trocknen mit Natriumsulfat eingedampft.
Der kristalline Rückstand lässt sich aus Hexan
umkristallisieren und ergibt 11,2 g (E)-6-(p-Fluor-α-
methylstyryl)-1,2,3,4-tetrahydro-1,1,4,4-tetramethylnaphthalin
in farblosen Kristallen, Schmelzpunkt 99-101°.

358 g [1-(5,6,7,8-Tetrahydro-5,5,8,8-tetramethyl-
2-naphthyl)-äthyl]-triphenylphosphoniumbromid werden in
600 ml Tetrahydrofuran suspendiert und bei 0° mit 400 ml
n-Butyllithium (1,6 molar in Hexan) versetzt. Nach 30-minütigem
Rühren bei 0° tropft man eine Lösung von 78,5 g
p-Methoxybenzaldehyd in 200 ml Tetrahydrofuran hinzu und
rührt weitere 2 Stunden bei Raumtemperatur. Anschliessend
giesst man das Reaktionsgemisch in 2 l Methanol/Wasser (6 : 4)
und extrahiert mehrfach mit Hexan. Die organische Phase wird
dreimal mit Wasser gewaschen und nach dem Trocknen mit
Natriumsulfat eingedampft. Der kristalline Rückstand lässt
sich aus Hexan umkristallisieren und ergibt 138 g 1,2,3,4-
Tetrahydro-6-[(E)-p-methoxy-α-methylstyryl]-1,1,4,4-
tetramethylnaphthalin, Schmelzpunkt 108-110°.

6 g 6-[p-Nitro-α-methylstryryl]-1,2,3,4-tetrahydro-
1,1,4,4-tetramethylnaphthalin werden in 200 ml Essigsäure
gelöst und nach Erwärmen auf 90° innerhalb von 20 Minuten
mit 4,5 g aktiviertem Eisenpulver versetzt. Danach gibt man
60 ml Wasser hinzu und versetzt nach weiteren 30 Minuten
erneut mit 60 ml Wasser. Nach 1-stündigem Rühren bei 90°
kühlt man ab, verdünnt das Reaktionsgemisch mit Wasser und
extrahiert mit Aether. Die organische Phase wird mit Wasser,
verdünnter Sodalösung und erneut mit Wasser gewaschen. Nach
dem Trocknen mit Natriumsulfat dampft man ein und erhält ein
braunes Oel, das durch Filtration über Kieselgel (Eluierungsmittel
Hexan/Essigsäure 4 : 1) gereinigt wird. Umkristallisation
aus Hexan ergibt 4,5 g p-[2-(5,6,7,8-Tetrahydro-
5,5,8,8-tetramethyl-2-naphthyl)propenyl]anilin in farblosen
Kristallen, Schmelzpunkt 106-108°.
320 mg (p-[2-(5,6,7,8-Tetrahydro-5,5,8,8-tetramethyl-
2-naphthyl)propenyl]anilin werden in 5 ml Acetonitril
gelöst und bei Raumtemperatur mit 440 mg Acetaldehyd und
190 mg Natriumcyanoborhydrid versetzt. Nach 30 Minuten
stellt man durch Zugabe von Essigsäure ein pH von 6-7 ein
und gibt erneut 440 mg Acetaldehyd hinzu. Nach 2-stündigem
Rühren bei Raumtemperatur giesst man das Reaktionsgemisch
auf Eiswasser, macht durch Zugabe von 2N Kalilauge alkalisch
und extrahiert mit Aether. Das nach Trocknen und Eindampfen
des organischen Lösungsmittels erhaltene bräunliche Oel wird
über Kieselgel filtriert (Eluierungsmittel Hexan/Essigester
9 : 1) und aus Hexan umkristallisiert. Man erhält 280 mg
N,N-Diäthyl-p-[(E)-2-(5,6,7,8-tetrahydro-5,5,8,8-tetramethyl-
2-naphthyl)propenyl]anilin in farblosen Kristallen,
Schmelzpunkt 89-90°.
6,1 g (1,2,3,4-Tetrahydro-1,1,4,4-tetramethyl-6-naphthyl)-
äthylketon werden in 25 ml abs. Aether gelöst und bei
0° zu einer Benzylmagnesiumchlorid-Lösung, hergestellt aus
0,6 g Magnesium und 4,3 g Benzylchlorid in 30 ml abs.
