本发明总体上涉及生物微阵列技术。更具体地说,本发明提供用于处理大量生物微阵列的系统和方法。仅举例而言,可将本发明描述为应用于96-栓钉(peg)仪器,但应该理解的是本发明具有更广阔的应用范围。
生物微阵列往往包含用于从核酸样品中提取序列信息的核酸探针。核酸样品在允许杂交的特定条件下与核酸探针接触。然后处理生物微阵列并扫描以测定核酸样品与哪些探针杂交。基于这种测定,提供比较杂交和非杂交的模式获得序列信息。例如,序列信息可用于核酸测序、或诊断性筛选遗传疾病或是否存在特定病原体或病原体的菌株。
生物微阵列在扫描前往往用流体系统进行处理。例如,该流体系统包括能洗涤和对微阵列染色的流体站。随着微阵列设计的进步,经常需要改进流体系统以提高自动化并降低成本。

发明简述

“阵列”是可通过合成或生物合成方法制备而有意建立的分子库。阵列中的分子可相同或互不相同。阵列可采用各种形式,例如可溶性分子文库;连接于树脂珠的化合物文库;硅芯片或其它固体支持物。核酸文库或阵列是可通过合成或生物合成方法制备而有意建立的核酸库并可以各种形式(例如可溶性分子文库;连接于树脂珠的化合物文库;硅芯片或其它固体支持物)来筛选生物活性。此外,术语“阵列”可包括通过将基本上任何长度的核酸(例如,长1-约100个核苷酸单体)点在基材上制备的核酸文库。本文使用的术语“核酸”指任何长度的核糖核苷酸、脱氧核糖核苷酸或肽核酸(PNA)的聚合形式的核苷酸,所述核苷酸含有嘌呤和嘧啶碱基,或其它天然、化学或生物化学修饰的、非天然的或衍生的核苷酸碱基。多核苷酸骨架可含有糖和磷酸基团,或修饰或取代的糖或磷酸基团,一般可在RNA或DNA中发现。多核苷酸可含有修饰的核苷酸,例如甲基化核苷酸或核苷酸类似物。核苷酸序列可为非核苷酸组分打断。因此,术语核苷、核苷酸、脱氧核苷和脱氧核苷酸一般包括如本文所述的类似物。这些类似物是与天然存在的核苷或核苷酸具有某些相同结构特征的分子,以致当掺入核酸或寡核苷酸序列时,它们可在溶液中与天然存在的核酸序列杂交。这些类似物一般通过取代和/或修饰碱基、核糖或磷酸二酯部分而衍生自天然存在的核苷和核苷酸。可有意设计这些变化来稳定或动摇杂交形成或提高与所需的互补核酸序列杂交的特异性。
生物聚合物或生物学聚合物是指生物或化学部分的重复单元。代表性的生物聚合物包括(但不限于)核酸、寡核苷酸、氨基酸、蛋白质、肽、激素、寡糖、脂质、糖脂、脂多糖、磷脂、上述物质的合成类似物,包括(但不限于)反向核苷酸、肽核酸、Meta-DNA和上述物质的组合。“生物聚合物合成”包括合成生产生物聚合物(有机和无机的)。
“生物单体”与生物聚合物相关,该术语指生物聚合物的单一的单元或非生物聚合物一部分的单一的单元。因此,例如核苷酸是寡核苷酸生物聚合物内的生物单体,氨基酸是蛋白质或肽生物聚合物的生物单体;例如亲和素、生物素、抗体、抗体片段等也是生物单体。“起始生物单体”或“起始物生物单体”指经反应性亲核试剂与聚合物表面共价相连的第一生物单体,或是与连接于聚合物的接头或间隔臂相连的第一生物单体,所述接头或间隔臂经反应性亲核试剂连接于聚合物。
“互补”指核苷酸或核酸之间的杂交或碱基配对,例如双链DNA分子的两条链之间或寡核苷酸引物和待测序或扩增的单链核酸上的引物结合位点之间。互补核苷酸一般是A和T(或A和U)或C和G。当进行最优对比和比较的具有合适核苷酸插入物或缺失物的一条链的核苷酸与另一条链的核苷酸的至少约80%配对、通常至少约90%-95%,更优选约98-100%时,则两条单链RNA或DNA分子认为是互补的。此外,当RNA或DNA链在选择性杂交条件下与其补体杂交时则存在互补性。当在14-25个核苷酸的延伸段上具有至少约65%互补、优选至少约75%、更优选至少约90%时,一般会发生选择性杂交。参见,纳入本文作为参考的M.KanehsiaNucleic Acid Res.12:203(1984)。
组合合成策略:组合合成策略是通过依次加入可由反应物矩阵和切换矩阵代表的试剂来平行合成各种聚合物序列的有序策略,其产物是产物矩阵。反应物矩阵是待加入的构件的1列m行矩阵。切换矩阵是1和安排在列中的m之间所有或一子集的二元数字(优选有序的)。“二元策略”是至少两个连续步骤照射基材上感兴趣区域的一部分(优选一半)的策略。在二元合成策略中,可从形成的能从有序组的反应物中形成所有可能的化合物。在最优选的实施策略中,二元策略指也因式分解(factor)前述加入步骤的合成策略。例如,所述策略可是遮掩策略的切换矩阵将以前照射的区域一分为二,照射约以前照射区域的一半并保护剩下一半(也保护约以前保护区域的一半并照射约以前照射区域的一半)。应理解的是二元循环可间插以非二元循环,那么仅部分基材可经受二元策略。组合“遮掩”策略是使用光或其它空间选择性脱保护或激活剂从材料中除去保护基团以加入其它物质,例如氨基酸的合成方法。
