Abstract

The present invention relates to a cap which can form an essentially leak- proof seal with an open-ended vessel capable of receiving and holding fluid specimens or other materials for analysis. To minimize potentially contaminating contact between a fluid sample present in the vessel and humans or the environment, the present invention features a cap having a frangible seal which is penetrable by a plastic pipette tip or other fluid transfer device. The cap further includes filtering means for limiting dissemination of an aerosol or bubbles once the frangible seal has been pierced. The filtering means is positioned between the frangible seal and retaining means. The retaining means is positioned on the cap above the filtering means and may be used to contain the filtering means within the cap. The retaining means may be comprised of a material which is penetrable by a fluid transfer device.

Claims (28)

1. A cap comprising:
a core structure comprising a closed side wall adapted to grip an open-ended vessel and having an opening formed therein; the opening being sized to receive a fluid transfer device therethrough;
a first frangible seal affixed to the core structure and covering the opening so as to provide a fluid barrier; and a filter for impeding or preventing the passage of an aerosol or bubbles through the cap, the filter being positioned above the first frangible seal and substantially within the cap.
2. The cap of claim 1, wherein the core structure is a unitary plastic piece having a generally cylindrical shape.
3. The cap of claim 1 or 2, wherein the first frangible seal is affixed to a top or bottom surface of a ledge extending inwardly from the closed side wall.
4. The cap of claim 1 or 2, wherein the first frangible seal is affixed to a top surface of a ledge extending inwardly from the closed side wall or to a bottom surface of a skirt depending from the ledge.
5. The cap of any one of claims 1 to 4, wherein the filter is comprised of a resilient material.
6. The cap of claim 5, wherein the filter comprises a fibrous material.
7. The cap of claim 6, wherein the filter is a pile fabric.
8. The cap of any one of claims 1 to 7, wherein the filter is constructed and arranged to facilitate the venting of air through the cap.
9. The cap of any one of claims 1 to 8 further comprising a retainer for containing the filter within the cap.
10. The cap of claim 9, wherein the retainer is a foil ring affixed to the core structure above the filter, the foil ring defining an opening which is sized to receive a fluid transfer device therethrough.
11. The cap of claim 9, wherein the retainer is a disc affixed to or seated in a shelf of the core structure above the filter, the retainer having a hole formed therein which is sized to receive a fluid transfer device therethrough.
12. The cap of claim 11, wherein the hole is covered by a removable seal having a tab.
13. The cap of claim 11, wherein the hole is covered by a second frangible seal.
14. The cap of claim 9, wherein the retainer is a second frangible seal affixed to the core structure above the filter.
15. The cap of claim 14, wherein the first and second frangible seals are comprised of the same material.
16. The cap of claim 14 or 15, wherein each of the first and second frangible seals comprises a foil or plastic film.
17. The cap of claim 14, wherein each of the first and second frangible seals comprises a foil.
18. The cap of claim 17, wherein each of the first and second frangible seals further comprise a brittle layer.
19. The cap of claim 17, wherein each of the first and second frangible seals comprises a foil layer, a heat seal layer and an intervening brittle layer.
20. The cap of claim 17, wherein each of the first and second frangible seals comprises a foil layer and a combined brittle/heat seal layer.
21. The cap of claim 14, wherein each of the first and second frangible seals comprises a plastic film.
22. The cap of any one of claims 13 to 21, wherein at least one of the first and second frangible seals is modified to facilitate penetration by a fluid transfer device by reducing the tensile strength of the at least one of the first and second frangible seals.
23. The cap of claim 22, wherein the at least one of the first and second frangible seals is modified to include creases, perforations, score lines or other mechanical impressions for reducing its tensile strength.
24. The cap of any one of claims 9 to 23, wherein the retainer is affixed to or seated in a top surface of the closed side wall of the core structure.
25. A system comprising the cap of any one of claims 1 to 24 in sealing engagement with an open-ended vessel adapted to hold a fluid substance.
26. A method for removing a fluid substance from the system of claim 25, the method comprising the steps of:

(a) penetrating the cap with a fluid transfer device such that air passageways are formed between the fluid transfer device and the first frangible seal, thereby permitting air to be vented from the system;

(b) drawing at least a portion of the fluid substance into the fluid transfer device;
and (c) removing the fluid transfer device from the system.
27. The method of claim 26, wherein the fluid transfer device comprises a plastic pipette tip.
28. The method of claim 26 or 27 further comprising the step of exposing the fluid sample removed in step (c) to reagents and conditions sufficient to amplify a target nucleic acid sequence suspected of being contained in the fluid sample.