Aether, getropft. Nach 2-stündigem Rühren bei Raumtemperatur
giesst man das Reaktionsgemisch auf eine gesättigte Ammoniumchlorid-
Lösung, extrahiert mit Aether, trocknet über
Natriumsulfat und dampft ein. Das so erhaltene Oel wird in
100 ml Toluol gelöst und nach Zugabe von 0,5 g p-Toluolsulfonsäure
über Nacht am Rückfluss gekocht. Nach dem Abkühlen
versetzt man mit 10%iger Natriumbicarbonat-Lösung, extrahiert
mit Aether, trocknet und dampft ein. Der Rückstand
wird durch Filtration über eine kurze Säule (Kieselgel,
Eluierungsmittel Hexan) gereinigt und aus Methylenchlorid/
Methanol umkristallisiert. Man erhält 3 g 6-(α-Aethylstyryl)-
1,2,3,4-tetrahydro-1,1,4,4-tetramethylnaphthalin in
farblosen Kristallen, Schmelzpunkt 65°.
1,1 g Natriumhydrid (50% in Mineralöl) werden mit abs.
Pentan gewaschen, getrocknet und in 20 ml Dimethylformamid
suspendiert. Unter Eiskühlung tropft man eine Lösung von
5,3 g Diäthyl-benzylphosphat in 50 ml Dimethylformamid
hinzu. Nach 1 Stunde lässt man eine Lösung von 5 g 1,2,3,4-
Tetrahydro-1,1,4,4-tetramethyl-6-naphthyl-aldehyd in 40 ml
Dimethylformamid hinzutropfen und rührt über Nacht bei 40°.
Man giesst das Reaktionsgemisch auf Eis, extrahiert mehrfach
mit Aether, trocknet und dampft ein. Das so erhaltene Oel
wird zur Abtrennung des Z-Isomeren chromatographiert
(Kieselgel, Eluierungsmittel Hexan) und aus Hexan umkristallisiert.
Man erhält 4,1 g (E)-1,2,3,4-Tetrahydro-1,1,4,4-
tetramethyl-6-styrylnaphthalin in farblosen Kristallen,
Schmelzpunkt 57-58°.
In Analogie zu Beispiel 6 werden durch Wittig-Horner-
Reaktion von 6,8 g Diäthyl-(5,6,7,8-tetrahydro-5,5,8,8-
tetramethyl-2-naphthyl)methanphosphonat mit 2,4 g Acetophenon
nach Umkristallisation aus Methanol 1,5 g (E)-1,2,3,4-Tetrahydro-
1,1,4,4-tetramethyl-6-(β-methylstyryl)naphthalin
in farblosen Kristallen erhalten, Schmelzpunkt 72-73°.
Das in diesem Beispiel verwendete Phosphonat lässt sich
in einfacher Weise ausgehend von 1,2,3,4-Tetrahydro-1,1,4,4-
tetramethyl-6-naphthyl-aldehyd herstellen: Reduktion mit
Natriumborhydrid in Aethanol zur entsprechenden Hydroxymethylverbindung
(Schmelzpunkt 78° aus Pentan), Ueberführung
in die Brommethylverbindung durch Reaktion mit Phosphortribromid
(Siedepunkt 125°/0,01 mm) und Reaktion mit Triäthylphosphit
(16 Stunden, 150°, Schmelzpunkt 55° aus Hexan).
In Analogie zu Beispiel 1 werden aus 20 g [1-[5,6,7,8-
Tetrahydro-2-naphthyl)äthyl]triphenylphosphoniumbromid und
4 g Benzaldehyd nach Chromatographie (Kieselgel, Eluierungsmittel
Hexan) 4,6 g 1,2,3,4-Tetrahydro-6-(α-methylstyryl)-
naphthalin als farbloses Oel erhalten, Siedepunkt ca.
170°/0,01 mm).
Zu einer Suspension von 3,7 g NaH (50% in Mineralöl) in
50 ml Dimethylformamid gibt man bei Raumtemperatur eine
Lösung von Diäthylbenzylphosphonat in 30 ml Dimethylformamid.