有效量指足以诱导所需结果的量。
基因组是生物体的染色体中所有的遗传物质。来源于特定生物体的染色体中遗传物质的DNA是基因组DNA。
基因组文库是从代表生物体全部基因组的一套随机产生的重叠DNA片段制得的克隆库。杂交条件通常包括低于约1M的盐浓度,更常见是低于约500mM,优选低于约200mM。杂交温度可低至5℃,但通常高于22℃、更常见的高于30℃,优选高于37℃。较长的片段可能需要更高的杂交温度来进行特异性杂交。由于其它因素也会影响杂交的严谨性,所述因素包括互补链的碱基组成和长度,存在有机溶剂和碱基错配的程度,所以参数的组合比单独任何一个绝对测量值更重要。
杂交,例如等位基因特异的探针杂交一般在严谨条件下进行。例如,条件为盐浓度不高于1摩尔(M),温度至少25℃,如750mM NaCl、50mM磷酸钠、5mMEDTA、pH7.4(5×SSPE)和约25-30的温度。
杂交通常在严谨条件下进行,例如盐浓度不高于1摩尔(M)和温度至少25℃。例如5×SSPE(750mM NaCl、50mM磷酸钠、5mM EDTA、pH7.4)和25-30的温度是适合于等位基因特异的探针杂交的条件。就严谨条件而言,参见,例如全文纳入本文作为参考的Sambrook,Fritsche和Maniatis,《分子克隆实验室手册》(MolecularCloning A Laboratory Manual),第二版,冷泉港出版社(1989)。
术语“杂交”指两条单链多核苷酸非共价结合成稳定的双链多核苷酸;三链杂交在理论上也是可能的。得到的双链多核苷酸(通常)是“杂交体”。本文将形成稳定的杂交体的多核苷酸群的比例称为“杂交程度”。
杂交探针是能以碱基特异性方式结合核酸的互补链的寡核苷酸。这种探针包括如Nielsen等,Science 254,1497-1500(1991)所述的肽核酸和其它核酸类似物和核酸模拟物。
“特异性杂交”指当序列存在于复杂的混合物(例如,总细胞的)DNA或RNA中时,一种分子仅与特定的核苷酸序列或多条序列在严谨条件下结合、形成双螺旋或杂交。
分离的核酸是作为主要种类存在的本发明目标种类(即,在摩尔基础上比组合物中任何其它单个种类更丰富)。分离的核酸优选包含所有存在的大分子种类的至少约50、80或90%(在摩尔基础上)。目标种类最优选纯化至基本上均一性(常规方法不能在组合物中检测到污染的种类)。
配体:配体是为特定的受体所识别的分子。与受体结合或反应的物质称为“配体”,该术语仅根据其对应的受体才明确有意义。除了讨论中的物质能结合或与受体相互作用以外,术语“配体”不涉及任何特定的分子大小或其它结构或构象特征。配体也可用作与受体结合的天然配体或作为激动剂或拮抗剂的功能性类似物。本发明可研究的配体的例子包括(但不限于)细胞膜受体的激动剂和拮抗剂、毒素或毒液、病毒表位、激素(例如,阿片剂、类固醇等)、激素受体、肽、酶、酶底物、底物类似物、过渡态类似物、辅助因子、药物、蛋白质和抗体。连锁不平衡或等位基因结合指特定的等位基因或遗传标记择优与染色体位置附近的特定等位基因或遗传标记结合的频率高于意外期望的该种群中任何特定等位基因的频率。例如,如果基因座X具有相同频率发生的等位基因a和b,而连锁基因座Y具有相同频率发生的等位基因c和d,则期望ac组合发生的频率为0.25。如果ac更频繁发生,则等位基因a和c是连锁不平衡的。连锁不平衡可起因于等位基因的某些组合的天然选择或由于等位基因被引入种群太近以至于不能与连锁等位基因达到平衡。
混合种群或复杂种群:指任何含有需要和不需要核酸的样品。作为非限制性的例子,核酸的复杂种群可是总基因组DNA、总基因组RNA或它们的组合。此外,核酸的复杂种群中可富集给定种群但也包括其它不需要的种群。例如,核酸的复杂种群可是富集所需的信使RNA(mRNA)序列但仍含有一些不需要的核糖体RNA序列(rRNA)的样品。
单体:指可连接到一起以形成寡聚物或聚合物的分子集合的任何成员。以(多)肽合成为例,用于本发明的单体集合包括(但不限于),L-氨基酸、D-氨基酸或合成的氨基酸集合。本文使用的“单体”指用于寡聚物合成的基础集合的任何成员。例如,L-氨基酸二聚体形成了用于多肽合成的400“单体”的基础集合。不同的单体的基础集合可用在聚合物合成的连续步骤中。
