技术领域
本发明一般涉及到生物微阵列技术。更具体说,本发明提供了一种加工生物微阵列的流体系统和方法。例如,可将所述发明应用于个人设备,但应知道本发明具有更广阔的应用范围。
背景技术
生物微阵列通常包括用于提取核酸样品序列信息的核酸探针,在能够发生杂交的某些条件下使核酸样品与该核酸探针接触,然后对形成的生物微阵列进行扫描,以测定该核酸样品与那种探针杂交。根据这种测定,通过比较杂交和非杂交模式从而获得序列信息。例如,可利用这种序列信息确定核酸的序列,或对遗传性疾病或特定病原体或病原株的存在进行诊断性筛检。
形成生物微阵列然后扫描常常通过流体系统进行。例如,这种流体系统包括一种流体工作台。此流体工作台可以洗涤和染色该微阵列。随着微阵列设计的进展,常常需要改进这种流体工作台以促进其自动化和降低成本。
因此,非常需要改进的加工微阵列的流体技术。
发明内容

组合性合成策略:组合性合成策略是一种通过依次加入反应性矩阵和转换性矩阵所提供的试剂来平行合成多种多样聚合物序列的顺序方法,其产物是一种产物矩阵。反应性矩阵是待加入的构建模块的m排矩阵所形成的1个柱。转换性矩阵是所有的二进数或其一亚组,优选顺序排列在柱中的1和m之间。“二进制方法”是可照射该基质上感兴趣区域一部分,常常是感兴趣区域一半的至少二个连续步骤的一种方法。在一种二进制合成方法中,可形成所有的可能从顺次反应性矩阵组所形成的化合物。在最优选实施方式中,二进制合成指其它因素作为预先加入步骤的一种合成方法。例如,该方法中进行遮盖方法的转换矩阵将已预先照射的区域对半分,照射已照射区域的约一半,并保护其余的一半(同时也保护原先已受保护区域的约一半并照射原先已受保护区域的约一半)。将认识到,双数轮次可相间散布着非双数轮次,并且只有基质的一部分可进行此二进制方式。组合性“遮盖”性方法,是利用光或其它空间选择性脱保护剂或激活剂来去除物质的保护性基团而能加入其它物质如氨基酸的合成方法。
有效量指能充分诱导产生所需结果的量。
基因组是某生物染色体中的所有遗传物质。某特定生物的染色体中遗传物质衍生的DNA是基因组DNA。
基因组文库是一系列随机产生的代表某生物整个基因组的重叠性DNA片段所制备克隆的集合体。杂交条件通常包括不到约1M,更常不到500mM,优选区不到约200mM的盐浓度。杂交温度可低至5℃,但通常高于22℃,更通常高于约30℃。优远超过约37℃。对于特异性杂交,较长的片段可能需要较高的杂交温度。其它影响杂交严格性的因素包括互补链的碱基组成和长度、有机溶剂的存在和碱基错配的程度,这些参数的组合比任何一个单独参数的绝对值更重要。
杂交,例如等位基因特异性探针的杂交通常在严格条件下进行。例如,在盐浓度不高于1M和温度至少25℃的条件,如750mM NaCl、50mM磷酸钠、5mM EDTA、pH7.4(5X SSPE)和温度约35-30℃的条件下进行。

术语“杂交”指二条单链多核苷酸非共价结合形成稳定的双链多核苷酸的过程,三链杂交在理论上也是可能的。所产生的(通常)双链多核苷酸即是“杂交体”。能形成稳定杂交体的多核苷酸群的比例本文称为“杂交程度”。
杂交探针是能以碱基特异性方式结合核酸互补链的寡核苷酸。这类探针包括肽核酸,如Nielsen等,Science 254:1497-1500(1991)所述,以及其它核酸类似物和核酸模拟物。
特异性杂交:指在严格条件下某分子只与存在于复杂的(例如,细胞总)DNA或RNA混合物中的特定核苷酸序列结合、形成双链体或杂交。
分离的核酸是本发明的优势存在(即以摩尔计比该组合物中任何其它单一核酸更丰富)的目标核酸。分离的核酸优选包括所有存在的巨分子核酸的至少约5%、80%或90%(以摩尔计)。最优选将该目标核酸纯化至基本上均一性(用常规检测方法不能在该组合物中检测到污染物质)。