Nach 15-minütigem Rühren bei Raumtemperatur tropft
man eine Lösung von 12,6 g 7-Acetyl-1,1,4,4,6-pentamethyltetralin
in 60 ml Dimethylformamid im Laufe von 2 Stunden
hinzu. Das Reaktionsgemisch wird über Nacht bei Raumtemperatur
gerührt und anschliessend noch 1 Stunde auf 60° erwärmt.
Nach dem Abkühlen giesst man auf Eis, extrahiert mit Aether,
trocknet und dampft ein. Nach Chromatographie des Rohproduktes
(Kieselgel, Eluierungsmittel Hexan) und Kristallisation
aus Hexan erhält man 3,9 g 1,2,3,4-Tetrahydro-1,1,4,4,7-
pentamethyl-6-[(E)-α-methylstyryl]naphthalin in farblosen
Kristallen, Schmelzpunkt 75-77°.
In Analogie zu Beispiel 9 wird aus Diäthylbenzylphosphonat
und 5,6,7,8-Tetrahydro-5,5,8,8-tetramethyl-3-octyl-2-
acetonaphthon 1,2,3,4-Tetrahydro-1,1,4,4-tetramethyl-
6-(α-methylstyryl)-7-octylnaphthalin hergestellt,
Schmelzpunkt 47-48° (aus Hexan).
In Analogie zu Beispiel 1 werden aus 7,1 g [1-(1,1,3,3-
Tetramethyl-indan-2-on-5-yl)äthyl]triphenylphosphoniumbromid
und 1,4 g Benzaldehyd nach Chromatographie (Kieselgel,
Eluierungsmittel Hexan/Aether = 9 : 1) und Umkristallisation
aus Hexan 800 mg das 1,1,3,3-Tetramethyl-5-[(E)-α-
methylstyryl]-2-indanon erhalten, Schmelzpunkt 83-85°.
Das als Ausgangsprodukt verwendete Phosphoniumbromid
lässt sich in einfacher Weise durch Friedel-Crafts-Acetylierung
von 1,1,3,3-Tetramethylindanon, Reduktion der
Acetylgruppe mit Natriumborhydrid und Umsetzung mit Triphenylphosphoniumbromid
herstellen.
1,4 g 1,1,3,3-Tetramethyl-5-[(E)-α-methylstyryl]-2-
indanon werden in 100 ml Aethanol gelöst und bei Raumtemperatur
mit 6 g Natriumborhydrid versetzt. Nach 16-
stündigem Rühren bei Raumtemperatur giesst man das Reaktionsgemisch
auf Eis und extrahiert mehrfach mit Aether. Die
organische Phase wird mit gesättigter Natriumchloridlösung
gewaschen, getrocknet und eingedampft. Der Rückstand lässt
sich aus Hexan umkristallisieren und ergibt 1,1 g 1,1,3,3-
Tetramethyl-5-[(E)-α-methylstyryl]-2-indanol in farblosen
Kristallen, Schmelzpunkt 63-67°.
Zu einer aus 1,90 g Benzylchlorid und 437 mg Mg-Spänen
in 30 ml abs. THF hergestellten Grignard-Lösung werden bei
0° 2,80 g 6′-(tert-Butyldimethylsiloxy)-5′,6′,7′,8′-tetrahydro-
5′,5′,8′,8′-tetramethyl-2-acetonaphthon, in 10 ml
abs. THF, getropft. Nach 15 Minuten hydrolysiert man mit
H2O, extrahiert mit Aether und wäscht die organischen
Phasen gründlich mit H2O. Nach Trocknen und Abziehen der
Lösungsmittel wird der Rückstand, ein viskoses Oel, in 20 ml
CH2Cl2 aufgenommen und mit 150 mg p-Toluolsulfonsäure
versetzt. Nach 6 Stunden filtriert man über Kieselgel, und
behandelt das Rohprodukt ca. 14 Stunden bei 40° mit 6,3 g
nBu4NF · 3 H2O in 20 ml THF. Das Reaktionsprodukt wird
zwischen Wasser und Aether verteilt, die organische Phase
mit Wasser gewaschen, getrocknet und eingedampft. Filtration
über Silicagel liefert 2,27 g eines Oels, das alle drei
möglichen Doppelbindungsisomeren enthält. Um diese zu
äquilibrieren, behandelt man mit 200 mg p-Toluolsulfonsäure
in 20 ml CHCl3 bei 45-50°. Nach 24 Stunden chromatographiert
man über eine kurze Silicagelsäule (Petroläther : AcOEt
= 7 : 3) und kristallisiert aus Hexan um. Einmalige Wiederholung
der Isomerisierung mit der Mutterlauge liefert insgesamt
1,32 g 1,2,3,4-Tetrahydro-1,1,4,4-tetramethyl-6[(E)-
α-methylstyryl]-2-naphthalenol, Smp. 102-103°.