术语“单体”也指化学亚单位,该化学亚单位可与不同的化学亚单位结合以形成大于每个亚单位的化合物。本文使用的“mRNA”或“mRNA转录物”包括(但不限于)前-mRNA转录物、转录物加工中间体、准备翻译的成熟mRNA、一个或多个基因的转录物,或来源于mRNA转录物的核酸。转录物加工包括剪接、编辑和降解。本文使用的“来源于mRNA转录物的核酸”指合成最终用作模板的mRNA转录物或其亚序列的核酸。因此,逆转录自mRNA的cDNA、转录自cDNA的RNA、扩增自cDNA的DNA、转录自扩增的DNA的RNA等均来源于mRNA转录物,检测这种衍生的产物是样品中原始转录物的存在和/或丰富程度的征兆。因此,mRNA衍生的样品包括(但不限于)一种或多种基因的mRNA转录物、逆转录自mRNA的cDNA、转录自cDNA的cRNA、扩增自基因的DNA、转录自扩增的DNA的RNA等。

本发明的核酸可含有嘧啶和嘌呤碱基的任何聚合物或寡聚物,分别优选胞嘧啶、胸腺嘧啶和尿嘧啶与腺嘌呤和鸟嘌呤。参见,Albert L.Lehninger,《生物化学原理》(Principles of Biochemistry),793-800页(Worth Pub,1982)。实际上,本发明考虑了任何脱氧核糖核苷酸、核糖核苷酸或肽核酸组分与它们的任何化学变体,例如这些碱基的甲基化、羟甲基化或糖基化形式等。这些聚合物或寡聚物在组合物中可是异质或均质的,并且可从天然来源分离或经人工或合成产生。此外,核酸可是DNA或RNA或它们的混合物,并且可以单链或双链形式永久或暂时存在,包括同源双链、异源双链和杂交体状态。
“寡核苷酸”或“多核苷酸”是长度范围在至少2个、优选至少8个、更优选至少20个核苷酸的核酸,或是特异性地与多核苷酸杂交的化合物。本发明的多核苷酸包括分离自天然来源、重组产生或人工合成的脱氧核糖核酸(DNA)或核糖核酸(RNA)的序列和它们的模拟物。本发明多核苷酸的其它例子可是肽核酸(PNA)。本发明也包括有非传统碱基配对的情况,例如在某些tRNA中已鉴定和假定存在于三螺旋中的Hoogsteen碱基配对。在本申请中“多核苷酸”和“寡聚核苷酸”可互换使用。
探针:探针是可由特定靶识别的表面固定的分子。以具有10、12和更多个碱基的探针的所有可能组合的阵列为例,参见美国专利号6,582,908。本发明可研究的探针的例子包括(但不限于)细胞膜受体的激动剂和拮抗剂、毒素和毒液、病毒表位、激素(例如,阿片样肽、类固醇等)、激素受体、肽、酶、酶的底物、辅因子、药物、凝集素、糖、寡核苷酸、核酸、寡糖、蛋白质和单克隆抗体。
引物是能在合适条件下用作模板定向DNA合成的起始点的单链寡核苷酸,所述条件例如缓冲液和温度、存在四种不同的核苷三磷酸和用于聚合的试剂,如DNA或RNA聚合酶或逆转录酶。在任何给定的情况中,引物的长度取决于,例如引物的用途,并且一般是15-30个核苷酸。短引物分子一般需要较冷的温度来与模板形成足够稳定的杂交复合物。引物无需反映模板的确切序列,但必须与这种模板具有足够的杂交互补性。引物位点是模板上与引物杂交的区域。引物对是包括与待扩增序列的5’端杂交的5’上游引物和与互补的待扩增序列的3’端的补体杂交的3’下游引物的一组引物。
多态性指在某个群体中发生两种或多种遗传确定的交替序列或等位基因。多态性标记或位点是趋异发生的基因座。优选的标记具有至少两个等位基因,其中每一个的发生频率大于选择的种群的1%,优选大于10%或20%。多态性可包括一个或多个碱基变化、插入、重复或缺失。多态性基因座可小至一个碱基对。多态性标记包括限制性片段长度多态性、可变数量的串联重复(VNTR)、超变区、小卫星(DNA)、二核苷酸重复、三核苷酸重复、四核苷酸重复、简单序列重复和插入元件,例如Alu。第一个鉴定的等位基因形式是任意设计为参考形式,另一个等位基因形式设计为交替或变体等位基因。选择的种群中发生频率最高的等位基因形式有时称为野生型形式。双倍体生物体的等位基因形式可是纯合或杂合的。双等位基因多态性具有两种形式。三等位基因多态性具有三种形式。多态性包括单核苷酸多态性(SNP)。
受体:对给定的配体具有亲和力的分子。受体可是天然存在或人造的分子。它们也可以其未改变的状态或与其它种类聚集的状态使用。受体可通过特定的结合物质直接或间接地、共价或非共价地与结合成员相连。本发明可用的受体的例子包括(但不限于)抗体、细胞膜受体、与特定的抗原决定簇(例如在病毒、细胞或其它物质上)具有反应性的单克隆抗体和抗血清、药物、多核苷酸、核酸、肽、辅因子、凝集素、糖、多糖、细胞、细胞膜和细胞器。在本领域受体有时指抗配体。本文使用的术语“受体”没有不同的含义。两个大分子通过分子识别形成复合物后,就形成了“配体受体对”。本发明研究的受体的其它例子包括(但不限于)全文纳入本文作为参考的美国专利号5,143,854所示的分子。