配体:配体是一种为特定受体所识别的分子。能与受体结合或反应的物质称为“配体”,此术语定义上只意味就其配对受体而言。术语“配体”不意味该分子有任何特定的大小或与所述物质相比具有其它结构上或组成上的特征,只要能与其受体结合或相互反应。配体也可以是能结合受体的天然配体,或能起着激动剂或拮抗剂作用的功能类似物。本发明所研究的配体例子包括但不限于:细胞膜受体的激动剂和拮抗剂,毒素和毒液、病毒表位、激素(如鸦片剂、类固醇等)、激素受体、肽、酶、酶底物、底物类似物、过渡态类似物、辅因子、药物、蛋白质和抗体。连锁失平衡或等位基因结合指特定的等位基因或遗传标记偏爱与附近染色体位置处的特异性等位基因或标记结合,其频率高于该群体中任何特定等位基因偶然性结合所预计的频率。例如,如果基因座X具有等位基因a和b,其发生频率相等,连锁的基因座Y具有等位基因c和d,其发生的频率相等,则预计ac结合发生的频率为0.25,如果ac发生的频率更高,那么等位基因a和c的连锁失平衡。连锁失平衡可发生自等位基因某些结合的天然性选择,或因为已将某等位基因太过于引入某群体而不能达到与连锁的等位基因相平衡。
混合群或复合群:指含有所需的和不需要的核酸二者的任何样品。作为非限制性例子,核酸的复合群可以是总基因组DNA、总基因组RNA或它们的混合物。然而,可富集某给定群的核酸复合群但不包含其它不需要的核酸群。例如,核酸的复合群可以是富集了所需信使RNA(mRNA)序列但仍含有某些不需要的核糖体RNA(rRNA)序列的样品。
单体:指可连接在一起形成寡聚物或多聚物的一组分子中的任何成员。可用于本发明例如多肽合成的单体组包括但不限于:L-氨基酸、D-氨基酸或合成的氨基酸组。“单体”本文用于指合成某寡聚物碱基组的任何成员。例如,L-氨基酸的二聚体形成了合成多肽的400个“单体”碱基组。在合成多聚体的连续步骤中可采用不同的单体碱基组。
术语“单体”也指一种化学亚单位,其可与不同的化学亚单位组合形成比单独亚单位更大的化合物。mRNA或mRNA转录物:如本文所用,包括但不限于:前mRNA转录物、转录加工中间体、成熟的已能进行基因翻译和转录的mRNA,或该mRNA转录物衍生的核酸。转录加工包括:剪接、编辑和降解。mRNA转录物衍生的核酸,本文用于指该mRNA转录物或其亚序列最终作为模板而合成的核酸。因此,cDNA逆转录自某mRNA,从该cDNA再转录生成RNA,从该cDNA扩增产生DNA,从拉增的DNA转录生成RNA等等都有衍生自该mRNA转录物,测到这种衍生产物表明样品中存在和/或富含该最初的转录物。因此,mRNA衍生的样品包括但不限于:基因的mRNA转录物、从该mRNA逆转录的cDNA、从该cDNA转录的cRNA、从该基因扩增的DNA、从扩增的DNA转录的RNA等。
核酸文库或阵列意思是可用化学合成法或生物合成法制备产生的核酸集合体,用于以各种不同模式筛选生物学活性(如可溶性分子的文库;结合于树脂珠、二氧化硅芯片、或其它固体支持物上的寡聚物文库)。此外,术语“阵列”意指包括点缀在一基质上的基本上可以是任何长度(如长1-1000个核苷酸的单体)的核酸所制备的那些核酸文库。术语“核酸”本文用于指任何长度的核苷酸,或核糖核苷酸、脱氧核糖核苷酸或肽核酸(PNA)的聚合形式,包括嘌呤和嘧啶碱基、或其它天然的、化学或生物化学方法修饰的非天然的、或衍生的核苷酸碱基。多核苷酸的骨架可包含糖或通常见于RNA或DNA中的磷酸基团,或修饰的或取代的糖或磷酸基团。多核苷酸可含有修饰的核苷酸,如甲基化的核苷酸和核苷酸类似物。核苷酸的序列可被非核苷酸组分间隔。因此术语核苷、核苷酸、脱氧核苷和脱氧核苷酸通常包括本文描述的那些类似物。这些类似物是具有与天然产生的核苷或核苷酸所共有的某些结构特征的那些分子,因此当将其掺入核酸或寡核苷序列中时,它们能在溶液中与天然产生的核酸序列杂交。通常这些类似物可通过对天然产生的核苷和核苷酸的碱基、核糖或磷酸二酯键部分的置换和/或修饰而产生。可定制这些变化以使杂交模式稳定或不稳定,或提高与所述互补核酸序列杂交的特异性。