Das Ausgangsmaterial kann wie folgt erhalten werden:
p-Bromphenylessigsäure wird durch doppelte Alkylierung,
Ueberführung in das Säurechlorid und Tandem-Friedel-Crafts-
Reaktion des Säurechlorids mit Isobutylen unter SnCl4-
bzw. AlCl3-Katalyse in 6-Brom-3,4-dihydro-1,1,4,4-tetramethyl-
2(lH)-naphthalenon übergeführt, aus dem man durch
NaBH3-Reduktion und Silylierung mit TBDMS-Cl/Imidazol das
[(6-Brom-1,2,3,4-tetrahydro-1,1,4,4-tetramethyl-(2-naphthyl)-
oxy]tert.-butyl-dimethylsilan erhält. Grignard-Reaktion mit
Acetaldehyd und MnO2-Oxidation liefert das 6′-(tert.-
Butyldimethylsilyloxy)-5′,6′,7′,8′-tetrahydro-5′,5′,8′,8′-
tetramethyl-2-acetonaphthon.
In Analogie zu Beispiel 13 erhält man aus Benzylmagnesiumchlorid
und 7′-(tert.-Butyldimethylsilyloxy)-
5′,6′,7′,8′-tetrahydro-5′,5′,8′,8′-tetramethyl-2-acetonaphthon
das 1,2,3,4-Tetrahydro-1,1,4,4-tetramethyl-
7-[(E)-α-methylstyryl]-2-naphthalenol, Smp. 89-91°.
Das Ausgangsmaterial kann wie folgt erhalten werden:
m-Brombenzylcyanid wird durch doppelte Alkylierung und
basische Hydrolyse in 2-(m-Bromphenyl)-2-methylpropionsäure
überführt, die in Analogie zu den in Beispiel 13 beschriebenen
Verfahrensstufen über das 7-Brom-3,4-dihydro-1,1,4,4-
tetramethyl-2(1H)-naphthalenon weiter umgesetzt wird.
144 mg einer 50%igen Natriumhydrid-Dispersion werden in
3 ml Dimethylformamid suspendiert und mit 740 mg Diäthylfluorphenylmethanphosphonat
(hergestellt am Benzylfluorid
durch radikalische Bromierung mit N-Bromsuccinimid und
Umsetzung mit Triäthylphosphit) versetzt. nach 2-stündigem
Rühren bei Raumtemperatur tropft man 1,15 g 5,6,7,8-Tetrahydro-
5,5,8,8-tetramethyl-2-acetonaphthon zu und erwärmt
über Nacht bei 55°. Nach dem Abkühlen giesst man auf Eiswasser,
und extrahiert mit Aether. Nach dem Trocknen und
Eindampfen der organischen Phase erhält man 1,3 g
Rohprodukt, das durch Chromatographie an Kieselgel (Eluierungsmittel
Hexan/Essigester = 99 : 1) gereinigt wird und nach
Umkristallisation aus Hexan 77 mg 6-[(Z)-β-Fluor-α-methylstyryl]-
1,2,3,4-tetrahydro-1,1,4,4-tetramethylnaphthalin
ergibt, Smp. 100°.
