Abstract

Proposed is a method of imaging part of a medical or biological object using electromagnetic radiation together with a contrast medium introduced before the exposure is made. The contrast medium is introduced by prior art biophysical and/or mechanical and/or chemical methods into the interior of a biological cell surrounded by a membrane and acting as a carrier, the membrane being impermeable to the contrast medium. By virtue of its medico-biological properties and/or by virtue of antigenes or antibodies placed on the surface of the membrane, the carrier is concentrated in the area of the object to be imaged, and simultaneously and/or subsequently the exposure is made.

Description

The invention relates to a method for spatially resolving image of a biological area object with the help of electromagnetic radiation before application of contrast medium guidance of the recording and a contrast medium for Use for this.

In medicine, there are various illustrations Principles based on and above all for diagnosis imaging processes used for physical purposes Entrance found. Some of the be most significant procedures are shown with Help of electromagnetic radiation, for what as a case pass on the X-rays familiar to everyone name is. But also fluorescence spectroscopy is attributable to this process. Will be a timed constant magnetic field can also be superimposed NMR spectroscopy (nuclear magnetic resonance) and the elec tron spin resonance (ESR) can be performed. At the The aim of mapping biological structures is Introduction of contrast media before implementation radiation. So with the sub search for vessels, muscles and tissues before  Contrast agents are introduced, which the better visualization of the vessels, the tissue etc. serve. There is a lot on the market number of different contrast agents, which are described in Ab dependence on the mapping process used the tissue to be examined and the load ability of the test person selected and appli be decorated. The contrast media must be sometimes after a short time in the body be dismantled or eliminated.

The decisive disadvantage is that in the case of imaging of the blood vessels or with Er averaging the regional blood volume (= volume of the Blood vessels divided by the total volume) often intravenously administered contrast medium in the Spread body, d. H. starting from the Blutge would diffuse into the neighboring cell tissue ren, with the result that the respective environment also experiences an image and therefore a overall low-contrast picture results. The region nale blood volume is an important physiological Parameters for assessing the function of an Or goose. Another disadvantage is that the contrast medium, going beyond ever areas to be examined throughout the body distributes what the application a corresponding large dose of contrast medium on the one hand and the in itself unnecessary stress on other parts of the body on the other hand has the consequence.

Proceeding from this, the invention has turned to  made in medical imaging or biology, in which the illustration by elek tromagnetic radiation occurs, the images to be imaged Areas of the object, such as body organs, etc. reproduce it clearly.

According to the invention, this object is achieved by that the contrast medium by known bio physical and / or mechanical and / or chemi cal process inside a carrier adjusting and by a biocompatible membrane closed container is introduced, wherein the membrane for the contrast medium is impermeable, that the wearer is medically biological property and / or due to antigen applied to the surface of the membrane or antibodies in the area to be imaged Object enriches and at the same time and / or finally the recording is made.

The two different proposed solutions serve the realization of the same goal, namely the targeted enrichment of the body to be imaged weave with contrast medium. Both solutions in common is the use of a biocompatible, from a Membrane enclosed container, wor among cells, liposomes and vesicles, as aqueous phase to be understood surrounded by a membrane. Inside the membrane Volumens with the help of known biophysical and / or mechanical processes and / or on chemi contrast medium introduced and inserted closed. All known contrasts can be used here  medium use, provided with regard to the respective membrane is impermeable, so that after it has been introduced into the interior of the enclosed container safely and permanently stay closed. Enclosing the contrast with through the membrane means no impairment and loss of later image quality. The Art and manner of introducing the contrast agent biophysical and / or mechanical and / or che Mixed processes are known to me and have an effect a brief, brief opening of the membrane, through which the contrast medium penetrates into the interior can, taking appropriate measures the membrane reappears after a short time is closed.

J. 14, 881) in Frage. Z. Physiol. Chem. 352, 527), ein hypotoner Schock (Borle & Snowdowne 1982: Science 217, 242-254), os motische Lyse von Pinosomen (Okada & Rechsteiner 1982: Cell 29, 33-41), mechanisches Abschaben der Zellen (McNeil 1984: J. Cell Biol. 98, 1556-1564) sowie Rühren mit Glaskugeln (McNeil & Warder 1987: J. Cell Sci. 88, 669-678). As a biophysical process, with the help of The contrast medium can be introduced, comes primarily electric injection (room mann et al 1974: Biophys. J. 14, 881) in question. But also the ultrasound treatment (Ganbihler et al 1994: J. Membrane Biol. 141, 257-275) and also High-speed microparticles Klein et al 1987: Nature 327, 70-73) can be considered. Mechanical processes can be used microinjection (Graessmann 1971: Hoppe Seyl. Z. Physiol. Chem. 352, 527), a hypotonic shock (Borle & Snowdowne 1982: Science 217, 242-254), os Motive lysis of pinosomes (Okada & Rechsteiner 1982: Cell 29, 33-41), mechanical scraping of the Cells (McNeil 1984: J. Cell Biol. 98, 1556-1564) and stirring with glass balls (McNeil & Warder 1987: J. Cell Sci. 88, 669-678).