“固体支持物”、“支持物”和“基材”可互换使用并指具有刚性或半刚性表面或多个表面的材料或材料组。虽然在一些实施方案中,需要用,例如孔、突起的区域、针、蚀刻槽等用物理方法分开用于不同化合物的合成区域,但在许多实施方案中,固体支持物的至少一个表面基本上是平的。根据其它实施方案,固体支持物可采用珠子、树脂、凝胶、微球或其它几何构造的形式。示范性的基材可参见美国专利号5,744,305。
靶:对给定的探针具有亲和力的分子。靶可是天然存在或人造的分子。它们也可以其未改变的状态或与其它种类聚集的状态使用。靶可通过特定的结合物质直接或间接地、共价或非共价地与结合成员相连。本发明可用的靶的例子包括(但不限于)抗体、细胞膜受体、与特定的抗原决定簇(例如在病毒、细胞或其它物质上)具有反应性的单克隆抗体和抗血清、药物、多核苷酸、核酸、肽、辅因子、凝集素、糖、多糖、细胞、细胞膜和细胞器。在本领域靶有时指抗探针。本文使用的术语“靶”没有不同的含义。当两个大分子通过分子识别形成复合物后,就形成了“探针靶配对”。
III.特定的实施方案
图1-2显示了本发明的一个实施方案的用于处理生物传感器的简化系统。这些示意图仅是例子,而不应过分限制权利要求的范围。本领域的普通技术人员可知晓许多变化、替代和改进形式。系统100包括流体部件、支持部件与电子和机械部件。虽然上文描述了使用系统100的一组选择的部件,但可有许多替代、改进和变化形式。例如,一些部件可扩展和/或组合。其它部件可插入上述部件。取决于实施方案,部件的安排可用其它替代部件来互换。例如,使用系统100的至少一部分,如流体站。这些部件的其它细节见本说明书并且在下文将更详细。
流体部件包括至少一些容器。每一个这些容器含有一种或多种用于处理至少一个生物传感器的流体。例如,每个容器是孔。在另一实施例中,孔分成一个或多个板和/或一个或多个条带。如图1-2所示,这些孔分成至少一个或多个孔板112、114和116。在一个实施方案中,孔板112、114和116中的每一个含有96个孔。在另一个实施方案中,每个孔含有一种或多种流体。在还有一个实施方案中,同一板或不同板上的不同孔含有相同或不同的流体。还有另一个实施方案中,不同的孔具有相同或不同的深度。
根据本发明的一个实施方案,流体部件110也包含一个或多个支架板。例如,每个支架板包含多个以多行和多列安排的多个孔并能放置一个或多个传感器。在一个实施方案中,支架板用于在杂交过程之前或之后将一个或多个传感器输送至系统100。在另一个实施方案中,支架板用于将一个或多个传感器输送出系统100。例如,支架板是包括至少一个比色皿支架的扫描托盘。
支持部件包括至少一个面板140和抽屉组件122、124和128。例如,抽屉组件122、124和128中的每一个包含一块可在水平方向滑动的面板。在一个实施方案中,孔板112置于抽屉组件122的面板上,孔板114置于抽屉组件124的面板上。例如,孔板112直接或间接地固定在抽屉组件122的面板的预定位置上。在另一个实施方案中,孔板114的一个或多个孔与抽屉组件122的面板基本上垂直。如图1-2所示,根据本发明的另一个实施方案,支持部件也包括额外的抽屉组件。每一个额外的抽屉组件包括一块能在水平方向滑动的面板。在一个实施方案中,孔板或支架板置于面板上。例如,孔板或支架板直接或间接地固定于面板上的预定位置。在另一个实施方案中,孔板或支架板的一个或多个孔基本上与面板垂直。
此外,支持部件也包括热板组件126。在一个实施方案中,热板组件126置于抽屉组件125的面板上。例如,热板组件126直接或间接地固定于抽屉组件128的面板上的预定位置。
图3(A)和3(B)显示了本发明一个实施方案的用于处理生物传感器的系统100中简化的热板组件126。这些示意图仅是例子,而不应过分限制权利要求的范围。本领域的普通技术人员可知晓许多变化、替代和改进形式。热板组件126包括绝缘结构310、加热器312、温度计314(例如热电偶)和传导结构316。虽然上文描述了使用热板组件126的一组选择的部件,但可有许多替代、改进和变化形式。例如,一些部件可扩展和/或组合。其它部件可插入上述部件。取决于实施方案,部件的安排可用其它替代部件来互换。例如,图3(A)是简化的顶视图,图3(B)是简化的底视图。这些部件的其它细节见本说明书并且在下文将更详细。