In Analogie zu Beispiel 1 erhält man aus 19,4 g
[1-(1,1,3,3-Tetramethyl-5-indanyl)äthyl]triphenylphosphoniumbromid,
6,9 g Aethyl-p-formylphenylcarbonat und
200 ml Butylenoxid nach Filtration des Rohproduktes über
Kieselgel (Eluierungsmittel Hexan/Essigester = 19 : 1) 5 g
Aethyl p-[2-(1,1,3,3-tetramethyl-5-indanyl)propenyl]-
phenylcarbonat als gelbliches Oel, das in der Kälte erstarrt
und aus Hexan umkristallisiert werden kann. 5 g des so
erhaltenen Produktes werden in 50 ml Aethanol gelöst und mit
einer Lösung von 7,4 g Kaliumhydroxid in 25 ml Wasser versetzt.
Nach 3-stündigem Rühren bei Raumtemperatur giesst man
auf Eiswasser, säuert mit 3N Salzsäure an, extrahiert mit
Essigester und dampft ein. Nach dem Umkristallisieren des
Rohproduktes aus Hexan erhält man 2,6 g p-[2-(1,1,3,3-
Tetramethyl-5-indanyl)propenyl]phenol in farblosen
Kristallen, Smp. 137°.
Das als Ausgangsprodukt verwendete Aethyl p-formyl-phenylcarbonat
lässt sich in einfacher Weise durch Reaktion
von p-Hydroxybenzaldehyd mit Chlorameisensäureäthylester
unter Zusatz von Triäthylamin herstellen. Destillation des
Rohproduktes ergibt Aethyl-p-formyl-phenylcarbonat als
farblose Flüssigkeit, Sdp. 111-113°/2,5 mm.
In Analogie zu Beispiel 1 erhält man aus 21,9 g
[1-(1,1,3,3-Tetramethyl-indan-2-on-5-yl)äthyl]triphenylphosphoniumbromid,
7,6 g Aethyl-p-formyl-phenylcarbonat und
400 ml Butylenoxid nach Filtration des Rohproduktes über
Kieselgel (Eluierungsmittel Hexan/Essigester = 4 : 1) und
Umkristallisation aus Hexan/Essigester 3,4 g Aethyl-p-[2-
(1,1,3,3-tetramethyl-2-oxo-5-indanyl)propenyl]phenylcarbonat,
Smp. 130-131°.
Hydrolyse dieses Produktes mit einem Ueberschuss Kaliumhydroxid
in Aethanol/Wasser ergibt nach Umkristallisation
aus Essigester/Hexan 1,7 g 5-(p-Hydroxy-α-methylstyryl)-
1,1,3,3-tetramethyl-2-indanon in farblosen Kristallen, Smp.
172-173°.
Zu einer Suspension von 100 mg Lithiumaluminiumhydrid in
5 ml Tetrahydrofuran tropft man unter Eiskühlung eine Lösung
von 1 g 5-(p-Hydroxy-α-methylstyryl)-1,1,3,3-tetramethyl-
2-indanon in 10 ml Tetrahydrofuran und lässt anschliessend
2 Stunden bei Raumtemperatur führen. Nach dem Zutropfen von
50 ml 2N Salzsäure bei 0° extrahiert man mit Essigester,
wäscht die organische Phase mit Wasser, trocknet und dampft
ein. Nach Filtration des Rohproduktes über Kieselgel (Eluierungsmittel
Hexan/Essigester = 1 : 1) kristallisiert man aus
Essigester/Hexan um und erhält 500 mg 5-(p-Hydroxy-α-
methylstyryl)-1,1,3,3-tetramethyl-2-indanol in farblosen
Kristallen, Smp. 148-149°.
In Analogie zu Beispiel 2 erhält man aus [1-(5,6,7,8-
Tetrahydro-3-methoxy-5,5,8,8-tetramethyl-2-naphthyl)äthyl]-
triphenylphosphoniumbromid und Aethyl-(p-formyl-phenyl)-
carbonat das Aethyl [p-[(E)-2-(5,6,7,8-tetrahydro-3-methoxy-
5,5,8,8-tetramethyl-2-naphthyl)propenyl]phenyl]carbonat,
Smp. 122-123°.
Hydrolyse dieses Produktes mit einem Ueberschuss Kaliumhydroxid
in Aethanol/Wasser ergibt nach Umkristallisation
aus Methanol p-[(E)-2-(5,6,7,8-Tetrahydro-3-methoxy-5,5,8,8-
tetramethyl-2-naphthyl)propenyl]phenol, Smp. 181°.