本发明的核酸可包括分别由嘧啶和嘌呤碱基,优选胞嘧啶、胸腺嘧啶和尿嘌呤、腺嘌呤与鸟嘌呤组成的聚合物或寡聚物。见Albert L.Lehninger,PRICIPLES OFBIOCHEMISTRY,第93-800页(Worth Pub.1982)。的确,本发明考虑了任何脱氧核糖核苷酸、核糖核苷酸、肽核酸组分,和它们的任何化学变体,如这些碱基的甲基化、羟甲基化或糖基化形式等。多聚物或寡聚物在组成上可以是异质性的或同质性的,可以分离自天然来源或经人工或合成方法产生。此外,核酸可以是DNA或RNA或它们的混合物,可以单链或双链形式永久或短时存在,包括同质双链体、异质双链体和杂交体。
“寡核苷酸”或“多核苷酸”是长至少2个,优选至少8个,更优选至少20个核苷酸的核酸,或能特异性杂交多核苷酸的化合物。本发明的多核苷酸包括:分离自天然来源、重组产生的或人工合成的脱氧核糖核酸(DNA)或核糖核酸(RNA)的序列,和它们的模拟物。本发明多核苷酸的另一例子可以是肽核酸(PNA)。本发明也包括非传统性碱基配对,如在某些tRNA分子中已鉴定到的推测以三螺旋存在的Hoogsteen碱基配对。“寡核苷酸”或“多核苷酸”在此申请书中可互换使用。
探针:探针是可被特定靶分子所识别的固定于表面的分子。见美国专利No.6,582,908中具有10、12和更多碱基的所有可能组合的阵列例子。可为本发明研究的探针例子包括但不限于:细胞膜受体的激动剂和拮抗剂,毒素和毒液、病毒表位、激素(如鸦片样肽、类固醇等)、激素受体、肽、酶、酶底物、辅因子、药物、凝集素、糖、寡核苷酸、核酸、寡糖、蛋白质和单克隆抗体。
引物是一种单链寡核苷酸,能在适当条件下,如缓冲液和温度、及存在4种不同核苷三磷酸和聚合试剂如DNA或RNA聚合酶或逆转录酶时,作为模板引导的DNA合成的起始点。在某些给定情况下,引物的长度取决于,例如,所用引物一般长15-30个核苷酸。短的引物分子通常需要较低的温度与模板形成充分稳定的杂交复合物。引物不需要反映模板的精确序列但应与此模板充分互补杂交。引导位点是引物与模板杂交的区域。引物对是一组引物包括能与被扩增序列5’端杂交的5’上游引物,和能与被扩增序列3’端互补序列杂交的3’下游引物。
多态性指群体中存在二种或多种遗传上决定性的可变序列或等位基因。多态性标记或位点是发生趋异性的基因座。在所选择的群体中优势标记具有至少二个等位基因,各自发生的频率大于1%,更优选大于10%或20%。多态性可包括一个或多个碱基的改变、插入、重复或缺失。多态性基因座可以小至一个碱基对。多态性标记包括片段长度限制的多态性、不同数目的串联重复序列(VNTR)、超变区、微小卫星现象、双核苷酸重复序列、三核苷酸重复序列、四核苷酸重复序列、单序列重复、和插入元件如Alu。最先鉴定到的等位基因形式是随意称为的参考形式和其它称为可变的或变异的等位基因形式。最常发生在所选群体中的等位基因形式有时称为野生型。二倍体生物可以是等位基因形式的纯合子或杂合子。双等位基因多态性具有二种形式。三等位基因多态性具有三种形式。单个核苷酸多态性(SNP)包括在多态性内。