如图1、2、3(A)和3(B)所示,根据本发明的一个实施方案,孔板116置于传导结构316上。例如,孔板116包括一个或多个孔,每个孔含有至少一种流体。该流体通过传导结构316加热。流体的温度通过热电偶314直接或间接地监测。响应于(温度),热电偶314向温度控制器发出信号。在一个实施方案中,温度控制器也是系统100的一个部件。温度控制器根据目标温度处理接收的信号并调节加热器312的功率来实现流体的目标温度。在一个实施方案中,使用者通过温度接口将目标温度提供给系统100。例如,温度接口是系统100的一个部件。在另一个实施方案中,通过孔板116本身将目标温度提供给系统100。例如,孔板116携带由系统100的条形码(barcode)读数仪读数的条形码。系统100可基于条形码确定目标温度。在另一个实施例中,条形码读数仪,例如一种检测器是系统100的一个部件。
再回到图1-2,电子和机械部件可将每一个所述的一个或多个传感器从一个位置移到另一个位置。在一个实施方案中,电子和机械部件用作输送部件。在另一个实施方案中,传感器的移动可在一、二或三维中进行。在还有另一个实施方案中,传感器可以任何速度移动。如图1-2所示,电子和机械部件将每一个所述的一个或多个传感器从流体部件的一个孔移至另一个孔。例如,每一个所述的一个或多个传感器从孔板112的一个孔移至孔板114的另一个孔。此外,电子和机械部件将每一个所述的一个或多个传感器在流体部件110的对应孔内移动,和/或将每一个所述的一个或多个传感器移入和/或移出流体部件110的对应孔。例如,对应的孔是孔板112或114的孔。
根据本发明的一个实施方案,电子和机械部件包括夹钳组件132和至少包括马达134和136。夹钳组件可直接或间接地夹住或松开一个或多个传感器。马达134和136的每一个可使一个或多个夹住的传感器在两个相对的一维方向中移动。例如,马达134可使一个或多个传感器沿着与抽屉组件122或124的面板垂直的Z轴移动。在另一个实施例中,马达136可使一个或多个夹住的传感器沿着与抽屉组件122或124的面板平行的X轴移动。此外,电子和机械部件包括第三马达。例如,第三马达可使一个或多个夹住的传感器在沿着如图1-2所示的Y轴的两个相对方向中移动。
图4(A)和4(B)显示了本发明一个实施方案的用于处理生物传感器的系统100中简化的夹钳组件132。这些示意图仅是例子,而不应过分限制权利要求的范围。本领域的普通技术人员可知晓许多变化、替代和改进形式。夹钳组件132包括传感器410、马达420、凸轮430、架子和小齿轮440、弹簧450、臂状体460和462、指状组件470和472。虽然上文描述了使用夹钳组件132的一组选择的部件,但可有许多替代、改进和变化形式。例如,一些部件可扩展和/或组合。其它部件可插入上述部件。取决于实施方案,部件的安排可用其它替代部件来互换。这些部件的其它细节见本说明书并且在下文将更详细。
指状组件470和472中的每一个包括一个或多个指状体,例如两个指状体。当指状体处于关闭位置时,可直接或间接地夹住所述的一个或多个传感器190。当指状体处于打开位置时,可直接或间接地松开所述的一个或多个传感器190。如图4(A)和4(B)所示,指状组件470和472分别连接于臂状体460和462。指状体470和472在关闭位置和打开位置之间的移动由马达420驱动。马达420可至少通过凸轮430与臂状体460和462向指状体施加力。在一个实施方案中,臂状体460延伸超过用于连接指状组件470的部分,臂状体462延伸超过用于连接指状组件472的部分。例如,臂状体460包括节段480,臂状体462包括节段482。在另一个实施例中,传感器410包括局部和极限(home and limit)传感器。

高严谨洗涤过程1020中,所述一个或多个传感器中的每个至少在一个孔中洗涤。在每个孔,流体处于升高的温度中,对应的传感器与流体混合一段时间。例如,升高的温度是48℃,时间是25分钟。在另一个实施例中,升高的温度范围是36-42℃,例如41℃,时间也是25分钟。
每个SAPE染色过程1030和1070中,所述一个或多个传感器中的每个于室温在至少一个孔中染色。就每个孔而言,对应的传感器与流体混合一段时间。例如,所述一段时间是10分钟。在AB染色过程1050中,所述一个或多个传感器中的每个于室温在至少一个孔中染色。就每个孔而言,对应的传感器与流体混合一段时间。例如,所述一段时间是10分钟。