In Analogie zu Beispiel 5 erhält man
aus 7-Acetyl-6-chlor-1,2,3,4-tetrahydro-1,1,4,4-tetramethylnaphthalin und Benzylmagnesiumchlorid (E)-6-Chlor- 1,2,3,4-tetrahydro-1,1,4,4-tetramethyl-7-(α-methylstyryl) naphthalin, Smp. 114°,

Die Herstellung von Gebrauchsformen der Verbindungen der
Formel I kann in üblicher Weise, z. B. anhand der nachstehenden
Beispiele erfolgen.

Das sprühgetrocknete Pulver, das auf dem Wirkstoff,
Gelatine und mikrokristalliner Cellulose basiert und eine
mittlere Korngrösse des Wirkstoffes von ≦ωτ1 µ aufweist
(mittels Autokorrelationsspektroskopie gemessen), wird mit
einer wässrigen Lösung von Natriumcarboxymethylcellulose und
Natriumdioctylsulfosuccinat befeuchtet und geknetet. Die
resultierende Masse wird granuliert, getrocknet und gesiebt,
und das erhaltene Granulat mit mikrokristalliner Cellulose,
Talk und Magnesiumstearat vermischt. Das Pulver wird in
Kapseln der Grösse 0 abgefüllt.
Tabletten können wie folgt hergestellt werden:

Die feingemahlene Substanz wird mit Milchzucker und
einem Teil der Maisstärke gemischt. Die Mischung wird mit
einer wässrigen Lösung von Povidone K30 befeuchtet und
geknetet, und die resultierende Masse granuliert, getrocknet
und gesiebt. Das Granulat wird mit der restlichen Maisstärke,
Talk und Magnesiumstearat vermischt und zu Tabletten
geeigneter Grösse verpresst.
Weichgelatinekapseln können wie folgt hergestellt werden:

10 g Verbindung I werden unter Rühren, Inertbegasung und
Lichtschutz in 90 g mittelkettigem Triglycerid gelöst. Diese
Lösung wird als Kapselfüllmasse zu Weichgelatinekapseln à
50 mg Wirkstoff verarbeitet.
Eine Lotion kann wie folgt hergestellt werden:

Der Wirkstoff wird unter Lichtschutz in die Mischung
Aethanol, 94%ig/Wasser eingearbeitet. Carbopol 934 wird bis
zur vollständigen Gelierung eingerührt und der pH-Wert mit
Natriumhydroxid eingestellt.

1. Verbindungen der allgemeinen Formel
worin X und Y -CH2- oder ≦λτC(CH3)2; Z einen Rest
-CHR8-, ≦λτCO, ≦λτCR8OR7, -CHR8-CHR8-,
-CHOR -CH2-, -CO-CHOR7 oder -CHOR7-CHOR7-
darstellt; R1 Fluor, Chlor, Jod, o- oder m-Brom,
nieder-Alkoxy, Acyloxy, Nitro, Hydroxy, Amino, nieder-
Alkylamino, di-nieder-Alkyl-amino oder Phenyl; oder
nieder-Alkyl in o- oder m-Stellung; R2 und R3
Wasserstoff, nieder-Alkyl, Trifluormethyl oder Halogen
und einer der Reste R2 und R3 Wasserstoff,
Trifluormethyl oder nieder-Alkyl darstellt, R4 und

1,2,3,4-Tetrahydro-1,1,4,4-tetramethyl-6-(α-methylstyryl)-
naphthalins und dessen im terminalen Phenylring hydroxylierten
Derivaten.
3. Verbindungen gemäss Anspruch 1 oder 2, in denen die
olefinische Doppelbindung trans-Konfiguration hat.
4. Verbindungen gemäss den Ansprüchen 1-3, in denen X
und Y eine Gruppe ≦λτC(CH3)2 darstellen.
5. Verbindungen gemäss den Ansprüchen 1-4, in denen Z =
-CH2-CH2- und m = 1 oder 2 ist.
6. Verbindungen gemäss den Ansprüchen 1-5, in denen R3
Methyl und R2 Wasserstoff ist.
7. Verbindungen gemäss den Ansprüchen 1-6, in denen
R4, R5 und R6 Wasserstoff sind.