受体:一种对给定配体具有亲和力的分子。受体可以是天然产生的或人造的分子。它们也可以其固定不变的状态或与其它物质的凝聚物使用。受体可直接或通过特异性结合物质共价或非共价结合于其结合伙伴。本发明可采用的受体的例子包括但不限于:抗体、细胞膜受体、单克隆抗体和能与特异性抗原决定簇(如病毒、细胞或其它物质上的)起反应的抗血清、药物、多核苷酸、核酸、肽、辅因子、凝集素、糖、多糖、细胞、细胞膜、和细胞器。受体有时本领域称之为抗-配体。本文所用的术语受体在含意上没有差别。当二个大分子通过分子识别而结合形成复合体时即形成“配体受体对”。本发明所研究的受体的其它例子包括但不限于美国专利No.5,143,854中所示的那些分子,其内容纳入本文作参考。
“固相支持物”、“支持物”和“基质”可互换使用,指具有坚硬或半坚硬表面的物质或一组物质。在许多实施方式中,该固相支持物的至少一个表面是基本上平的。虽然在某些实施方式中可能需要为不同的化合物准备物理上分隔的合成区域,例如孔格、抬高区域、尖点、蚀刻槽等。按照其它实施方式,此固相支持物将采取小珠、树脂、凝胶、微球形式或其它几何形态。示范性的基质见美国专利5,744,305。
靶分子:对某给定探针具有亲和力的一种分子。靶分子可以是天然产生的或人造的分子。它们也可以其固定不变的状态或与其它物质的凝聚物使用。靶分子可直接或通过特异性结合物质共价或非共价结合于其结合伙伴。本发明可采用的靶分子的例子包括但不限于:抗体、细胞膜受体、单克隆抗体和能与特异性抗原决定簇(如病毒、细胞或其它物质上的)起反应的抗血清、药物、多核苷酸、核酸、肽、辅因子、凝集素、糖、多糖、细胞、细胞膜和细胞器。靶分子有时本领域称之为抗-探针。本文所用的术语靶分子在含意上没有差别。当二个大分子通过分子识别而结合形成复合体时即形成“探针靶分子对”。

流体组件110包括有至少几个容器,这些容器各装有用于加工至少一个生物传感器的一种或多种流体。例如,每个容器是一孔格。另一实施例中,这些孔格成组分布在一个或多个平板和/或一个或多个板条中。如图1-4所示,诸孔格成组分布在至少一个孔格板112和孔格条114中。一实施方式中,孔格板112含有96个孔格,孔格条114含有4个孔格。另一实施方式中,每孔格装有一种或多种流体,而不同的孔格装有相同或不同的流体。还有一实施方式中,不同的孔格具有相同或不同的深度。

数个管510为一个或多个传感器提供了数个孔格。例如,数个管510的每个管中装有至少一种流体,加热器512加热此流体,温差电偶514直接或间接监测该流体的温度。温差电偶514起反应时将信号发给温度控制器。一实施方式中,温度控制器也是该流体系统的一个组件。温度控制器加工接受到的目标物温度信号并调节加热器512的功率以使该流体达到目标温度。如图1所示,用户可通过本发明实施方式的温度显示面板140向该流体系统110提供目标温度。温度显示面板140是流体系统110的一个组件。
图6是显示本发明一实施方式加工生物传感器的流体系统110中对孔格条114而言温度与时间函数关系的简图。此图只是示范例,不应错误理解为对本发明权利要求范围的限制。本领域普通技术人员知道可作出许多变化、替代和修饰。如图6所示,曲线610可提供温度计514所测定的作为时间函数的温度。曲线620提供了数个管510例如孔格中流体的温度,也是时间的函数,其它温度计如温差电偶测定了数个管中的流体温度。曲线620显示在给定的时间互相靠近的不同管中的流体温度,这些流体的温度在一段时间后稳定在约50℃。
如图1-4所示,流体组件110还包含板条座116和118。在一实施方式中,板条座用于在进行杂交过程后装入一个或多个传感器运送给流体系统110。例如,板条座118是一种杂交用托盘,在另一实施方式中,利用板条座将一个或多个传感器运送到流体系统110之外。例如,板条座118包含至少一透明小杯座。
支持组件120包含至少一底板122。例如,底板122水平安置。如图1-4所示,流体组件110放置在底板122上。例如,流体组件110可直接或间接固定在底板122上的预定位置。在另一实施例中,该流体组件中的一个或多个孔格基本垂直于底板122。