Zur chemischen Permeabilisierung können eingesetzt werden: ATP (Rozengurt & Heppel 1975: BBRC 67, 1581-1588, EDTA (Johnsen et al 1985: JBC 260, 2069-2076), CaPO4 (Graham & van Erb 1973: Virology 52, 456-467), DEAE-Dextran (Vaheri & Pagano 1965: Viro logy 27, 435-436).Chemical permeabilization can be used become: ATP (Rozengurt & Heppel 1975: BBRC 67, 1581-1588, EDTA (Johnsen et al 1985: JBC 260, 2069-2076), CaPO4 (Graham & van Erb 1973: Virology 52, 456-467), DEAE dextran (Vaheri & Pagano 1965: Viro logy 27, 435-436).

It is crucial for the invention to cause this container, which is enclosed by a membrane and is now doped with contrast medium, to migrate and accumulate in a targeted manner in those areas that need to be examined and reproduced. To this end, the invention proposes two un different solutions:
On the one hand, the use of certain containers, cells, etc. can ensure that this desired accumulation occurs in certain areas of the body. Thus, the use of white blood cell cells can transport the enclosed contrast medium to inflammation sites and accumulate there accordingly. On the other hand, if erythrocytes (= red blood cells) are used, the result is that the cells doped accordingly with contrast agent remain in the bloodstream, so that they map the vessels and not the environment. Erythrocytes have the property of not penetrating into and diffusing into the tissue. In this case, known properties of the container used as a carrier, primarily the cell or liposomes, are used and exploited.

In the proposed alternatives, too Cells etc. are used as carriers that are not the property of concentration on a particular th area in the subject's body. The Information about the goal to be achieved by applying antigens and antibodies on the surface of the membrane, which is after the Key-lock principle on the corresponding com attach complementary cells of the object. In the appropriate choice of an antibody he can suffice that the carriers are only at certain Zel len, such as B. attach tumor cells and him mark follow. The carriers themselves then need in Deviation from the first described alternatives not to contain any target information.

With both proposed solutions you get one targeted enrichment of the contrast medium in be matched fabrics and thus targeted marking of the areas of the body to be imaged, consequently the production of high-contrast and meaningful eng illustrations allowed. The contrast medium is in comparatively small amounts targeted used for later recording.

It has already been mentioned that in terms of choice of the to be introduced in the carrier and through the membrane enclosed contrast medium all already known connections are used and used can, provided they are impermeable to the membrane are. As a preferred contrast medium for NMR Imaging is gadolinium-DTPA (= Gadolinium-Di  called ethylenetriaminepentaacetate); a special one would be a suitable contrast medium for X-rays Barium sulfate.

In the case of the possibility of quick execution the admission and / or the subjects Contrast agent compounds would be a short life duration or dwell time of the contrast medium in the body per advantage. On the other hand, with several images to be recorded at intervals and to avoid repeated use tel can be used with a long service life. According to Another proposal of the invention is a ge targeted influencing of the lifespan possible Lich that through the well-known biophysical and / or mechanical processes and / or on chemi openings made in the membrane, through which the contrast medium to be absorbed into the In nere of the wearer can penetrate in their through knife can be varied. If it is chosen low, the membrane closes very quickly and also in the With regard to the entire surface largely Completely. The result is a long service life of the contrast medium inside. Become however, introduced large diameters increases in contrast, the damage caused to the membrane and the probability that the original Liche restores state of the membrane correspondingly lower. The result is that at Lö chern large diameter a shorter life occurs and the decay of the contrast agent goes faster. With appropriate choice the process parameters during doping he  the lifetime of the Contrast agent.

In terms of NMR imaging, there are a lot Number of admission procedures is basically conceivable. In Dependence on the chosen contrast medium sub punch and from the desired image (vessels, Functions, structure) one becomes the corresponding one choose the best admission procedure. Especially before is added when using the Gadolinium DTPA Recording method in which the NMR relaxation time is detected. In addition to NMR imaging, is also suitable the method according to the invention in particular also in the production of images by X-ray recordings, by electron spin resonance (ESR), through short-term laser spectroscopic imaging or by fluorescence spectroscopy. Clarify remains that when using the NMR or ESR method which the electromagnetic rays additionally by a constant magnetic field in itself are known to be superimposed.

In addition to the procedure outlined above Requests for protection expressly also in the inventions senses prepared and immediately ap contrast medium ready for application. It be is found inside one closed by one sene membrane of limited container, which as Serves the carrier and is provided with information, that serve as a target for transportation. Here for example, this is used as a cell reali container itself or through on the  Surface of the membrane attached to the outside Tigens or antibodies that develop after the key sel-lock principle only to the corresponding com attach complementary cells of the object.