根据本发明还有的实施方案,处理生物传感器的方法包括将第一传感器和第二传感器转移入处理生物传感器的系统。该系统包括至少一个传送部件、一个杂交部件和一个流体部件。传送部件包括一个夹钳,流体部件包括第一容器和第二容器。第一容器容纳第一体积的第一流体,第二容器容纳第二体积的第二流体。此外,该方法包括在转移第一传感器和第二传感器之后,通过杂交部件在至少第一传感器和第二传感器上进行杂交过程。另外,该方法包括在杂交过程之后,直接或间接地将第一传感器从杂交部件移入第一容器并与第一体积的第一流体接触。该方法也包括在杂交过程之后,直接或间接地将第二传感器从杂交部件移入第二容器并与第二体积的第二流体接触。移动第一传感器和移动第二传感器至少由夹钳进行,移动第一传感器和移动第二传感器基本上同时进行。例如,该系统可根据系统100实施。
本发明有许多优点。本发明的某些实施方案提供自动化的流体和杂交系统。例如,该系统包括杂交炉组件并提供在杂交炉组件和该系统的其它部件之间自动输送。在另一个实施方案中,该系统提供与扫描系统之间自动地来回传送。本发明的一些实施方案提供了能进行杂交、洗涤和染色的整合及自动处理的系统。此外,联合扫描系统后,该系统也能进行整合及自动的扫描处理。本发明的某些实施方案可提高杂交和流体系统的处理量。例如,可平行处理多个生物传感器(如微阵列)。本发明的一些实施方案可降低杂交和流体系统以及扫描系统的体积。本发明的某些实施方案可减少在一个或多个生物传感器上进行的不同处理之间的交叉污染。例如,在某个给定的处理中,不同的传感器在不同的孔中洗涤、染色和/或储存。在另一个例子中,用于不同的处理的某给定传感器分别在不同的孔中洗涤、染色和存放。在又一个例子中,每个孔最多用于一次处理的最多一个传感器,例如微阵列。
虽然已描述了本发明的某些实施方案,本领域的技术人员应该理解的是有与所述实施方案等效的其它实施方案。因此,应该理解的是本发明不受具体阐述的实施方案的限制,而仅受限于附加的权利要求的范围。
Claims (64)
1.一种处理生物传感器的系统,所述系统包括:
装配为支持流体部件的支持部件,所述流体部件至少包括第一容器和第二容器,该第一容器可容纳第一体积的第一流体,该第二容器可容纳第二体积的第二流体;
装配为在第一传感器和第二传感器上进行杂交过程的杂交部件;
传送部件,该传送部件被装配成:
直接或间接地将第一传感器从杂交部件移入第一容器并与第一体积的第一流体接触;直接或间接地将第二传感器从杂交部件移入第二容器并与第二体积的第二流体接触;
其中该传送部件还装配为基本上同时移动第一传感器和第二传感器。
2.如权利要求1所述的系统,其特征在于,所述传送部件还装配为自动地移动第一传感器和第二传感器。
3.如权利要求1所述的系统,其特征在于,所述杂交部件包括炉。
4.如权利要求1所述的系统,其特征在于,所述传送部件还装配为:
将第一传感器移入杂交部件,所述第一传感器与第一杂交孔相连;
将第二传感器移入杂交部件,所述第二传感器与第二杂交孔相连。
5.如权利要求1所述的系统,其特征在于,还包括卸卡部件。
6.如权利要求5所述的系统,其特征在于,所述卸卡部件包括松开夹钳站。
7.如权利要求5所述的系统,其特征在于,所述传送部件还装配为:
将第一传感器从杂交部件移入卸卡部件,所述第一传感器与第一杂交孔相连;
将第二传感器杂交部件移入卸卡部件,所述第二传感器与第二杂交孔相连。
8.如权利要求7所述的系统,其特征在于,所述卸卡部件装配为使第一传感器与第一杂交孔分离,使第二传感器与第二杂交孔分离。
9.如权利要求1所述的系统,其特征在于,所述传送部件还装配为:
将第一传感器移至用于扫描的第一位置;
将第二传感器移至用于扫描的第二位置。
10.如权利要求1所述的系统,其特征在于:
当所述第一传感器在第一容器内并与第一体积的第一流体接触时,第一体积的第一流体完全保持在第一容器内;
当所述第二传感器在第二容器内并与第二体积的第二流体接触时,第二体积的第二流体完全保持在第二容器内。
11.如权利要求1所述的系统,其特征在于,所述第一传感器与第一支持元件相连。
12.如权利要求11所述的系统,其特征在于,所述第一支持元件是栓钉。
13.如权利要求11所述的系统,其特征在于,所述第二传感器与第二支持元件相连。
14.如权利要求13所述的系统,其特征在于,所述每个第一支持元件和第二支持元件是一块板的一部分。
15.如权利要求1所述的系统,其特征在于:
第一传感器与传感器的长度、传感器的宽度和传感器的厚度相关;
传感器的长度等于或短于10mm;