如图1-4所示,孔格板112包含排列成多行和多列的多个孔格。例如,行数等于或大于列数。另一实施例中,多行的每一行从该孔格板的一侧伸展至该孔格板的另一侧。在一实施方式中,孔格条114和二个板条座116与118都安置在紧靠孔格板112的一侧。另一实施例中,多行孔格与多列孔格相互垂直。此外孔格条114可包含一行孔格。板条座116和118各包含一行孔格,能容纳一个或多个传感器。一实施方式中,孔格条114与板条座116与118的多行孔格相平行。另一实施方式中,孔格条114与板条座116与118的多列孔格与多行孔格相平行。
按照一实施方式,孔格板112、孔格条114、板条座116和/或板条座118包含多个孔格。该多个孔格中至少一孔格装有一种或多种流体。例如,各孔格对一种或多种流体的容量等于或小于3毫升或5毫升。一实施方式中,进行低严格性洗涤和染色的一种或多种流体的体积约为1.9毫升,进行高严格性洗涤的约为3.7毫升。另一实施方式中,不同的孔格具有相同或不同的深度。
电子机械组件130可将一个或多个传感器之一从一个位置移动到另一位置。例如,一个或多个传感器不是该电子机械组件的一部分。另一实施例中,电子机械组件130用作运送组件。又一实施例中,以一维、二维或三维方式移动这些传感器。还有一实施例中,以不同速度移动这些传感器。如图1-4所示,电子机械组件130可将一个或多个传感器之一移入流体组件110的相应孔格中,和/或可将一个或多个传感器之一移入或移出流体组件110的相应孔格中。
按照本发明的一个实施方式,电子机械组件130包含至少夹具132和电机134、136和138。此外,电机134、136和138各能在一维平面的两个相反方向上移动一个或多个传感器。例如,电机134可以垂直于底板122的两个相反方向移动一个或多个传感器。另一实施方式中,电机136和138可以平行于底板122的方向移动一个或多个传感器。在一实施方式中,电机136可以平行于孔格条114的方向移动一个或多个传感器。在还有一个实施方式中,电机138可以垂直于孔格条114的方向移动一个或多个传感器。
此外,电子机械组件130可包含其它组件。例如,电子机械组件130可包含至少一高压电源和低压电源。一实施方式中,例如,利用此电源向电机134、136和138提供预定值的和/或预定范围内的电压。另一实施方式中,该电源从外部接受115伏的AC电压。

驱动器710可用来移动臂720和730。一实施方式中,驱动器710是电子驱动器。另一实施方式中,驱动器710是气动驱动器。例如,气动驱动器可接受气体调制仪的第一压力气体,而气体调制仪可将第二压力的气体转变成第一压力的气体。另一实施例中,所述气体是干燥的清洁气体。如图1所示,通过流体系统100的组件压力计150监测第一压力。此外,臂720包括多个爪指,如爪指722、724、726和728,见图7(A)。类似地,臂730也包括多个爪指。如图7(B)所示,利用夹具132来夹住一个或多个传感器。例如,在一个或多个传感器不是系统100的一部分时。

例如,系统800的流体组件可用机盖810部分或完全封闭。另一实施例中,系统800的流体组件包括孔格板、孔格条和二个板条座。该孔格板包含排列成多行和多列的多个孔格。例如,行数等于或大于列数。另一实施例中,多行孔格的每一行从该孔格板的一侧伸展至该孔格板的另一侧。在一实施方式中,孔格条和二个板条座都安置在紧靠孔格板的第一侧。另一实施例中,多行与多列孔格相垂直。此外孔格条可包含一行孔格。板条座各包含一行孔格,能容纳一个或多个传感器。一实施方式中,孔格条和板条座的多行孔格相平行。另一实施方式中,孔格条和板条座的多列孔格与多行孔格相平行。

