Claims (1)
- Patentanspruch:Verfahren zur Herstellung einer Pyruvatoxidase für ein Reagenz für analytische Zwecke durch Züchten eines Mikroorganismus und Isolieren der Pyruvatoxidase aus den gezüchteten Zellen, dadurch gekennzeichnet, daß man als Mikroorganismus Aerococcus viridans IFO 12219, Aerococcus viridans IFO 12317, Pediococcus sp. FERM BP-465 oder Streptococcus sp. FERM BP-466 züchtet .Die Erfindung bezieht sich auf ein Verfahren zur Gewinnung von Pyruvatoxidase.Pyruvatoxidase ist ein schon bekanntes Enzym, das die Umsetzung von Brenztraubensäure in Anwesenheit von Phosphat und Sauerstoff zu Acetylphosphat, Kohlendioxid und Wasserstoffperoxid katalysiert und das aus einer Spezies von Lactobacillus delbrücki gewonnen werden konnte.Es wurde nun gefunden, daß ein Enzym Pyruvatoxidase produziert wird von einem Stamm, der zum Genus Pediococcus gehört und hinterlegt ist beim FRI unter der Hinterlegungsnummer FERM BP-465 und von einem Stamm, der zum Genus Streptococcus gehört und hinterlegt ist beim FRI unter der Hinterlegungsnummer FERM BP-466, isoliert aus einer Bodenprobe, die an dem Rettichfeld in Ohito-cho, Tagatagun, Shizuoka-ken, Japan, gesammelt wurde, sowie aus den Bakterienstämmen IFO12219 und IFO 12317, die zum Genus Aerococcus gehörecGe-gen-stad der vorliegenden Erfindung ist dementsprechend ein Verfahren zur Herstellung einer Pyruvatoxidase durch Züchten eines Mikroorganismus und Isolieren der Pyruvatoxidase aus den gezüchteten Zellen, das erfindungsgemäß dadurch gekennzeichnet ist, daß man als Mikroorganismus Aerococcus viridans IFO 12219, Aerococcus viridans IFO 12317, Pediococcus sp. FERM BP-465 oder Streptococcus sp. FERM BP-466 züchtet. Die erfindungsgemäß gewonnene Pyruvatoxidase ist brauchbar als Reagenz für analytische Zwecke; sie ist insbesondere für diagnostische Analysen gut geeignet.Für das erfindungsgemäße Verfahren brauchbare Organismenstämme Efdiococcus sp. B-0667 (= FERM BP-465) und Streptococcus sp. B-0668 (= FERM BP-466) haben die folgenden taxonomischen Eigenschaften.In der Zeichnung zeigtFig. 1 in grafischer Darstellung das Optimum des pH-Wertes für Pyruvatoxidase,Fig. 2 in p.rafischer Darstellung die Wärmestabilität von Pyruvatoxidase, undFig. 3 in grafischer Darstellung die pH-Stabilität von Pyruvatoxidase.
