Verfahren zur Herstellung und Isolierun, hor- haltiger Gardenolid- und ,enolidgl kos de Die Erfindung betrifft die Herstellung und Iso- lierung bonhaltiger Cardenoli.d- und !!tx#tdien- olidglykoside (im folgenden Dorsäureeeter ge- nannt). Die Borsäure ist in den erfindungsge- mäß hergestellten Verbindungen eeterartig ge- bunden. Die Borsäureester der Gardenolid- und _ Butadienolidglykoside (im folgenden Herzgly-- koside öenannt) zeichnen eich durch eine-be$-- sere enterale Wirksamkeit und Vertxäglichkei.t aus als sie die unveränderten @erzglykoside haben. he Ist bekannt, Borsäureester von z. B. Oxy- anthrachinonen mittels Tetraacetatoborat her- zustellen (DIIMOTH, 0. und FAUST, T, über die Borsäureester der Oxyanthrachinone."#' Berichte der Dtsch. Ohem. Ges. ,54, 3029 (1921». :-[ei- terhin sind bonhaltige Gardenolidglykoeide als Zwischenprodukte bei der Reduktion von 0-19 Aldehydaardenolidglykosiden mittels Na- triumborhydrid bekannt. Die Herstellung von Dorsäureestern der Herz. glykoside mittels Tetraacetatoborat ist je- doch nicht in der Literatur beschrieben, da offenbar eine Anwendung der Methode auf die chemisch und thermisch instabilen Herzglyko- side allgemein für nicht möglich gehalten wuxzde. Die Darstellung von bonhaltigen Zwi.- schen=ierbi.ndun3en bei der Reduktion von C-19 Aldehydherzglykosiden ist experimentell auf- wendig und naturgemäß nur bei Aldehydglyko- si.den anwendbar. Der Erfindung liegt die Aufgabe zugrunde, .die j wie festgestellt-wurde, therapeutisch wertvollen Borsäuroester der Herzglykoeide in guter Ausbeute auf einfache Yleise heriustgl- len, zu isolieren und wenn gewünscht wieder zu spalten. Erfindungsgemäß wird das zu veresternde Herz- glykosid mit einem 2 bis 5 molarem Ubersch4ß von auf übliche Weise hergestelltem Tetraaze-. tatoborat (z. B. nach 2ICTET und GHLEZNOW, Be- richte Dtsch. Chem. Ges. 26, 2219 (1903» bei Temperaturen innerhalb*des Stabilitätsberei- chesder gelösten Glykoside in einem geeigne- ten Lösunsniittel, z. B. Dioaan, umgesetzt. Dabei fällt der entstandene Borsäureester fast quantitativ aus und kann durch Filtration oder Zentrifugation isoliert werden. Dabei ist es gleichgültig, ob die Herzglykoeide in reiner oder angereicherter, roher Form vorliegen, da Nebenbestandteile und ni.cht- glykosidische Verunreinigungen in der Reäel keine schwerlöslichen Borsäureester ergeben. Je nach dem Grad der Schwerlöslichkeit bil- den sich Mono, Di, Tri- oder höhere Bof- säureester.