按照本发明的一实施方式,可利用加工系统的电子机械组件接受的指令控制一个或多个传感器移入和/或移出流体系统100或800。例如,该加工系统可外接流体系统。另一实施例中,该加工系统是此流体系统的一个组件。一实施方式中,该加工系统包括计算机或处理器。例如,可通过密码指导该计算机或处理器。另一实施例中,通过计算机程序产品中的计算机可读媒介包含的指令指导此计算机或处理器。

进行低严格性洗涤的各步骤910、940和960时,在室温下洗涤多个孔格中的一个或多个传感器之一。例如,所述多孔格包括二个孔格。另一实施例中,多孔格包括4个孔格。每个孔格中,相应的传感器与孔格中的流体混合多次。例如,混合多次包括在3分钟内混合23次。另一实施例中,混合多次包括在2分钟内混合36次。
在高严格性洗涤步骤920时,至少一孔格中有一个或多个传感器。升高每个孔格中的流体温度使相应的传感器与流体混合一段时间。例如,温度升高到48℃,时间是25分钟。另一实施例中,温度升高到41℃,时间也是25分钟。
进行SAPE染色各步骤930和970时,在室温下至少一个孔格中染色一个或多个传感器之一。每个孔格中,相应的传感器与流体混合一段时间,例如,混合时间为10分钟。
进行AB染色步骤950时,在室温下至少一个孔格中染色一个或多个传感器之一。每个孔格中,相应的传感器与流体混合一段时间,例如,混合时间为10分钟
如上所述和此处进一步强调的,步骤910、920、930、940、950、960、970和980都用本发明一实施方式的流体系统100或800进行。在步骤910之前,使一个或多个传感器杂交。例如,杂交步骤在48℃下进行16小时。进行步骤980后扫描一个或多个传感器。

按照一实施方式,图10(A)-(V)的各图显示了孔格板112、孔格条114和板条座116与118。例如,孔格板含有排列成1-12行和A-H列的96个孔格。行1用于SAPE染色,行3用于AB染色,行5-12用于低严格性洗涤。另一实施例中,孔格条112用于高严格性洗涤,在还有一实施例中,板条座116是一种在进行杂交后将一个或多个传感器送至流体系统100的杂交托盘。在还有一实施例中,板条座118包含一透明小杯座,用于从流体系统100中输送出一个或多个微阵。
为了描述一个或多个传感器的移动,所用的孔格时或已用后的孔格以影线方块标识。如图10(A)所示,通过板条座116将一个或多个传感器运送到流体系统100。然后,按图10(B)-(E)所示步骤910以低严格性洗涤处理一个或多个传感器。分别在行12、11、10和9的4个孔格中洗涤各传感器。每个孔格中,以基本上平行于孔格深度的方向搅拌传感器使其与孔格中的流体混合多次。进行步骤920后,将一个或多个传感器移入孔格条114中进行图10(F)所示的高严格性洗涤。
图10(G)显示一个或多个传感器从孔格条114移动到孔格板112的行1孔格中。在行1孔格中,按步骤930用SAPE染色该一个或多个传感器。进行步骤940时,如图10(H)-(K)所示对一个或多个传感器再进行低严格性洗涤。分别洗涤行8、7、6和5的4个孔格中的各传感器。每个孔格中,以基本上平行于孔格深度的方向搅拌传感器使其与孔格中的流体混合多次。

如上所述和此处进一步强调的,流体系统100或800可用于加工本发明某些实施方式的生物传感器。例如可按方法900进行此加工。另一实施例中,生物传感器可有不同类型。见2004年4月16日提交的美国专利No.10/826,577和2005年10月4日提交的11/243,621,其各自内容纳入本文作参考。另一实施例中,一个或多个生物传感器是生物微阵列。一实施方式中,该生物微阵列具有传感器的长度、传感器的宽度和传感器的厚度。例如,传感器的长度等于或短于10毫米,传感器的宽度等于或窄于10毫米,传感器的厚度等于或薄于1000微米。另一实施例中,传感器的长度等于或短于6.3毫米,传感器的宽度等于或窄于6.3毫米m,传感器的厚度等于约700微米。