8. Eine Verbindung aus der Reihe der folgenden:

14. Pharmazeutische Präparate, enthaltend eine Verbindung
der Formel I und übliche Träger- oder Hilfsstoffe.
15. Pharmazeutische Präparate, in Dosiseinheitsform,
enthaltend 5 bis 200 mg einer Verbindung der Formel I pro
Dosierungseinheit.
16. Die Verbindungen der Formel I zur Anwendung als
Heilmittel.
17. Verwendung einer Verbindung der Formel I zur Herstellung
von pharmazeutischen Präparaten zur Behandlung von
Neoplasien und Dermatosen.
18. Verfahren zur Herstellung der Verbindungen der
Formel I aus Anspruch 1, dadurch gekennzeichnet, dass man
eine Verbindung der allgemeinen Formel
mit einer Verbindung der allgemeinen Formel
umsetzt,
wobei entweder
A einen der Reste -CH(R3)P⁺(Q)3Y- oder -CH(R3)-P(O)COAlk)2 darstellt, und B einen Rest R21-CO- darstellt; oder
A einen Rest R31-CO- und B einen der Reste -CH(R2)P⁺(Q)3Y-, oder -CH(R2)-P(O)(OAlk)2 oder -CH(R21)MgHal darstellt; oder
A einen Rest -CH(R31)MgHal und B einen Rest R21-CO- darstellt; wobei in den obigen Formeln Q Aryl; Y- das Anion einer organischen oder anorganischen Säure; Alk eine niedere Alkylgruppe; Hal Halogen; R21 und R31 Wasserstoff, Trifluormethyl oder nieder-Alkyl ist; und R1, R2, R3, R4, R5, X, Y, Z und m die in Anspruch 1 angegebene Bedeutung haben und das 1,2,3,4-Tetrahydro-1,1,4,4-tetramethyl- 6-(α-methylstyryl)naphthalin und dessen im terminalen Phenylring hydroxylierte Derivate von der Herstellung ausgenommen sind,
worauf gewünschtenfalls eine Nitrogruppe R1 zur Aminogruppe reduziert wird, gewünschtenfalls eine Aminogruppe R1 mono- oder di-alkyliert, gewünschtenfalls eine Acyloxygruppe R1 oder OR7 verseift, eine in Z enthaltene Carbonylgruppe zur Hydroxygruppe reduziert und gewünschtenfalls eine Hydroxygruppe R1 oder OR7 oder eine in Z enthaltene Hydroxygruppe alkyliert oder acyliert.
wobei entweder
A einen der Reste -CH(R3)P⁺(Q)3Y- oder -CH(R3)-P(O)COAlk)2 darstellt, und B einen Rest R21-CO- darstellt; oder
A einen Rest R31-CO- und B einen der Reste -CH(R2)P⁺(Q)3Y-, oder -CH(R2)-P(O)(OAlk)2 oder -CH(R21)MgHal darstellt; oder
A einen Rest -CH(R31)MgHal und B einen Rest R21-CO- darstellt; wobei in den obigen Formeln Q Aryl; Y- das Anion einer organischen oder anorganischen Säure; Alk eine niedere Alkylgruppe; Hal Halogen; R21 und R31 Wasserstoff, Trifluormethyl oder nieder-Alkyl ist; und R1, R2, R3, R4, R5, X, Y, Z und m die in Anspruch 1 angegebene Bedeutung haben und das 1,2,3,4-Tetrahydro-1,1,4,4-tetramethyl- 6-(α-methylstyryl)naphthalin und dessen im terminalen Phenylring hydroxylierte Derivate von der Herstellung ausgenommen sind,
worauf gewünschtenfalls eine Nitrogruppe R1 zur Aminogruppe reduziert wird, gewünschtenfalls eine Aminogruppe R1 mono- oder di-alkyliert, gewünschtenfalls eine Acyloxygruppe R1 oder OR7 verseift, eine in Z enthaltene Carbonylgruppe zur Hydroxygruppe reduziert und gewünschtenfalls eine Hydroxygruppe R1 oder OR7 oder eine in Z enthaltene Hydroxygruppe alkyliert oder acyliert.