传感器的宽度等于或狭于10mm;
传感器的厚度等于或薄于1000μm。
16.如权利要求1所述的系统,其特征在于,所述第一传感器是生物传感器。
17.如权利要求16所述的系统,其特征在于,所述生物传感器包括微阵列。
18.如权利要求1所述的系统,其特征在于,所述第一流体的第一体积等于或小于5ml。
19.如权利要求1所述的系统,其特征在于,所述第一流体和第二流体种类相同或不同。
20.如权利要求1所述的系统,其特征在于,所述第一体积和第二体积的大小相同或不同。
21.如权利要求1所述的系统,其特征在于,所述每个第一容器和第二容器均是孔。
22.如权利要求1所述的系统,其特征在于,所述第一容器和第二容器是多个容器中的两个,所述多个容器直接或间接连接于共同部件。
23.如权利要求22所述的系统,其特征在于,所述多个容器排列为多行和多列。
24.一种用于处理生物传感器的系统,所述系统包括:
装配为支持流体部件的支持部件,所述流体部件至少包括第一容器和第二容器,所述第一容器可容纳第一体积的第一流体,所述第二容器可容纳第二体积的第二流体;
装配为基于至少与预定温度相关的信息在第一传感器和第二传感器上进行杂交过程的杂交部件;
包括夹钳和至少一个马达的传送部件;
其中:
支持部件包括支持所述至少第一容器和第二容器的抽屉,所述第一容器和第二容器相对于抽屉基本上静止;
夹钳可基本上同时夹住所述第一传感器和第二传感器并基本上同时松开所述第一传感器和第二传感器;
所述至少一个马达装配为:
将夹住的第一传感器移入第一容器并与第一体积的第一流体接触;
将夹住的第二传感器移入第二容器并与第二体积的第二流体接触;
基本上同时移动第一传感器和第二传感器。
25.如权利要求24所述的系统,其特征在于,所述系统包括装配检测与所述第一容器和第二容器相连的指示器的检测器,所述指示器与预定的温度相关。
26.如权利要求25所述的系统,其特征在于,
所述指示器包括条形码;
所述检测器包括条形码读数器。
27.如权利要求24所述的系统,其特征在于,所述夹钳还能将所述第一容器和第二容器放于抽屉上和将所述第一容器和第二容器从抽屉上移开,
28.如权利要求24所述的系统,其特征在于,所述夹钳还能将所述抽屉从第一位置推至第二位置和将所述抽屉从第二位置拉回第一位置。
29.如权利要求24所述的系统,其特征在于:
所述至少一个马达能以至少6个方向移动夹住的第一传感器;
所述6个方向中的每一个均在所述6个方向中的另一个方向对面并垂直于所述6个方向中的4个方向;
所述6个方向中的4个方向不同于另一个方向。
30.如权利要求24所述的系统,其特征在于:
所述支持部件包括与第二温度相关的加热物;
所述加热物包括装配为加热所述第一体积的第一流体和所述第二体积的第二流体的加热器;
所述加热物还包括装配测量第一温度的温度计。
31.如权利要求30所述的系统,其特征在于,所述加热物是一块板。
32.如权利要求24所述的系统,其特征在于,所述系统还包括装配与加热器和温度计耦联并将第二温度升高至预定值的温度控制器。
33.如权利要求24所述的系统,其特征在于,所述传送部件与处理系统耦联,所述处理系统装配为向传送部件提供夹住、松开或移动所述第一传感器和第二传感器的指令。
34.如权利要求33所述的系统,其特征在于,所述处理系统包括计算机。
35.如权利要求24所述的系统,其特征在于,所述系统装配为处理所述第一传感器和第二传感器从而使所述处理过的第一传感器和第二传感器随时可用于扫描。
36.如权利要求24所述的系统,其特征在于:
当所述第一传感器在第一容器内并与第一体积的第一流体接触时,第一体积的第一流体完全保持在第一容器内;
当所述第二传感器在第二容器内并与第二体积的第二流体接触时,第二体积的第二流体完全保持在第二容器内。
37.如权利要求24所述的系统,其特征在于,所述第一传感器与第一支持元件相连。
38.如权利要求37所述的系统,其特征在于,所述第一支持元件是栓钉。
39.如权利要求37所述的系统,其特征在于,所述第二传感器与第二支持元件相连。
40.如权利要求38所述的系统,其特征在于,所述第一支持元件和第二支持元件各自是板的一部分。
41.如权利要求24所述的系统,其特征在于:
第一传感器与传感器的长度、传感器的宽度和传感器的厚度相关;
传感器的长度等于或短于10mm;
传感器的宽度等于或狭于10mm;
传感器的厚度等于或薄于1000μm。
42.如权利要求24所述的系统,其特征在于,所述第一传感器是生物传感器。