按照再一实施方式,加工生物传感器的方法包括在至少第一传感器和第二传感器上进行杂交步骤。杂交步骤后,将第一传感器和第二传感器称转移到一流体系统。该流体系统包括至少一第一容器和一第二容器。第一容器容纳第一体积的第一流体,而第二容器容纳第二体积的第二流体。此外,该方法包括将第一传感器移入第一容器中与第一体积的第一流体接触,和将第二传感器移入第二容器中与第二体积的第二流体接触。移动第一传感器和第二传感器基本上是同时进行的。例如,可按照系统100和/或系统800提供此流体系统。
本发明具有各种应用。例如,流体系统100或流体系统800可用作流体工作台。一实施方式中,如图1所示该流体工作台的底宽19.5英寸深17.5英寸。此外,该流体工作台高15英寸。另一实施例中,本发明的某些实施方式可用于个人和手提仪器以及加工生物微阵列的方法。
本发明具有各种优点。本发明的某些实施方式提供自动化的流体系统。本发明的某些实施方式提供低成本的流体系统。本发明的某些实施方式可提供整个流体系统。例如,可平行加工多个生物传感器如微阵列。本发明的某些实施方式可减少在一个或多个生物传感器上进行不同加工时的交叉污染。例如,在一给定加工中,可在不同的孔格中洗涤、染色和/或保持不同的传感器。另一实施例中,在不同的孔格中分别对某给定的传感器进行洗涤、染色和/或保持进行不同加工。在还有一实施例中,各个最多用于一次加工最多一个传感器如一个微阵列。
虽然已描述了本发明的具体实施方式,但本领域技术人员知道还有与描述的这些实施方式相等价的其它实施方式。因此,应理解本发明的范围不限于这些具体描述的实施方式,而只受本说明书所附的权利要求书的限制。
Claims (37)
1.一种加工生物传感器的流体系统,该流体系统包括:
流体组件,此组件至少包括第一容器和第二容器,第一容器能容纳第一体积的第一流体,而第二容器能容纳第二体积的第二流体;
支持组件,其结构支持至少所述第一容器和第二容器,所述第一容器和第二容器相对于该支持组件基本上是静止不动的;
运送组件,该组件的结构为:
相对于支持组件将第一传感器移入第一容器中与第一体积的第一流体接触;相对于支持组件将第二传感器移入第二容器中与第二体积的第二流体接触;
其中所述第一传感器和第二传感器基本上是同时移动的。
2.如权利要求1所述的流体系统,其特征在于,
当第一传感器位于第一容器内并与第一体积的第一流体接触时,该第一体积的第一流体完全留在第一容器中;
当第二传感器位于第二容器内并与第二体积的第二流体接触时,该第二体积的第二流体完全留在第二容器中。
3.如权利要求1所述的流体系统,其特征在于,所述第一传感器结合于第一支持组件。
4.如权利要求4所述的流体系统,其特征在于,所述第一支持组件是一种销钉桩。
5.如权利要求4所述的流体系统,其特征在于,所述第二传感器结合于第二支持组件。
6.如权利要求5所述的流体系统,其特征在于,其中所述第一支持组件和第二支持组件各自是一孔格板的一部分。
7.如权利要求1所述的流体系统,其特征在于,
所述第一传感器具有传感器的长度、传感器的宽度和传感器的厚度;
传感器的长度等于或短于10毫米;
传感器的宽度等于或窄于10毫米;
传感器的厚度等于或薄于1000微米。
8.如权利要求1所述的流体系统,其特征在于,所述第一传感器是生物传感器。
9.如权利要求8所述的流体系统,其特征在于,所述生物传感器包括微阵列。
10.如权利要求1所述的流体系统,其特征在于,所述第一流体传感器的第一体积等于或小于5毫升。
11.如权利要求1所述的流体系统,其特征在于,所述支持组件还构成对至少第一容器和第二容器的直接支持。
12.如权利要求1所述的流体系统,其特征在于,所述支持组件还通过另一物体构成对至少第一容器和第二容器的间接支持。
13.如权利要求1所述的流体系统,其特征在于,所述第一流体和第二流体在种类上相同或不相同。
14.如权利要求1所述的流体系统,其特征在于,所述第一体积和第二体积在容量上相同或不相同。
15.如权利要求1所述的流体系统,其特征在于,所述第一容器和第二容器各为一孔格。
16.如权利要求1所述的流体系统,其特征在于,所述第一容器和第二容器是多个容器中的二个,该多个容器直接或间接结合于一共有组件上。
17.如权利要求16所述的流体系统,其特征在于,所述多个容器排列成一行或多行和一列或多列。
18.一种加工生物传感器的流体系统,该流体系统包括:
流体组件,此组件至少包括第一容器和一第二容器,第一容器能容纳第一体积的第一流体,而第二容器能容纳第二体积的第二流体;
支持组件,其包括支持至少所述第一容器和第二容器的底板,所述第一容器和第二容器相对于该底板基本上是静止不动的;
运送组件,该组件包括一夹具和至少一个电机;其中:
所述夹具能基本上同时夹住第一传感器和第二传感器,并基本上同时松开第一传感器和第二传感器;
所述至少一个电机的结构能相对于底板将夹住的第一传感器移入第一容器并与第一体积的第一流体接触;
所述至少一个电机的结构还能相对于底板将夹住的第二传感器移入第二容器并与第二体积的第二流体接触;
其中所述第一传感器和第二传感器基本上是同时移动的。
19.如权利要求18所述的流体系统,其特征在于,所述夹具包括一驱动器。
20.如权利要求19所述的流体系统,其特征在于,所述驱动器是气动驱动器。
21.如权利要求19所述的流体系统,其特征在于,所述驱动器是电动驱动器。
22.如权利要求18所述的流体系统,其特征在于,
所述至少一个电机能在至少6个方向移动夹住的第一传感器;
所述6个方向的每一个与6个方向中的另一个方向相反并与6个方向中的4个方向垂直;
所述6个方向中的4个方向不同于所述另一个方向。
23.如权利要求18所述的流体系统,其特征在于,所述流体组件至少被一机盖部分封闭。
24.如权利要求18所述的流体系统,其特征在于,
所述第一容器和第二容器连接于一温度相关的组件;
该组件包括结构能加热第一体积的第一流体和第二体积的第二流体的加热器;
该组件还包括结构能测量温度的温度计。
25.如权利要求18所述的流体系统,其特征在于,该系统还包括连接于加热器和温度计的温度控制仪,且其结构能将温度加热至预定值。
26.如权利要求18所述的流体系统,其特征在于,所述运送组件连接于一处理系统,该处理系统可向此运送组件提供夹持、松开或移动第一传感器和第二传感器的指令。
27.如权利要求26所述的流体系统,其特征在于,所述处理系统包括一计算机。
28.如权利要求18所述的流体系统,其特征在于;
当第一传感器位于第一容器内并与第一体积的第一流体接触时,该第一体积的第一流体完全留在第一容器中;
当第二传感器位于第二容器内并与第二体积的第二流体接触时,该第二体积的第二流体完全留在第二容器中。
29.如权利要求18所述的流体系统,其特征在于,所述第一传感器结合于第一支持组件。
30.如权利要求29所述的流体系统,其特征在于,所述第一支持组件是一种销钉桩。
31.如权利要求29所述的流体系统,其特征在于,所述第二传感器结合于第二支持组件。
32.如权利要求30述的流体系统,其特征在于,其中所述第一支持组件和第二支持组件是一孔格板的一部分。

所述第一传感器具有传感器的长度、传感器的宽度和传感器的厚度;