43.如权利要求42所述的系统,其特征在于,所述生物传感器包括微阵列。
44.如权利要求24所述的系统,其特征在于,所述第一流体的第一体积等于或小于5ml。
45.一种用于处理生物传感器的方法,所述方法包括:
将第一传感器和第二传感器转移入处理生物传感器的系统,所述系统至少包括传送部件、杂交部件和流体部件,所述传送部件包括夹钳,所述流体部件包括第一容器和第二容器,所述第一容器容纳第一体积的第一流体,所述第二容器容纳第二体积的第二流体;
在转移第一传感器和第二传感器之后,通过杂交部件在所述至少第一传感器和第二传感器上进行杂交过程;
杂交过程之后,直接或间接地将第一传感器从杂交部件移入第一容器并与第一体积的第一流体接触;
杂交过程之后,直接或间接地将第二传感器从杂交部件移入第二容器并与第二体积的第二流体接触;
其中:
至少由夹钳移动第一传感器和第二传感器;
基本上同时移动第一传感器和移动第二传感器。
46.如权利要求45所述的方法,其特征在于,所述第一传感器和第二传感器的移动自动进行。
47.如权利要求45所述的方法,其特征在于,所述方法还包括:
移动第一传感器和第二传感器之后,至少由夹钳将第一传感器移入杂交部件,所述第一传感器连接于第一杂交孔;
移动第一传感器和第二传感器之后,至少由夹钳将第二传感器移入杂交部件,所述第二传感器连接于第二杂交孔。
48.如权利要求47所述的方法,其特征在于,所述方法还包括:
杂交过程后,将所述第一传感器从杂交部件移至卸卡部件,所述第一传感器连接于第一杂交孔;
杂交过程后,将所述第二传感器从杂交部件移至卸卡部件,所述第二传感器连接于第二杂交孔;
其中所述卸卡部件是用于处理生物传感器的系统的一部分。
49.如权利要求48所述的方法,其特征在于,所述方法还包括通过卸卡部件将第一传感器和第一杂交孔分开与将第二传感器和第二杂交孔分开。
50.如权利要求45所述的方法,其特征在于,所述方法还包括将所述第一传感器从杂交部件移开后和将第二传感器从杂交部件移开后,处理所述第一传感器和所述第二传感器从而使处理过的第一传感器和处理过的第二传感器随时可用于扫描。
51.如权利要求50所述的方法,其特征在于,所述处理第一传感器和第二传感器包括进行至少一次低严谨洗涤、至少一次高严谨洗涤和对所述第一传感器和所述第二传感器的至少一次染色过程。
52.如权利要求45所述的方法,其特征在于,所述方法还包括至少由夹钳将所述第一传感器和所述第二传感器移至处理生物传感器的系统内的交接位置。
53.如权利要求52所述的方法,其特征在于,所述方法还包括:
将所述第一传感器和所述第二传感器从移交位置移出处理生物传感器的系统;
对处理过的第一传感器和处理过的第二传感器进行扫描过程。
54.如权利要求52所述的方法,其特征在于,在至少由夹钳将第一传感器和第二传感器移入处理生物传感器的系统和将第一传感器和第二传感器移至交接位置之间:
所述第一体积的第一流体不与除第一传感器以外的任何传感器接触;
所述第二体积的第二流体不与除第二传感器以外的任何传感器接触。
55.如权利要求52所述的方法,其特征在于,在至少由夹钳将第一传感器和第二传感器移入处理生物传感器的系统和将第一传感器和第二传感器移至交接位置之间:
所述第一容器不提供不同于第一体积的第一流体且不由第一传感器提供的任何体积的任何流体;
所述第二容器不提供不同于第二体积的第二流体且不由第二传感器提供的任何体积的任何流体。
56.如权利要求45所述的方法,其特征在于:
所述第一传感器从杂交部件的移动包括当所述第一传感器在所述第一容器内并与第一体积的第一流体接触时,所述第一体积的第一流体完全维持在第一容器中;
所述第二传感器从杂交部件的移动包括当所述第二传感器在所述第二容器内并与第二体积的第二流体接触时,所述第二体积的第二流体完全维持在第二容器中。
57.如权利要求45所述的方法,其特征在于,第一传感器与第一支持元件相连。
58.如权利要求57所述的方法,其特征在于,所述第一支持元件是栓钉。
59.如权利要求57所述的方法,其特征在于,所述第二传感器与第二支持元件相连。
60.如权利要求59所述的系统,其特征在于,所述每一个第一支持元件和第二支持元件是板的一部分。
61.如权利要求45所述的系统,其特征在于,所述第一传感器是生物传感器。
62.如权利要求61所述的系统,其特征在于,所述生物传感器包括微阵列。
63.如权利要求45所述的方法,其特征在于,所述第一流体和所述第二流体的种类相同或不同。
64.如权利要求45所述的方法,其特征在于,所述第一体积和所述第二体积的大小相同或不同。
